In order to use steels for hydrogen applications, it is important to secure safety through the evaluation of the hydrogen embrittlement (HE) behaviors. Therefore, it is important to develop a simple test method to examine the HE behaviors under high-pressure hydrogen environment. From this reasons, it is necessary to understand the effect of the surface finish condition of specimens depending on fabrication processes on the HE behaviors. In this study, in addition to austenitic steels, carbon (API-X70 steel) and alloy steels (4Cr-Mo steel) are used for in-situ SP tests in high-pressure environments. The obtained load-displacement curves and absorbed SP energy until gas leakage were obtained. In addition, the relative reduction of thickness (RRT) was evaluated by measuring the thickness of the fractured part of the specimen, and the HE sensitivity depending on the surface roughness was evaluated quantitatively. As a result, it can be found that the effect of the surface roughness of specimens on the HE sensitivity was not clearly observed in both kinds of steels with austenitic and ferritic phases.

In this study, we investigated in vitro anti-obesity activity of Parthenocissus tricuspidata (Siebold & Zucc.) Planch stem (PTS) in mouse adipocytes 3T3-L1 cells. PTS inhibited lipid accumulation and reduced the expression level of the proteins such as C/EBPα, PPARγ and FABP4 associated with adipogenesis in 3T3-L1 cells. In addition, PTS increased lipolysis-related protein expression such as p-AMPK, ATGL and p-HSL, and reduced perilipin-1 in 3T3-L1 cells. Taken together, PTS may have anti-obesity activity through inhibiting adipogenesis and inducing lipid lipolysis in adipocytes. Based on these results, PTS is expected to be used as a potential functional agent for anti-obesity.

Bok choy is one of Brassica vegetables widely consumed worldwide. Brassica vegetables have been reported to exert various pharmacological activities such as antioxidant, anti-cancer and cardioprotective activity. However, studies on immunostimulatory activity of bok choy sprout have not been conducted properly. Thus, in this study, we investigated in vitro immunostimulatory activity of bok choy sprout extract (BCS) using mouse macrophage RAW264.7 cells. Our results showed that BCS increased the production of immunomodulators such as NO, iNOS, IL-1β, IL-6, IL-12, TNF-α and MCP-1, and phagocytic activity in RAW264.7 cells. BCS activated MAPK, NF-κB and PI3K/AKT signaling pathways. However, BCS-mediated production of immunomodulators was dependent on JNK, NF-κB and PI3K/AKT signaling pathways. the mRNA expression of TLR2 were significantly increased by BCS, TLR2 inhibition by anti-TLR2 dramatically suppressed the production of immunomodulators by BCS. In addition, TLR2 inhibition by anti-TLR2 significantly reduced BCS-mediated phosphorylation level of AKT, JNK and NF-κB. From these results, BCS may have immunostimulatory activity via TLR2-MAPK, NF-κB and PI3K/AKT signaling pathways. Therefore, BCS expected to be used as a potential immune-enhancing agent.

Background: The leaves of Staphylea bumalda (S. bumalda), a deciduous tree distributed in Korea, China, and Japan, are used to treat respiratory diseases or inflammation. However, the immune-enhancing activity of S. bumalda leaves has yet to be examined. Thus, we investigated the effect of water extracts of S. bumalda leaves (SBL) on macrophage activity using RAW264.7 mouse macrophage cells. Methods and Results: SBL increased the production of immunomodulators such as NO, iNOS, IL-1β, IL-6, TNF-α, and MCP-1 and activated the phagocytic activity of RAW264.7 cells. SBL-mediated production of immunomodulators in RAW264.7 cells was blocked by inhibition of TLR2 and TLR4 and was attenuated by JNK inhibition. Moreover, SBL increased JNK phosphorylation, while inhibition of TLR2 and TLR4 blocked SBL-mediated JNK phosphorylation. These results suggest that SBL activates JNK through stimulating TLR2 and TLR4 in macrophages to induce immunomodulator production. In lipopolysaccharide-stimulated RAW264.7 cells, SBL inhibited the overproduction of immunomodulators. Conclusions: Given these results, SBL showed immunostimulatory activity under normal conditions and immunosuppressive activity during LPS-induced excessive immune response.
Immunostimulatory Activity of Hibiscus manihot Flower in Mouse Macrophages, RAW264.7 Cells

In this study, we investigated whether Hibiscus manihot flowers (HMF) exhibits immunostimulatory activity in RAW264.7 cells. HMF increased the production of immunostimulatory factors such as NO, iNOS, IL-1β and TNF-α in RAW264.7 cells. TLR2 and TLR4 blocked HMF-mediated production of immunostimulatory factors in RAW264.7 cells. In addition, the inhibition of MAPK signaling pathway reduced HMF-mediated production of immunostimulatory factors. From these results, HMF is thought to promote the production of immunostimulatory factors through activating TLR2/4/MAPK signaling in macrophages. It is believed that HMF can be developed as an agent related to immune enhancement in the future.

In this study, we measured the inhibitory activity of Pinus densiflora leaf (PDL) against excessive lipid accumulation in mouse preadipocyte, 3T3-L1 cells to investigate whether PDL exerts anti-obesity activity. Lipid accumulation and the protein level were measured using Oil red O staining assay and Western blot analysis, respectively. We observed that PDL inhibited excessive lipid accumulation and decreased the expression of CEBPα, PPARγ and perilipin-1 related to lipid accumulation in 3T3-L1 cells. Therefore, considering these results, PDL can be used as a potential agent for anti-obesity.
