        Identification of CM1 as a Pathogenic Factor in Inflammatory Diseases and Cancer

        Background: CM1 (centrocyte/-blast marker 1) was defined by a mAb against concavabalin-A (ConA) activated PBMC. It is expressed in germinal center of human tonsil and on the surface of activated PBMC as well as cancer cells. Recently,increased productions of pro-inflammatory mediators were detected from activated PBMC by CM1 ligation. Methods:However, there is a limitation to explain the exact role of CM1 on inflammation and its related mechanisms, since the identity of CM1 is still not clarified. In our previous study, we have already confirmed that soluble form of CM1 was produced by Raji. Therefore, we performed Q-TOF analysis after immunoprecipitation of concentrated Raji culture supernatant using anti-CM1 mAbs. Results: As a result, we found that CM1 is identical to enolase-1(ENO1), a glycolytic enzyme,and we confirmed that results by silencing ENO1 using siRNA. It was also confirmed through competition assay between anti-CM1 and anti-ENO1 mAbs. Finally, we investigated the possible role of CM1 in inflammatory response and cancer. The ligation of CM1 on Raji cells with anti-CM1mAbs induces the extensive production of prostaglandin E2(PGE2). In addition, the increased activity of matrix metalloproteinase (MMP)-2/9 was shown in NCI-N87, stomach cancer cell line by CM1 stimulation. Conlusion: CM1 is identical to ENO1 and it might be an important role in the regulation of inflammatory responses.

        지목과 건축물용도간 불부합 현황 및 개선방안

        ㅂㄴ 연구는 지적공부상 지목과 실제의 토지이용현황간의 불부합이 발생하는 문제점을 해결하기 위해 불부합 현황분석을 토대로 표본조사에 따른 개선방안을 제시하였다. 분석결과, 조사대상 281필 중 건축물용도와 현장은 일치하나 지적공부상 지목과 현장은 불일치하여 지목변경을 필요로 하는 필지가 신암동 121필, 용제동 110필로 총 231필로 나타났다. 또한 지적공부상 지목과 현장은 일치하나 건추물용도와 현장은 불일치하여 지목변경을 필요로 하지 않는 필지가 신암동32필, 용계동 18필로 총 50필로 나타났다. 따라서 토지대장의 지목과 건축물 용도간의 지목불부합지 해소를 위한 실증적인 개선방알을 법.제도적, 행정적, 홍보적 측면에서 제시하였다. In this study was presented a improvement of method based on the sample survey with analyzed of cadastral discrepancy situation that in order to solve the problem of cadastral discrepancy between the actual land use and cadastral record on the land category. As a result, Object of investigation the 281 parcels of land use are inconsistent with the Building Usage, But the cadastral record and land use are inconsistent that need to change land category was a total 231 parcel with SINAM-Dong 121, Yonggye-Dong 110. Also, used on cadastral land category and land use and building Usage is consistent, but the parcels of land are inconsistent that does not require a change land category a total of 50 parcels, with SINAM-Dong 32 parcels, Yonggye-Dong 18 parcels. Therefore, In the land register of the land category and in the building register of the building usage discrepancy parcel base of the two dong in most sample surveys with a focus on cadastre and building usage cadastral discrepancy that was presented as empirical solve for the in the aspects of improvement of law and institutional, administrative, public relations.

        내러티브 메모리얼의 경험공간 구조에 관한 연구

        This study aims to investigate the spatial structure of the memorial in terms of experiential space, targeting the ‘narrative memorial’, which has its own unique identity. The narrative memorial, which has a defined path to deliver story of the events through space, has a different order of experience different to the order of floor plan, so it is necessary to understand the space structure from the perspective of the visitor"s experience. Therefore, the research was conducted by using "visitor journey map" originated from "customer (user) journey map" which has been widely used in the service design field to check the actual experience of users in the entire process of using specific products or services. The selected memorials for the research are US Holocaust Memorial Museum, Yad Vashem Holocaust history museum, and Jewish museum Berlin, which are representative of the modern memorial. The main spatial structure of narrative memorial found in terms of experiential space is as follows: 1. Intersecting structure of emotional experience space and practical educational space that are repeatedly experienced, 2. Emotional experience space that forms step-by-step experiences, 3. The role of a connecting space that allows the memorial to become a background itself telling the story of event, 4. a spatial experience that forms different meanings according to the experience stage even in the same space, 5. a transitional space that generates a staged background change. In summary, in the narrative memorial, all spaces participate in the story of an event commemorated, and these experience spaces are repeatedly experienced with each other, forming a staged experience structure based on the intersection of experiences as a whole. In this way, it can be confirmed that the story and emotional experience of the event conveyed in the memorial are consistently delivered to the visitor, and the memorial acts as a medium of memory to be told as the space itself.

