To test the protective effect of herbal medicine on myocardial damage against adriamycin-induced myocardiotoxicity, cytotoxicity of adriamycin was examined using MTT and SRB assay in the presence of water extract of Gamdu-tang, Gamigamdu-tang, or single constituents of these proscriptions. Myocardial toxicity was evaluated in neonatal mouse myocardiocytes in cultures. The results of these experiments were obtained as follows : Adriamycin resulted in a decrease in viability and total protein in cultured myocardial cells. Gamdu-tang water extract shows effects of protection from the cardiocyte toxicity induced by adriamycin treatment such as increases in viability and protein synthesis. Gamigamdu-tang water extract shows the similar protective effect as shown in gamdu-tang treatment on the cardiotoxicity induced by adriamycin. Radix Glycyrrhizae water extract shows a significant effect of protection for cardiotoxicity induced by adriamycin in cultured myocardial cells. Semen Glycine water extract has a significant effect of protection for cardiotoxicity induced by adriamycin in cultured myocardial cells. Folium Phyllostachyos water extract has a significant effect of protection for cardiotoxicity induced by adriamycin in cultured myocardial cells. These results show that adriamycin elicits toxic effects in cultured myocardial cells derived from neonatal mouse, and suggest that water extract of gamdu-tang, gamigamdu-tang, Radix Glycyrrhizae, Semen Glycine, or Folium Phyllostachyos is very effective in the prevention of adriamycin-induced cardiotoxicity.

Myocardial toxicity was evaluated in neonatal mouse myocardiocytes in cultures. The results of these experiments were obtained as follows ; Adriamycin resulted in a decrease in viability, increases in LDH activity in cultured myocardial cells. Gamdu-tang water extract shows effects of protection from the cardiocyte toxicity induced by adriamycin treatment such as decreases in lactate dehydrogenase activity. Gamigamdutang water extract shows the similar protective elect as shown in gamdutang treatment on the cardiotoxicity induced by adriamycin. Single constuents of these prescriptions shows a significant effect of protection for cardiotoxicity induced by adriamycin in cultured myocardial cells. These results show that adriamycin elicits toxic effects in cultured myocardial cells derived from neonatal mouse, and suggest that water extract of gamdu-tang, gamigamdu-tang, Radix Glycyrrhizae, Semen Glycine, or Folium Phyllostachyos is very effective in the prevention of adriamycin-induced cardiotoxicity.
Structural Study of Monomethyl Fumarate-Bound Human GAPDH

Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) is a core enzyme of the aerobic glycolytic pathway with versatile functions and is associated with cancer development. Recently, Kornberg et al . published the detailed correlation between GAPDH and di- or monomethyl fumarate (DMF or MMF), which are well-known GAPDH antagonists in the immune system. As an extension, herein, we report the crystal structure of MMF-bound human GAPDH at 2.29 Å. The MMF molecule is covalently linked to the catalytic Cys152 of human GAPDH, and inhibits the catalytic activity of the residue and dramatically reduces the enzymatic activity of GAPDH. Structural comparisons between NAD+bound GAPDH and MMF-bound GAPDH revealed that the covalently linked MMF can block the binding of the NAD+ cosubstrate due to steric hindrance of the nicotinamide portion of the NAD+ molecule, illuminating the specific mechanism by which MMF inhibits GAPDH. Our data provide insights into GAPDH antagonist development for GAPDH-mediated disease treatment.
