핵산염기 유도체와 salicylate들간의 분자수준에서의 결합양식에 관한 연구를 분광학적 및 구조화학적 수단으로 시행하였다. 비극성용매중 극히 희박한 농도에서 IR, H, C-NMR을 측정하여 결합부위, 결합상수를 조사하였다. salicylic acid는 핵산염기 adenine 및 uracil과 1 : 1로 결합했다. 핵산염기 cytosine과 guanosine도 결합에 참여하였다. 핵산염기 guanosine의 경우 강한 자기회합이 일부 파괴되었다. salicylamide는 핵산염기유도체와 아주 약하게 회합했다. The interactions of salicylic acid(SA) and salicylamide(SM) with nucleic acid base derivatives such as 9-ethyl adenine(A), 1-cyclohexl uracil(U), 2',3'-benzylidine-5'-trityl-cytidine(C) and guanosine-2',3',5'-isobutyrate(G) have been spectroscopically investigated to determine the binding mechanism. NMR and IR spectra were measured in nonpolar solvents at very low concentration. The association constant K of the complex was calculated from the IR spectra. Compounds SA and A form a 1:1 cyclic hydrogen-bonded complex. Compounds SA and U form a 1:1 hydrogen-bonded complex. Compounds SA and C form hydrogen-bonded complex. Compound SA binds to compound G, but its binding does not completely break the self-association of compound G. Compound SM binds to compounds A, U, C and G very weakly.

The interaction of polyene antibiotics, amphotericin B and nystatin, with liposomes has been investigated. The activity of amphotericin B and nystatin as a function of cholesterol and ergosterol contents was studied. The polyene were added at approximate final molarities of 10^(-6)-10^(-4)M. The molar percentage of sterols was increased from 0 ㏖% to 50 ㏖%. At this concentration range, nystatin had very little effect and amphotericin B had a little stronger effect. At a higher concentration above 8×10^(-5)M, amphotericin B showed strong lytic activity on liposomes with or without sterols. These studies suggest that amphotericin B is more potent lytic agent than nystatin and that the affinites for sterols of two polyene antibiotics depend strongly on their concentrations and sterol contents.

The effects of water soluble derivatives of polyene antibiotics, amphotericin B methyl ester (AME) and nystatin methyl ester, on the osmotic behavior of sterol-containing liposomes were investigated. The activity of polyene methyl esters on different sterol-containing liposomes as a function of sterol contents was observed. Cholesterol, ergosterol and 7-dehydrocholesterol were incorporated to egg phosphatidylcholine/dicetyl phosphate liposomes, which showed a good osmotic stability. When polyene derivatives were added to liposomes containing ergosterol, the osmotic stability was disturbed at final concentration of 6.45×10^(-5)M. Cholesterol-containing liposomes were not affected by these antibiotic solutions of the same concentration. 7-Dehydrocholesterol-containing liposomes have shown the intermediate sensitivity to AME when compared with cholesterol- and ergosterol-containing liposomes. Selectiveactivity on ergosterol-containing liposomes was also observed for nystatin methyl ester at a higher concentration above 2.31×10^(-4)M. These results suggest that AME is more potent than nytatin methyl ester and that the selectivity of polyene antibiotics for fungi is due to a stronger interaction of polyenes with ergosterol than with cholesterol.

It was attempted to elucidate the selective associational properties of nucleic acid bases with peptides by use of spectroscopic tools. Monopeptide and dipeptide models interact with uracil through two hydrogen bond bridges. The addition of peptidyl group produces the binding with guanosine derivative, while monopeptide cannot interact. Adenine and uracil derivatives associate with glutamic acid much more strongly than any other amino acids. In the case of guanosine, it is conspicuous that glutamine can uniquely interact with it. These results might give the information on the interaction between peptide and nucleic acid.
