        초음파진단기의 빔포밍 기술

        초음파진단기는 1950년대부터 사용되기 시작했고 그 동안 꾸준한 기술 발전을 통해 현재 대부분의 병원에서 필수불가결한 영상진단장비로 널리 활용되고 있다. 1970년대 초음파진단기에 어레이 프로브가 사용되기 시작한 이래로 전자적 신호처리를 통한 빔포밍 기술이 초음파진단기에 적용되었고, 꾸준히 개선되어 왔다. 빔포밍 기술은 초음파진단기의 해상도를 결정짓는 중요한 신호처리 기술이다. 이 논문에서는 이 빔포밍 기술의 원리부터 최근 동향까지 간략히 소개하고자 한다. 여기에는 어레이 프로브(array probe)를 사용하는 빔포밍의 원리, 기본적 이론, 실제 구현 등이 포함되고, 또 최근 기술 중 합성구경영상(synthetic aperture imaging: SAI), 적응형 빔포밍(adaptive beamforming), 2차원 어레이 프로브를 사용하는 2차원 빔포밍 기술 등의 주제도 소개한다. 이런 다양한 빔포밍 기술들은 다양한 다른 분야의 기술들과 여러 가지 형태로 발전적으로 융합하면서 시스템의 성능을 지속적으로 향상시켜 갈 것이다. Medical ultrasound systems have been used since 1950s, and are now widely used in most hospitals as indispensable diagnostic imaging systems. Since array probe was introduced in 1970s, beamforming technology using electronic signal processing has been adopted to the medical ultrasound system, and has been improved. Beamforming is a important technology which defines the resolution of the ultrasound system. In this paper, the technologies are introduced from basic beamforming principles to current trend. They include principles of beamforming using array probe, basic theory, and practical implementation, and recent topics of synthetic aperture imaging, adaptive beamforming, 2-dimensional beamforming using 2-dimensional array are also introduced. These various technologies will improve system performances continuously by merging innovatively with various technologies in other fields.

        제한효소인 Sdi I을 생성하는 Streptomyces 분리 균주의 수리동정

        Numerical identification was aplied for Streptomyces sp.264, an isolate producing a new restriction endonuclease Sdi I. The restriction enzyme would appear to be an isoschisomer of Xho I. Fifty taxonomic unit characters were tested and the data obtained were analyzed numerically by using the TAXON program. The isolate was identified to be the major cluster 19 of Streptomyces and best matched to S. diastatochromogenes. It was, therefore, concluded that the isolate was identified to be a member of Streptomyces diastatochromogenes.

        Arthrobacter simplex의 Steroid ${\Delta}^1$-dehydrogenase의 유도와 유도성 스테로이드의 성질

        Arthrobacter simplex IAM 1660의 스테로이드 $\Delta^1$-dehydrogenase에 대한 각종 steroid의 유도조건과 효과를 검토하였다. 그 결과 시험한 스테로이드 중 hydrocortisone이 가장 큰 효과를 나타냈고 progesterone, prednisolone, prednisone, androstenstendione의 순으로 유도효과를 보였고, 스테롤에 의해서는 유도효과가 낮았다. 유도 스테로이드의 특성으로는 스테로이드 모핵 C-3위가 수산기인 것보다 캐톤기이며 C-4위에 이중결합이 존재하고 스테로이드 D고리의 측쇄가 짧은 것이 유도 효과가 높았다. - Since steroid $\Delta^1$-dehydrogenase synthesis has been known to be inducible, the mechanism of the enzyme induction of Arthrobacter simplex IAM 1660 was investigated. Among various steroids tested for inducers, hydrocortisone was the most effective inducer when hydrocortisone was used as a substrate for steroid $\Delta^1$-dehydrogenase. Steroid $\Delta^1$-dehydrogenase synthesis was effectively induced by progesterone, prednisolone and androstenedione, while the enzyme was less induced by cholesterol and not by phytosterols. The results suggest that the presence of 3-keto group and short side chain of steroids are the favorable factors for the induction of the $\Delta^1$-dehydrogenase synthesis. The enzyme was induced at the highest level when hydrocortisone was added at early log phase to the concentration of 0.01% of the culture and the culture was grown for 15 hours.

