N/A CKD602, a camptothecin derivative, is a synthetic and water-soluble anticancer agent possessing of topoisomerase I inhibiting activity. DPPC and DSPE-PEG liposomal formulations entrapped with CKD602 were developed. DSPE-PEG liposome, or PEGylated liposome, encapsulating CKD602 composed of dipalmitoylphosphatidylcholine (DPPC), cholesterol and distearoyl-N-monoethoxy poly (ethyleneglycol) succinylphosphatidylethanolamine (DSPEPEG_2000) (22:11:2) was prepared by reverse-phase evaporation method. Formed liposomes were characterized in terms of the morphology, size and encapsulation efficiency. To elucidate the in vitro stability, PEGylated liposome was incubated in human plasma, and the adsorbed proteins onto the surface of liposomes were applied to the SDS-PAGE. In vitro cytotoxicity of CKD602 encapsulated in PEGylated liposome was studied in human cervical cancer cell line (HeLa). CKD602 in PEGylated liposome was found to be 40-fold more effective (IC_50 = 1 nM) than free CKD602 (IC_50 = 40 nM) in inhibiting the growth of HeLa cells in vitro.

Cordyceps is reputed for its broad biological activities and as a tonic for replenishing vital function in Chinese traditional medicines. As an attempt to obtain fundamental data for the development of a new type Cordyceps, the effects of the fruiting bodies of cultivated fungus of Paecilomyces japonica grown on silkworm larvae on hyperglycemia induced by streptozotocin(STZ) and by epinephrine in rats and in mice as well as on immunological functions in mice were investigated. The 70% methanol extract of the fungus, when administered orally at 100 and 300 mg/kg in STZ-induced hyperglycemic rats, caused a significant decrease in blood glucose level 3hr after sample treatments. The methanol extract, when administered p.o. at the same dose levels in epinephrine-induced hyperglycemic mice, also caused a significant decrease in serum glucose levels as well as a significant reversal of the liver glycogen contents suggesting its hypoglycemic activity might be due to glycogen breakdown in the liver. Treatment of normoglycaemic mice with the methanol extract of the fungus exhibited a significant glucose tolerance up to 3hr after oral glucose load(2.0 g/kg). The methanol extract also showed immuno-stimulating activity as measured by carbon clearance in mice and a significant antifatigue effect as measured by weight loaded forced swimming performance in mice.

Development of Liposomal Formulations of bcl-2 Antisense Oligodeoxynucleotide

Liposomal formulations of bcl-2 antisense oligodeoxynucleotide (ODN) were developed and tested for its apoptotic effect on SK-OV-3 cells in vitro. The SGTA119-2009(S), a synthetic conjugate of 20-mer bcl-2 oligodeoxy-nucleotide (2009) with a GeneChem's proprietary mitomycin derivative by covalent bond, was used throughout the experiment. Negatively charged SGTA119-2009(S) was incorporated by the REV method into various liposomal formulations; either neutral one using DPPC: Choi: l,2-dioleoyl-sn-glycero-3-phosphoethanolamine (DOPE) (6:3:1) or cationic one using stearylamine (SA): DPPC : Choi: DOPE (1:6:3:1). Formed liposomes were characterized in terms of morphology, size and encapsulation efficiency. Average size of liposomes is 400~500nm in diameter. Structure of liposomes observed by scanning electron microscope (SEM) was globular shape and had multi lamellars. SGTA119-2009(S) had 12% encapsulation efficiency on neutral liposome and 34% encapsulation efficiency on cationic liposome. IC_(50) of SGTA119-2009(S) in DPPC liposomes was about lOOOnM in SK-OV-3 cells, whereas free antisense ODN did not show cytotoxic effect at this range. Western blotting was also carried out to confirm the Bcl-2 protein downregulation, though it was not semi-quantitatively matched with the growth inhibition results. More vigorous studies are under going to evaluate this and other basic matters.
