        동해-삼척지역 석회석의 물리화학적 특성이 탈황성능에 미치는 영향

        동해-삼척지역 석회석을 활용하여 습식배연탈황공정기술에 대한 적용성을 검토하기 위해 석회석의 물리화학적 특성이 탈황성능에 미치는 영향을 연구하였다. 실험은 ASTM 표준 시험방법인 염산적정법을 적용한 실험실 규모의 실험 장치를 이용하여 총중화능력을 측정하였다. 실험결과 총중화능력은 CaO 함량 등의 화학성분 보다 입자크기의 물리적 특성에 더 큰 영향이 있음을 알 수 있었으며, 입자크기가 미립화 될수록 총중화능력의 값은 높아지는 것으로 확인되었다. It studies that effect of limestone of physicochemical characteristic on the performance of flue gas desulfurization (FGD) and application examination for technology of wet type FGD process and to utilize the limestone in Donghae-Samcheok. The experiment method was measured total neutralizing capability (TNC) using the lab scale experimental apparatus based on the HCl titration test. The results of TNC of limestone samples were more dependent on the physical characteristics including particle size rather than chemical compositions such as CaO content and particle size of limestone get smaller, TNC is increased.

        A new approach of successful denaturation of human sperm chromatin without undergoing decondensation treatment

        Due to the highly folded chromatin in human sperm, a proper nuclear swelling was highly required to localize certain DNA inside the sperm nuclei. Therefore,previous method for denaturation of sperm chromatin had to adopt chemical agents of decondensation treatment using Heparin/DTT or LIS, directly applied into the sperm cell before further examinations by FISH. Nevertheless,authors still had questions arising on the efficiency of decondensation process which is directly related to the quality of fluorescence signals, which, in turn, underlies the reliability of the results in frequencies and compositions as that still not a proper solution to overcome the major limitation in sperm studies. In this study, we approached a newly improved denaturation process of sperm chromatin without undergoing decondensation treatments that intact human sperms were used as the first time to localize examined DNA, and also two rounds of sequential FISH was carried out in the same sperm cell for the first time to investigate an idea of nullisomy of given chromosomes. From the results, all the variable centromeric compositions of sperm chromosomes 7, 8, and sex chromosomes revealed with significantly given frequencies of monosomy,disomy and nullisomy. Moreover, nullisomy was identified as a true absence of given chromosome rather than technical error of hybridization failure under decondensation. From the findings by our modified denaturation of human sperm chromatin without undergoing decondensation treatment, we strongly believe that more advanced and deep studies in human sperm of nuclear architecture and frequencies can be progressed with significantly reliable results.

        국내 고품위 석회석의 로내탈황 반응특성 연구

        국내 발전소 등에서는 배출되는 가스 내 황산화물을 제거하기 위한 탈황공정을 운영하고 있으며 주로 고품위 석회석을 탈황용 흡수제로 활용하고 있다. 이에 본 연구는 국내 고품위 석회석을 대상으로 편광현미경을 통한 광물학적 결정구조 특성, 열량측정 장치를 활용한 수화활성도, 기공률 측정기를 활용한 기공크기 특성을 평가하 였으며, 이러한 결과를 토대로 로내 탈황 반응장치를 활용한 고품위 석회석의 탈황 특성을 평가하여 활용 방안 을 확보하고자 하였다. 실험결과 광물학적 특성 및 소성 후 형성되는 석회석 입자의 기공크기 및 기공 면적은 수화반응 및 탈황반응의 반응속도와 전환율에 중요한 인자로 작용하는 것을 확인하였다.