        재생 뮤지엄 공간에서의 방문객 경험 특성 연구 - 1990년대 이후 재생된 뮤지엄 공간을 대상으로 -

        This study aims to understand what makes visitor experience special in refurbished museum space. For this study, three characteristics of refurbished space -Placeness, Historicity, Creativity- and five characteristics of experiential design in space -Interaction, Senses, Dynamism, Reminiscence, Event- were derived from advanced research analysis. The research method is to analyse the correlation between each item using characteristics of the refurbished space as independent variables and the characteristics of experiential design in space as dependent variables. And four cases of refurbished museums-Tate modern, Red dot design museum (Essen), Punta della dogana museum, Neues museum-were selected for this analysis. Through this process, the research draws following characteristics of experiential design in refurbished museum space: First, as visitor enter the refurbished museum space from outside to inside, the placeness, historicity, and creativity tends to appear in stages. Second, according to this, reminiscence, dynamism, interaction, senses and event which have strong correlation with placeness, historicity, and creativity also gradually appear as visitor enter the space. Third, experience of the refurbished museum space is not a holistic experience of multiple touchpoints but a stratified experience of individual touchpoints in spaces. Four, refurbished museum can be seen that the time axis is combined with the layer of experience by the temporality of the refurbished space, so that the process of visitors experiencing the space is formed more multi-layered.

        디자인 스튜디오의 소규모 브랜드 공간 아이덴티티 구축에 관한 연구

        One of the characteristics that can be seen in the field of interior design today is the emergence of design studios. The characteristic of their design work is that it aims for total branding. In the past, such processes have been mainly applied to brand stores under companies, but with the advent of design studios, they have also begun to be applied to small scale brand stores. Commercial spaces that has gone through this process that achieve significant commercial results through SNS by establishing store identity. There may be differences in the elements that form store identity, but most of them are furniture, lighting, decoration, color, logo, finishing materials, etc. The important point here is that the elements act as integrated elements that form a style, not as individual beings. In this way, the elements the space form a mutually related relationship through total branding. In this study, The identity of space is viewed based on Whitehead’s aesthetic sensory theory. Based on this theory, it can be seen that different elements of space exist independently, but are recognized as a completed space through the theory of sensory theory. This shows that the design method called total branding plays a significant role in building spatial identity. Finally, analyzing the site that went through the branding work of the design studio through Whitehead’s sensory theory, it was confirmed that the harmony, space form, and function of the spatial components interact with each other to firmly build the functional and aesthetic identity that space identity.

        1930년대 소설 속에 나타난 경성의 도시공간적 특성이 반영된 시민일상연구 - 경성의 중심부의 공간 특성을 인용한 소설 속 공간과 일상을 대상으로

        This study aims to investigate the relationship between the characteristics of architecture and urban space that formed the urban landscape of the colonial city ‘Gyeongseong’ in the 1930s and 'everyday life' of those living in the city at that time. In colonial period, there was coexistence of ‘non-everyday life’ and ‘everyday life’. Studying everyday life of this period makes it possible to find a "gap in everyday life" that was not known from a macro perspective. In addition, it is meaningful to reconstruct history through everyday life of ordinary people who lived in that age as an individual rather than as a nation. It is difficult to find the scenery and everyday life of the 1930s in current Seoul, where landscapes of various eras are mixed due to the rapid development brought since the early 1900s. For this reason, this study used novels written by the eyes of that era as research data. A total of 75 sentences related to urban spaces of Gyeongseong were extracted from 9 novels published in the 1930s, and these are compared with the urban spatial characteristics of Gyeongseong classified from macro perspective. Although the landscape was formed by the modernisation and urban planning implemented by Japan, ordinary people generally used whole Gyeongseong as their daily space. And the influence of capital on people's everyday life was stronger than the plan for colonial rule. In addition, there were people who lost their way in modernised city and felt the tragedy of the era. Finally, the simultaneous changes and newly introduced cultures led to introduce of a new space, but those who lived at that time were able to see that they were surviving beyond their everyday life as a matter of survival.

