The experiment was carried out to investigate some factors affecting mycelial growth of Pleurotus sajorcaju and P. ostreatus. The optimal temperature for mycelial growth of both the species were from 25 to 30℃ whereas the formation of primordia of the sporophores was 20 to 25℃ in P. sajor-caju and 15℃ in P. ostreatus respectively on potato sucrose yeast agar medium. The optimal pH of the medium for the mycelial growth of both the species were ranged 5 to 6. The effect of carbon and nitrogen sources on mycelial growth of the species were studied using modified Waksman's special medium as the basal medium with the test substance. Nine carbon and eight nitrogen sources were evaluated by replacing the carbohydrates or nitrogen in the basal medium. Generally, disaccharides and polysaccharides showed good effect for the mycelial growth of P. sajor-caju, but polysaccharides were superior to the other classes of carbon sources for the mycelial growth of P. ostreatus. Generally, organic nitrogen compounds showed good mycelial growth of both the species. The mycelial growth of the species were favored by ammonium more than by nitrate nitrogen source. Asparagine was superior to the other kinds of amino acid. Six percentages of glucose and 0.6% of ammonium tartrate was optimum for mycelial growth of P. sajor-caju whereas the optimum concentration for the P. ostreatus was 4% of glucose and 0.4% ammonium tartrate.

This experiment was performed to select the mast suitable species of oyster mushroom for cultivation during summer season in Korea. The selection was made based upon mycelial growth on the ricestraw substrates, fruiting under hot weather conditions during the summer season in Korea and potential high productivity using six cultures from South-east Asia. The newly selected strain of Pleurotus sajor-caju (ASI 2070) grew well on the rice straw substrates and produced the higher yields of sporophores than other cultures tested. The primordia of the mushroom were formed between 10 to 35℃. The optimal temperature for fruiting ranged from 20 to 25℃.

Some factors affecting the protoplast release from mycelia of Plurotus ostreatus using commercial lytic enzymes were investigated. The highest yields of the protoplast were obtained from four days old mycelia grown in mushroom complete medium. The solution of 0.8M MgSO₄ or KCl showed good results as the osmotic stabilizer for releasing the protoplast. Novozym 234 was the most effective among commercials tested. The concentration of the enzyme and pH of the enzyme solution were optimal at 15㎎/㎖ and 5.5∼6.0 for the protoplast release, respectively. Mycelial digestion was optimal at about 28℃ and was better in the reciprocal shaking bath (75 oscillations/min) than the stationary culture.

Interspecific heterokaryons were obtained between auxotrophic mutants of Pieurotus ostreatus and Pleurotus florida by polyethylene glycol-Ca^(++) induced fusion of somatic protuplasts. The fusion products produced colonies of dense growing mycelium after 10-14 days culture on hypertonic Mushroom Minimal Medium. When they were transferred to Mushroom Minimal Medium plates, the sectors showed normal vegetative morphology but the colonies of irregular shape varied in growth rate. All of the colonies produced fruit body of normal pilei. Some heterokaryon colonies bearing none or only a small amounts of basidiospores were isolated. Fusion products, generally, gave higher basidiocarp yields than the parents.