        Cellulomonas 속 종간 원형질 융합체의 특성

        In order to investigate physiological characteristics of fusants by interspecific protoplast fusion of the genus Cellulomonas, protoplasts of Cellulomonas flavigena NCIB 12901 and Cellulomonas bibula NCIB 8142 were fused and cell wall regenerated. To give gene maker, C. bibula was treated with 500 ug/ml NTG for 1 hr and arginine requiring auxotrophic mutants were isolated. Protoplasts of the genus Cellulomonas were obtained by treatment with $600{\mu}{\textrm{g}}$/ml lysozyme, and 0.5M sorbitol was optimal for osmotic stabilizer on protoplast fromation. Protoplast fusion was enhanced by 40% PEG)M.W.6,000) containing 25 mM $CaCl_{2}$ at $30^{\circ}C$ for 30 min and fusion frequency between C. bibula and C. flavigena was $5\times 10^{-4}$. Processes of protoplast formation, cell wall regeneration and protoplast fusion were obsdrved by scanning electron microscope. By comparing enzyme activities of cellulase, exocellobiohydrolase, .betha.-glucosidase of the parent strains of Cellulomonas with those of thier mutants and fusants, fusants with increased enzyme activity were obtained. By the studies on nutritional requirement, antibiotic resistance, cellulolytic enzyme activities, type of peptidoglycan and motility of two mutants and fusants, fusants were proved to be recombinant of both mutant strains. 섬유소를 분해할 수 있는 Cellutomonas flavigena NCIB 12901 과 Cellulomonas bibula NCIB 8142의 종간 원형질체 융합을 하고 그로부터 얻은 융합체의 특성을 조사하였다. C. bibuta의 원형질체는 lysozyme(600Ieιllulomoηas libula NCIB 8142의 종간 원형 섣처l 융 한응 하고:1로부터 얻은융합셰익 특성을 조사하였다 . C bibula의 원형선셰는 lysozyme $600{\mu}{\textrm{g}}$/ml)를 6시간 처리하여 얻었으며, 삼투안정제로는 O.5M sorbitol의 최저이었고 원형질체 재생률은 6.7%였다. 원형질체 융합은 25mM $CaCl_{2}$를 포함하는 40% PEG (M.W.6000)를 $30^{\circ}C$에서 30분간 처리함으로 수행하였고 C. bibuιl와 C. flavigeηa의 융합 빈도는 $5\times 10^{-4}$으로 원형질체 형성 및 합체를 얻었고, 모균주와 융합체의 영양요구성, 항생물질 내성, 효소활성, 세포벽 성분, 운동성 등을 비교하여 융합체는 모균주의 재조합체임을 밝히었다.

        纖維質 資化性菌의 分子育種에 관한 硏究 -Cellulomonas속균의 ${\beta}$-glucosidase gene의 E. coli에의 cloning -

        The cellabiase (${\beta}$-glucosidase) gene in a Cellulomonas sp. CS1-1 was cloned into E. coli HB101 using the vector plasmid pBR322, and the expression of the gene in E. coli studied. The chromosomal DNA of the cellulomonas was digested by seveal restriction enzymes, each of which has only one cleaving site in plasmid pBR322. The recombinant plasmid, pSB2, created with Sal I frament, was expressed for the cellobiase gene in E. coli. The recombiant plasmid was estimated to contain 6.4 Kb foreign DNA at the Sal I site of plasmid pBR322 and the inserted DNA was mapped by single and double digestion with several enzymes. E. coli HB101(pSB2) has slowly grown in a mineral liquid medium containing cellobiose as a sole carbon source. The cellobiase activity in the transformed E. coli was 132 units per liter, which is equivalent to one twenty fifth of that in doner strain Cellulomonas sp. CS1-1. The transforned cell with plasmid containing cellulase gene grow well in the LB mediuns. The synthesis of cellobiase in the strain, E. coli HB101 (pSB2), was inhibited by glucose and at high concentration of cellobiose, and induced by cellobiose at low concentration.

        미생물에 의한 핵산관련물질의 생산에 관한 연구 1

        As the first step in the production of nucleic acid derivatives by microorganisms, adenineless mutants were derived from Brevibacterium ammoniagenes ATCC6872. A culture of Br. ammoniagenes was exposed to ultraviolet rays for 120 second and treated with diethylsulfate in phosphate buffer for 2 hours to reach the designed death rate. The yield of mutants induced was 0.28% by the ultraviolet irradiation and 0.66% by the diethylsulfate treatment. By the diethylsulfate treatment. By the treatment of penicillin G in a hypertonic minimal medium, the yield of mutants was increased from 0.28% to 0.54% and from 0.66% to 1.5%, respectively. Thus, in was demonstrated that diethylsulfate treatment was much more efficient than UV irradiation to induce adenineless mutants of the bacteria, and total strains of 120 adenineless mutants were obtained.

