칡소와 비경흑색 한우의 Melanocortin Receptor 1(MC1R) 유전자형 분석

칡소와 비경흑색 한우의 MC1R 유전자의 유전자형를 조사하고 칡소의 모색 발현과 한우비경색과의 관계를 규명하기 위하여 MC1R 유전자의 PCR-RFLP 분석을 수행하였다. 소에서 나타나는 6가지 유전자형(E^D/E^D, E^D/E^+, E^D/e, E^+/E^+, E^+/e와 e/e) 중 칡소에서는 단지 2개의 유전자형 E^+/E^+, E^+/e 만이 출현하였고, e/e 유전자형을 가지는 개체는 전혀 출현하지 않아 제주재래흑우에서와 같이 흑색 호반모가 발현되기 위해서는 기본적으로 E^+ allele이 필요한 것으로 사료되었다. 비경흑색 한우와 비경황색 한우에서는 모두 E^+/e 혹은 e/e 유전자형이 출현하였고 E^+와 e allele의 빈도는 각각 0.37, 0.63과 0.11, 0.89였다. 비록 E^+ allele의 빈도가 비경흑색 한우에서 비경황색 한우에서 보다 높았지만, E^+ allele과 비경흑색과는 완전한 연관성은 없었다. 이러한 결과는 MCIR 유전자 분석이 칡소 뿐만 아니라 한우의 모색 고정에도 유용한 도구로 이용 가능함을 제시하고 있다. PCR-RFLP analysis was carried out to investigate the genotype of Melanocortin receptor 1 (MC1R) gene in Korean Brindle Cattle and Korean Cattle with dark muzzle, which are coat color and muzzle pigmentation variants of Korean Cattle, respectively. Allelic variants of MC1R in cattle were analyzed by digestion with BsrFⅠ, AciⅠ. Among six genotypes, E^D/E^D, E^D/E^+, E^D/e, E^+/E^+, E^+/e and e/e, detected in cattle, only two genotypes, E^+/E^+ and E^+/e, were observed in Korean Brindle Cattle, probably reflecting the necessary of E^+ allele for the expression of black brindle coat color. As in Korean Cattle with light muzzle, the E^+/e and e/e genotypes were detected in Korean Cattle with dark muzzle. The E^+ and e alleles frequencies in two populations of Korean Cattle with dark muzzle and with light muzzle were 0.37, 0.63 and 0.11, 0.89, respectively. Although the frequency of E^+ allele in Korean Cattle with dark muzzle was higher than in Korean Cattle with light muzzle, the E^+ allele was not completely associated with dark muzzle pigmentation. The results of this experiment indicate that the difference of MC1R genotype and frequency may be useful for fixation of coat color in Korean Cattle as well as Korean Brindle Cattle.

PCR-RFLP analysis of Melanocortin receptor 1 (MC1R) gene was carried out to investigate the genotype distribution in Korean-Native and Chinese-Native pig breeds(Neijiang, Putian, Wannanhua and Jinhua). Allelic variants of MC1R in pigs were analyzed by digestion of BspH I , AccII and Hha I . Ncijiang. Wannanhua and Jinhua had only MC1R*2 allele which is considered to be typical genotype for Meishan and Large Black. Among 20 Korean-Native Pigs and 5 Putian pigs, 10 and 2 heads had the MC1R*3 allele (*2/3 or 3/3), respectively, which was detected in European pig breeds such as Hampshire, Large White, and Pietrain. The remaining animals possessed the genotype MC1R*2/2, probably reflecting the genetic introgression of MC1R*3 allele into Korean-Native and Putian pig breeds by the crossbreeding for improvement. The alleles MC1R*l and MC1R*4, which are considered to be typical allele for European Wild Boar with wild-type coat color and Duroc with red coat color, respectively, were not detected in pig breeds used in this experiment. These results indicate that the analysis of genotype frequencies of MC1R gene may be a useful tool for the conservation of Korean-Native and Chinese-Native pig breeds.
한우 , 제주재래흑우 , 흑모화우와 갈모화우에서의 MSH Receptor(MC1R) 유전자의 유전자형 및 빈도 비교

PCR-RFLP analysis of MSH receptor (melanocyte-stimulating hormone receptor; MCIR) gene was carried out to confirm the genotypes and frequencies in Korean Cattle, Cheju Native Black Cattle, Japnaese Black, Japanese Brown, Holstein and Angus. Genetic relationship of these breeds and coat colors to alleles at the Extension locus, that encodes MCIR were also determined. Allele E^D E^+ and a were detected by digestion with BsrF I (or Msp I ) and Aci I . A Dominant allele E^D, which is considered to be typical allele for dominant black coat color in cattle breeds, such as Holstein and Angus, was not detected in Korean Cattle and Japnases Brown with yellowish-brown coat color. Among sixty-two Cheju Native Black Cattle and fifteen Japanese Black, only ten and eleven heads had the dominant allele E? respectively. The remaining animals possessed the genotype E^+/E^+ or E^+/e, which probably reflect alleles at the A-locus which are supposed to give rise to the recessive black color. Several animals of Cheju Native Black Cattle and Japanese Black possessed the genotype E^D/-, probably reflecting the genetic introgression of E^D allele into these breeds during the crossbreeding period for improving. These results show that the difference of MCIR genotype and frequency among these breeds may be useful for conservation of endangered Cheju native Black Cattle and also genetically discreminating between meat of Korean Cattle and that of Holstein