        Z세대의 소비패턴과 플래그십 스토어의 체험적 특성 관계에 관한 연구

        The purpose of this study is to understand the correlation between the consumption patterns of Generation Z and the experiential characteristics of the flagship store space after the COVID-19 pandemic. This study has implications in terms of examining the development strategies of offline stores to keep pace with consumers and trends in a rapidly changing consumer market. The research method is to establish criteria for the case study analysis based on items such as consumer characteristics of Generation Z and flagship store experience characteristics derived from the literature review. The selected research subjects—five flagship stores—are explored in the field, and the subjects are analyzed through cross-evaluation based on the previously derived case study criteria to form a comprehensive discussion. Through this research method, the study concluded that the main consumption patterns of Generation Z are well reflected in the flagship store space established after the COVID-19 pandemic. The consumption patterns of Generation Z and the “non-daily” items in the flagship stores show a deep correlation. Following the “non-daily”, Generation Z’s consumption patterns are highly correlated with the “symbolism” and “dynamic” of flagship stores. In such a conclusion, in order to meet the diverse consumer needs of Generation Z, the flagship store has developed the space experience into a personalization type while maintaining the non-daily experience, leading consumers to interact with the space on a deeper level. At the same time, flagship stores need to build a digital-friendly experience environment where consumers can participate more actively and become an important means of effectively communicating brand messages.

        The Analysis of Vitamin C Concentration in Organs of Gulo−/− Mice Upon Vitamin C Withdrawal

        Background: Vitamin C is an essential nutrient for maintaining human life. Vitamin C insufficiency in the plasma is closely related with the development of scurvy. However, in vivo kinetics of vitamin C regarding its storage and consumption is still largely unknown. Methods: We used Gulo−/− mice, which cannot synthesize vitamin C like human. Vitamin C level in plasma and organs from Gulo−/−mice was examined, and it compared with the level of wild-type mice during 5 weeks. Results: The significant weight loss of Gulo−/− mice was shown at 3 weeks after vitamin C withdrawal. However, there was no differences between wild-type and vitamin C-supplemented Gulo−/−mice (3.3 g/L in drinking water). The concentration of vitamin C in plasma and organs was significantly decreased at 1week after vitamin C withdrawal. Vitamin C is preferentially deposited in adrenal gland, lymph node, lung, and brain. There were no significant changes in the numbers and CD4/CD8 ratio of splenocytes in Gulo−/− mice with vitamin C withdrawal for 4 weeks. And the architecture of spleen in Gulo−/− mice was disrupted at 5 weeks after vitamin C withdrawal. Conclusion: The vitamin C level of Gulo−/− mice was considerably decreased from 1 week after vitamin C withdrawal.         Vitamin C Is an Essential Factor on the Anti-viral Immune Responses through the Production of Interferon-α/β at the Initial Stage of Influenza A Virus (H3N2) Infection

        L-ascorbic acid (vitamin C) is one of the well-known antiviral agents, especially to influenza virus. Since the in vivo antiviral effect is still controversial, we investigated whether vitamin C could regulate influenza virus infection in vivo by using Gluo (-/-) mice, which cannot synthesize vitamin C like humans. First, we found that vitamin C-insufficient Gluo (-/-)mice expired within 1 week after intranasal inoculation of influenza virus (H3N2/Hongkong). Viral titers in the lung of vitamin C-insufficient Gluo (-/-) mice were definitely increased but production of anti-viral cytokine, interferon (IFN)-α/β, was decreased. On the contrary, the infiltration of inflammatory cells into the lung and production of pro-inflammatory cytokines, tumor necrosis factor (TNF)-α and interleukin (IL)-α/β, were increased in the lung. Taken together,vitamin C shows in vivo antiviral immune responses at the early time of infection, especially against influenza virus,through increased production of IFN-α/β.

        Vitamin C Up-regulates Expression of CD80, CD86 and MHC Class II on Dendritic Cell Line, DC-1 Via the Activation of p38 MAPK

        Vitamin C is an essential water-soluble nutrient which primarily exerts its effect on host defense mechanisms and immune homeostasis, but the mechanism related to immune- potentiation is poorly understood. Since dendritic cells (DCs) are known as a potent antigen presenting cell (APC) that could enhance the antigen specific immune responses, we investigate the effects of vitamin C on activation of DCs and its related mechanism by using dendritic cell lines, DC-1. First, we found that there was no damage on DC-1 by 2.5 mM of vitamin C. In the presence of vitamin C, the expression of CD80,CD86, and MHC molecules was increased, but it was decreased by the pre-treatment of SB203580, p38 MAPK-specific inhibitor. We confirmed the phosphorylation of p38MAPK was increased by the treatment of vitamin C. Taken together, these results suggest that vitamin C could enhance the activity of dendritic cells via the up-regulation of the expression of CD80, CD86, and MHC molecules and the activation of p38 MAPK is related to this process.