      • 내열성 제한효소인 Svi Ⅰ을 생성하는 Streptomyces 분리균주의 수리 동정

        새로운 제한효소인 Svi I을 생성하는 Streptomyces속 한 균주(D2-5)를 토양으로부터 분리하여 형태적 관찰 및 수리동정을 실시하였다. 50개의 분류 단위 형질을 분석하였고 이 실험결과를 TAXON program에 적용하여 종의 수리 동정을 실시하였다. 그 결과 분리균 D2-5는 Streptomyces의 제 18 주군집에 속하며 이 군집에 속하는 S. violochromogenes와 유사성이 가장 높은 것으로 나타났다. 따라서 분리주는 Streptomyces violochromogenes의 한 균주로 동정하였다. Numerical identification was carried out for an isolate of Streptomyces D2-5 producing a new restriction endonuclease Svi I. Fifty taxonomic unit characters were tested and the data were analyzed numerically using the TAXON program. the isolate was best matched to Streptomyces violochromogenes in the major cluster 18 of Streptomyces. Therefore, it was concluded that the isolate was identified to be a member of Streptomyces violochromogenes.

        Rhodopseudomonas sphaeroides K-7에 의한 수소생성에 미치는 생리적 조건에 관한 연구

        Physiological conditions for hydrogen evolution by Rhodopseudomonas sphaeroides K-7 are examined. Larger amount of molecular hydrogen was evolved at $30^{\circ}C$, pH6.8-7.0 under anaerobically illuminated condition of about 12, 000 lux by the organism. The heighest rates of hydrogen evolution were observed in the culture with the organic acids such as acetate, DL-lactate or DL-malate in media containing L-glutamate as a nitrogen source. Hydrogen was also evolved from glucose with the rate of $178.9\;{\mu}l/hr/mg$ cells (dry weight). When glucose was adopted a a sole carbon source, however, conside erable time lag of about 20hours was required for hydrogen evolution. The resting cells stored at $30^{\circ}C$ under argon maintained the rate of hydrogen evolution in nearly about 90% of initial one even 40 days of storation. R. sphaeroides K-7에 의한 수소 생성에 미치는 생리적 조건을 조사한 결과 수소생성은 혐기성조건하에서 $30^{\circ}C$, pH 6.8-7.0s 조명도 약 12,000 lux을 쉰지하였을 때 할발히 이루어지는 것으로 내타났다. 아미노산으로서는 글루타민산, 유기산으로는 초산, 젖산, 혹은 사과산을 이용했을 때 높은 수소생성이 나타났으며 포도당과 같은 당류에서도 수소생성이 이루어졌다. 휴지기 세포는 약 40일 보관후에도 90% 정도의 수소생성능을 유지하고 있었다.

        Purification and Characterzation of a Restriction Endonuclease from Pseudomonas syringae pv.phaselicola

        A restriction endonuclease, PsyI, has been isolated from Pseudomonas syringae pv. pha- seolicola, and its catalytic properties have been studied. This enzyme was purified through strepto- mycin sulfate and ammonium sulfate fractionation, phosphocellulose Pll, DEAE-cellulose, hydroxy- apatite and Sephadex G-100 column chromatography. It's molecular weight was about 50,000 dalton as determined by 7.5% polyacrylamide gel electrophoresis containing 0.1% SDS. In catalytic proper- ties, PsyI shows stable at wide ranges of pH between 7.0 and 10.0, of temperature between 30$\circ$C and 37$\circ$C, and its thermal stability is between 25$\circ$C, and 45$\circ$C, at the presence Of 10 mM MgCl$_{2}$-PsyI essentially require Na salt for enzyme reaction, is rather inhibited in the high Na salt concent- ration. The presence of 2-mercaptoethanol is absolutely required for the enzyme activity. This endonuclease, PsyI was determined to be an isoschizomer of SalI from the results of the restriction mapping and DNA sequencing.
