Differential Potential of Stem Cells Following Their Origin

목적: 세가지 기원의 줄기 세포 분화능과 면역표현형을 평가하고자 하였다. 대상 및 방법: 견봉하 점액낭과 골수, 탯줄 혈액 세 개의 군에서 세포를 채취하였다. 견봉하 점액낭과 골수는 견관절 수술 환자군에게 임상적 동의 하에 수술중 채취하였다. 각각의 채취된 세포 및 탯줄 혈액에 대하여 계대 배양을 시행하여 신경 분화군, 지방 분화군, 골 분화군을 평가하였으며 세포 표면 항체를 밝히기 위해 유동세포분석법을 이용하였다. 결과: 견봉하 점액낭 유래 세포에서는 신경분화와 지방 분화는 8예 모두 (100%)에서, 골분화는 8례 중 5예 (62.5%)에서 성공할 수 있었으며 골수 유래 세포의 경우 신경 및 지방 분화 유도한 6례 및 5예 모두 (100%) 분화에 성공하였으나 골분화 유도는 5예 중 4예 (80%)에서 얻을 수 있었다. 반면 탯줄 유래 세포 분화 연구의 경우 신경 분화 유도 67례 중 65예 (97%)에서 지방 분화연구 54예 중 29예 (53.7%)에서 골 분화 연구 57예 중 39예 (68.4%)에서 성공할 수 있었다. 결론: 탯줄 유래 줄기세포의 분화능과 비교하였을 때 견봉하 점액낭 및 골수 유래 줄기세포의 분화능이 우수함을 알 수 있으며 이는 향후 세포 치료에 있어서 안정성 있는 치료 제공자가 될 수 있을 것으로 보이며 향후 생체 실험 연구의 참고 자료로서도 가치가 있을 것으로 보인다. Purpose: To evaluate the differentiation potential of stem cells and their immunophenotype from 3 different sources. Methods: Our study involved three stem cell sources-subacromial bursal tissue, bone marrow, and umbilical cord blood. We obtained the subacromial bursal tissue and bone marrow from the patients undergoing shoulder surgery. After collecting the sample, we applied specific induction media for neurogenic, adipogenic and osteogenic differentiation. Also, flow-cytometry analysis was done to reveal the cell surface antigens. Results: We obtained 100% (8 cases) neural and adipogenic differentiation, but 62.5% (5 of 8 cases) osseous differentiation among the subacromial bursal tissue group. Bone marrow derived cells showed 100% neural (6 cases) and adipogenic (5 cases) differentiation, but 80% (4 of 5 cases) osseous differentiation. Umbilical cord blood derived cells revealed 97% (65 of 67 cases) neural, 53.7% (29 of 54 cases) adipogenic and 68.4% (39 of 57 cases) osseous differentiation. Immunophenotype analysis revealed that surface markers of bone marrow, subacromial bursal cell and umbilical cord blood derived mesenchymal stem cells are different from each other. Conclusions: Mesenchymal stem cells are potential agents in regenerative medicine and are characterized by expression of surface markers and by their differentiation potential. Our study with stem cells from subacromial bursal tissue, bone marrow and umbilical cord discovered that each stem cell has unique differentiation potential and function based on its origin. Various stem cells show multi-lineage differentiations in vitro which can be correlated to in vivo conditions.

Despite of numerous research on motivation in the field of tourism, little research has been conducted for identifying motivations of the Korean DMZ visitors and then segmenting them with distinct subgroups for practical marketing strategies. In this respect, this study aims to identify the underlying dimensions of motivation for the DMZ visitors, using a factor analysis; to segment the DMZ market, using a two step cluster analysis based on derived motivation factors; to investigate any difference between two segments with respect to value; and to examine any differences between market segments with respect to socio-demographic and behavioral variables. The results of this study show that five underlying motivation dimensions were derived: novelty, adventure, information, peace, and nostalgia. The results of two step cluster analysis indicate that two distinctive segments were identified: passive and active motivation visitors. These two segments were significantly different with respect to purpose of visit, type of partner, information sources, income, age, marital status, and occupation. Additional factor analysis for the value of visiting DMZ was performed. In the process of factor analysis, three factors of value(overall value, emotional value, and monetary value) were derived. The results of t-test indicate that statistically significant difference existed between two segments with respect to overall value.

The purpose of this study is to examine how mentoring functions affect organizational commitment and the probable existence of moderating effect by emotional intelligence. The functions of mentoring are classified into thre categories; career development, psychosocial, role modeling. Frequency analysis, reliability, factor analysis and regression analysis used to conduct the data analysis on 303 out of 324 questionnaires. Major suggestions and significances of this study can be outlined as follows; Firstly, the functions of mentoring like career development, psychosocial, and role modeling function had a positive effect on organizational commitment. Second, We found that there was significant difference about model fit by emotional intelligence. but effectiveness of organizational commitment by interaction effect was a little insignificant. As a result, based on these findings, We recognize that mentoring is useful tool in organizational commitment and emotional intelligence of employees need to be developed by hotel organization though result was insignificant.
내재적 학습동기의 선행요인으로 긍정심리자본에 대한 연구 - 관광전공 2년제 대학생을 중심으로 -

The purpose of this study is to identify positive psychological capital as an antecedent of intrinsic learning motivation. Data were collected from college students majoring in tourism in Seoul. Positive psychological capital was measured by four factors; self-efficacy, optimism, hope, resilience. Intrinsic learning motivation was measured by three factors: challenging assignment, curiosity, independent achievement. Structural equation model analysis was conducted to examine hypotheses. The results are as follows. First, two factors of positive psychological capital, self-efficacy and hope, had a significantly positive influence on intrinsic learning motivation. On the other hand, optimism of positive psychological capital had a significantly negative influence on intrinsic learning motivation, and resilience of positive psychological capital had no impact on it. The results can suggest practical implications to improve intrinsic learning motivation by creating ways to enhance self-efficacy and hope of positive psychological capital.