        Sequence Variation among Tandem Repeat Units of 5S rRNA Gene and Phylogenetic Relationship in Four Taxa of Dendranthema

        5S rRNA genes are valuable in genetic and phylogenetic studies, and also in physical mapping. Amplification by polymerase chain reaction of the 5S rRNA gene repeat units from 4 diploid taxa of Dendranthema examined in this study, has resulted integral multiples from the primary band of approximately 0.35 kb product, which is corresponded to the full-length of 5S rRNA gene unit. In sequence analysis, the repeat units of 5S rRNA genes in each species showed considerable nucleotide polymorphisms as autapomorphies or synapomorphies in several positions among clones within species. Even though the numbers of polymorphic sites revealed, the reasonable consensus seguence of each species were constructed by alignment of clones. In result of alignment, all 4 taxa in this study showed completely conserved coding region of 120 bp, but two different distinctive sizes revealed as 243 bp and 245 bp, respectively in NTS regions. In comparison of two distinctive NTS regions of the consensus sequences, phylogenetic factors of nucleotides according to the presence or absence of GACG (320 bp - 323 bp) and GA (332 bp - 333bp) revealed to indicate a specific molecular phylogenetic relationships which lead a cladogram of two distinctive groups by 5S rRNA gene unit. By constructing the parsimonious tree with all the clones examined within 4 taxa of Dendranthema, the molecular phylogenetic factors were accepted by indicating two specific major groups independently which are one with D. boreale, D. indicum and D. indicum for. albescens, and another with D. zawadskii var. latilobum.

        볼밀을 활용한 국내 탈황용 석회석의 분쇄일지수 측정

        In this study, It is secured calibration data for the Bond’s work index of Korean desulfurization limestones according to the mining location with the standard-scale ball mill, work index were measured using JIS M 4002 and KS E 3600. The results, Average work index of limestone are 15.05 kWh/ton in Donghae-Samcheok and 12.44 kWh/ton in Jecheon-Danyang. Jecheon-Danyang limestones were about 20% lower than Donghae- Samcheok limestones. Therefore, besides chemical composition, mining location of limestones has been considered to influence on the grindability of limestones. 본 연구는 표준규격에 준하는 볼밀을 활용하여 채광 위치에 따른 국내 탈황용 석회석의 분쇄일지수데이터를 확보하고자 하였으며, 분쇄일지수는 JIS M 4002 및 KS E 3600에 준하여 측정하였다. 석회석의 분쇄일지수 측정결과, 동해-삼척지역은 평균 15.05 kWh/ton, 제천-단양지역은 평균 12.44 kWh/ton으로 제천-단양지역석회석이 약 20% 정도 낮은 결과를 나타냄을 확인하였다. 따라서, 화학조성 외에 석회석의 채광위치에도 석회석의 분쇄성은 영향을 미치는 것으로 사료된다.

        H<sub>2</sub>S 제거를 위한 고온건식 탈황제의 연구 및 기술개발동향

        청정석탄기술 중 하나인 석탄가스화 복합발전기술의 이론적 고찰 및 주요단위공정에 사용되는 고온건식 탈황제의 연구 및 기술개발 동향을 조사하였다. 고온건식 탈황제 개발의 중요한 요소는 반응성, 내구성 및 내마모성에 있으며, 탈황제 종류는 칼슘계, 아연계, 망간계, 철계, 구리계 등이 있다. 현재 고온건식 탈황제의 경우 선진국을 중심으로 아연계 탈황제 제조기술이 가장 상용화 단계에 있으며, 국내에서도 다양한 지지체를 사용하여 가격이 저렴하고 성능이 우수한 아연계 및 비아연계 고온건식 탈황제의 상용화를 위한 연구개발이 진행 중에 있다. Theoretical reviews of integrated gasification combined cycle one of the clean coal technologies and trends in the study and technology development for high temperature desulfurization sorbents were investigated. Reactivity, durability and abrasion resistance is an important key for development of high temperature desulfurization sorbents, the kind of things include calcium, zinc, manganese, iron and copper-based sorbents. Current status of high temperature desulfurization sorbents, manufacturing techniques of zinc-based sorbent in advanced countries has commercialized. In case of Korea, various research studies are underway to commercialize the Zn and non Zn-based high temperature desulfurization sorbents to cheaper and superior capability using various supports.

