        심장근육모세포주에서 Adriamycin으로 유발된 세포 사멸에 대한 Cardiotrophin-1의 효과

        Background and Objectives: Adriamycin (doxorubicin, ADR) is a highly effective anti-neoplastic drug, but its clinical use is limited by its adverse side effects on the heart. Cardiotrophin (CT-1), a potent cardiac survival factor, is capable of inhibiting apoptosis in cardiac myocytes. The aim of this study was to investigate the cytoprotective effects of CT-1 against ADR-induced apoptosis in vitro. Materials and Methods: We determined a reasonable ADR concentration for inducing cell death by utilizing a cell survival test performed in a dosedependent manner. To determine the requirements for apoptosis in ADR-treated cardiac myocytes (H9c2 cells), we examined the effect of CT-1 on survival and apoptotic changes using a cell counting kit (CCK), RT-PCR, and Western blotting. Results: In analyzing cell survival as determined by CCK, ADR-induced cell death was found to occur in a dose-dependent manner (50% death at 24 hours after 2 μM of ADR), and ADR was shown to decrease procaspase-3. On RT-PCR, expression of Bax-α mRNA increased and Bcl-2 decreased during the 24 hours after ADR treatment. Consequently, the ratio of Bax-α/Bcl-2 mRNA peaked at 24 hours after ADR treatment. In contrast, CT-1 effectively attenuated the ADR-induced cell death in a dose-dependent manner. The changes in Bax-α and Bcl-2 mRNA expression after ADR treatment were reversed by CT-1 (1 ng/mL) treatment. The protein levels of procaspase-3 decreased after ADR treatment, an effect which was reversed by CT-1 treatment. Akt phosphorylation was also increased by CT-1, demonstrating that CT-1 inhibited apoptosis induced by ADR. Conclusion: These data demonstrated that ADR-induced apoptosis of cardiomyocytes can be prevented by CT-1; therefore, it may be possible to use CT-1 as a cardioprotective agent during ADR chemotherapy in patients with cancer.       • 正常마우스 卵巢組織 生後發育에 關한 自記放射法的 硏究

        Postaatal growth and aging effect on the cell renewal system of ovary were observed by means of microradioautography. The 39 female mice were divided into 3 age groups, namely suckling young, young adult and old adult. The age ranges from 4-day to more than 50-week. The mice were treated with a single dose of H³-thymidine, 0.8 microcurie per gram ef body weight, intraperitoneally. The tissues were removed 2-hour after the treatment, and were examined histologically and labeling index were calculated after the necessary procedures. 1. Cell renewal of the ovary showed a generally high rate throughout the various postnatal ages. The highest labeling was observed in 4-day old group and thereafter gradual downcast was seen during suckling stage. At the young adult, the radioactive indices reached nearly maximal value when it was estrus. On the contrary, old adult showed lowest cell renewing during diestrus. 2. The ovarian cycle has much to do with the renewal patterns of all the cellular elements and quite an active patters was demonstrated during estrus, except in connective tissue. 3. All the connective tissue element appeared scantily in the younger age and gradually increased their amount with age proceed. At the old adult, many of the structures were replaced by abundant blood vessels and connective tissue. 4. The primordial follicle and the primary follicle should be considered separately as a different unit under the histological base. 5. None of the labeling on the oocytes was noted throughout this observation.

      • 실리콘 산화공정에 대한 실험적 연구

        Dry Oxidation and Wet Oxidation process of (100) P-type silicon have been examined experimentally. The oxidation temperature was 1,100℃ and Oxygen flow rate was changed from 200 cc/min to 1000 cc/min. From the experimental measurements, moxid₄atmon temperature, time and Oxygen flow rate have been graphed for oxide layer 0.24 μ∼0.9 μ in thickness. The optimum value of O₂ flow rate has been found to be 1.4 cm/sec. The quality of the grown oxide layer has been investigated in terms of the dielectric constant, breakdown voltage, fixed surface charge density(Ωf), and mobile ion charge density(Ωm/g). From these measurements, it is concluded that the quality of the oxide layer is sufficient to expect the normal operation of MOS transistors.

        There cases of traumatic intrauterine adhesions (Asherman`s syndrome) which developed at an interval of 10 months to 6 years after curettage were studied. Amenorrhea and sterility occurred in all the cases. And its definition, incidence, etiology, pathology, pathologic anatomy, symptoms, diagnosis, treatment, prognosis, and prevention are reviewed briefly.

        This study is determined a optimum conditions of apparatus operations and estimated a conversion factor of depth-averaged velocity (DAVCF) in puting a DAVCF to practical use through the field tests of a electromagnetic wave surface velocimeter (EWSVM). As a result the efficiency of a EWSVM show excellent in comparison with water-submerged current meters for a peculiarity of mechanical and a capacity at field tests. It show in case of a facile operations of vertical angle and declination as well as a convenience of measured method at performance tests. Even though field test are three stations of restricted conditions, the DAVCF values showed 0.472∼0.663, 0.732∼0.869, and 0.736∼1.047 for variations of vertical angle when velocity and bed material conditions are lower of gravel, medium of clay, and higher of gravel, respectively. Those ranges of flow velocity conditions are 0.3∼0.5㎧, 0.5∼1.3㎧ and 1.3∼1.5㎧ for lower, medium and higher in river channel, respectively. And the mean value of a DAVCF showed 0.757 for three conditions, but excluding condition of lower velocity, it showed 0.833, and only it showed 0.875 for the higher velocity condition.
