3 investigate the different mechanism between ATP and compound 48/80 (C48/80)-induced histamine release, we observed effects of calcium antagonists in histamine release of rat peritoneal mast cells.Verapamil and diltiazgem (voltage-dependent calcium channel blocker) and TMB-8 (a blocker of intracellular calcium release) significantly inhibited ATP-induced histamine release, but did not inhibit C48/80 induced histamine release. Econazole (a blocker of receptor-operated calcium channel) dose-dependently inhibited both and C48/80-inducted histamine release,but inhibitory effect of econarole in ATP-induced histamine release was more potent than that in C48/80-induced histamine. EGTA dote-dependently inhibited ATP and C48/80-induced histamine release, and C48/80-induced histamine release was slightly inhibited by high concentrations (>2 mM) of EGTA. These results suggest that ATP·induced histamine release is related to both intraoellular calcium release and extracellular calcium influx via voltage-dependent calcium channel and receptor-operated calcium channel. C48/80-induced histamine release is related to extracellular calcium influx, specially recetor-operated calcium channel rather tllan voltage-dependent calcium channel.

To investigate action of ethanol on histamine release from rat peritoneal mast cells, we compared the inhibitory effect of ethanol with those of calcium antagonists in mechanism of between ATP and compound 48/80-induced histamine release. Ethanol dose- dependently inhibited 1007M ATP-induced histamine release, whereas did not inhibit 1 mcg/ml compound 4a/80-induced histamine release. Verapamil,TMB-8 and EGTA dose- dependently inhibited ATP-induced histamine release, but did not inhibit compound 48/80-induced histamine release. Such an inhibitory effect of calcium antagonist was similar to that of ethanol. These results suggest that the inhibitory effect of ethanol on histamine release from rat peritoneal mast cells is mediated via disturbance of calcium mobilization.
RAW 264.7 세포에서 silica에 의한 염증 반응에 미치는 calcium chelator의 영향

To investigate the action of calcium in silica-induced inflammatory response, we observed examined effect of calcium chelator on arachidonic acid (AA) release, reactive oxygen species (ROS) and peroxynitrite (PON) in RAW 264.7 cells stimulated by silica dose-dependently increased [^3H]AA release, ROS and PON generation. Extracellular Ca^2+ chelator, EGTA, dose dependently inhibited silica-induced AA release and PON generation, but did not affect ROS generation. Intracellular Ca^2+ chelator, BAPTA, significantly inhibited ROS and PON generation but did not have any influence on AA release. These results suggest that PON generation is associated with both extracellular Ca^2+ influx and intracellular Ca^2+ release, whereas AA release and ROS generation is related to extracellular CA^2+ influx and intracellular Ca^2+ release in RAW 264.7 cells, respectively.