        Sequence polymorphisms in ribosomal RNA genes and variations in chromosomal loci of Oenothera odorata and O. laciniata

        Numerous studies on Oenothera species have been investigated for the physiological and ecological characteristics. However, no such an information based on molecular cytogenetic has yet been introduced, in turn, is very essential for identifying sequence polymorphisms of rRNA genes with their loci on mitotic phases for further biological researches. In this study, sequence variations of rRNA genes in Oenothera odorata and O. laciniata were examined to identify informative factors as unique or repeat sequences in intra- and inter-specific variations. Intra-specific variation revealed that the sequences of the rRNA genes including the spacer regions were highly conserved revealing only a few variations. From the interspecific variation, spacer regions of species were completely different as (1) non-homologous sequences in NTS and (2) different type repeat sequences in ITS 1, 2 and 5.8S rRNA, whereas the remaining coding regions were highly conserved. FISH was carried out on mitotic phases using the 5S rDNA of the analyzed sequences. From the interphase and metaphase chromosomes of the examined species,two loci of 5S rDNA in O. odorata and four loci in O. laciniata were confirmed on the telomeric region of the short arm. Due to the small size and unclear centromere of the chromosomes, karyotype could not be completed. However, we confirmed that the chromosomes are organized by meta- and acrocentric chromosomes and the chromosomes with identified loci were assumed to be paired by the location of loci at the telomeric region on the short arm with relative lengths.

        스프레이 모사 흡수탑에서 국내 고품위 석회석의 결정구조가 분쇄 및 용해도에 미치는 영향 연구

        국내 석탄화력발전소의 습식배연탈황공정에서 탈황용 흡수제로 사용하는 국내산 고품위석회석의 결정구조가 분쇄특성 및 용해특성에 미치는 영향을 연구하였다. 광물학적 결정구조가 다른 제천-단양 지역 석회석 4종을 대상으로 결정구조, 분쇄일지수 및 용해도를 비교한 결과 석회석의 결정크기와 구조에 따라 분쇄일지수 차이가 발생하였으며 분쇄일지수가 상대적으로 낮은 석회석의 용해도가 높은 것으로 나타났다. 이에 따라 석회석의 CaO 함량과 함께 결정구조에 따른 분쇄특성도 중요한 반응인자로 확인되었다. It is studied the effect of crystal structure of domestic High-Ca limestones using absorbent for desulfurization on the grinding characteristics and dissolution rates in wet flue gas desulfurization process of domestic coal fired power plant. It compared the crystal structure, grinding work index and dissolution rates with 4 Limestones from Jecheon-Danyang with different crystal structure, and we found that grinding work index differ in crystal size and crystal structure of limestones : The lower the value of the grinding work index is, the higher the dissolution rates of limestones. Confirmed that we have important indicators of grinding characteristics for crystal structure with CaO content of limestones.

        Rapid and Accurate Method for Determining Th-234 in Seawater: Fe Co-precipitation, UTEVA Extraction, and Micro-precipitation

        Thorium-234 (Th-234; t(1/2) = 24.1 days) has been widely used as a tracer of particle settling and organic carbon export in the ocean. However, the use of Th-234 in the ocean has been hampered by labor-intensive procedures, low throughputs, and large uncertainties related to source impurities. Here, we demonstrate a more efficient technique developed to analyze Th-234 in seawater by modifying the traditional Fe(OH)(3) co-precipitation method. The advantages and shortcomings of this method were compared with the Th-234 analytical method using MnO2 precipitation. In the new method, following the equilibration of the Th-230 spike with Th-234, Th was co-precipitated with Fe. Although the precipitates include a fraction of U, Fe precipitation was fast, and the supernatant can be easily siphoned off, allowing simple handling of several samples simultaneously. The Th adsorbed onto particles, collected by Fe precipitation, was desorbed in similar to 4 M HNO3 solution by heating at 230 degrees C for 30 min in a sealed Teflon bottle. Then, Th was separated from U using UTEVA resin. The counting source of Th was prepared by micro-precipitation of Ce. We were able to process similar to 60 samples for 5 days onboard, and the first counting was completed within a week upon returning to the laboratory. The main advantages of this method include (1) easy increase in the sample volume, (2) rapid Fe precipitation, (3) high-purity Th sources for beta and alpha counting, (4) no need for mass spectrometry, and (5) the high throughput.
