        The main focus of family therapy is to look at a family as a one unit and understand overall context of a family issue. Since family is formed by family members, a negative interaction among family members causes an adverse effect on family itself. Therefore, individual's family has impact on his issues and vice versa. As the problem does not end with an individual, the family is considered as a treatment target. From Bowen's family theory, where he tries to find a cause of a family problem internally and externally, we can assume that a family isn't just an individual unit but a unit with an impact of a past. Bowen does not just look at the person's current problem but also look at the person's past relationship and even understand his parents' or even his grand parents' lives. From this theory, we can see that the paranoia symptom that ran in the family have caused Prince Sado to become pathological. "Differentiation of Self" and "Multi-generational Transmission Process" concepts, from Bowen's family therapy theory, were very effective in analyzing Prince Sado's family. The signs of personality problem of Sukjong, Gyeongjong, Yeongjo and Sado, which are shown in literatures,can be explained by Bowen's multi-generational transmission process concept. Person with a poorly differentiated "self" creates fake image of oneself. A person not differentiated himself from his family, has high possibility to shape its individual "self" similar to that of his family's. Bowen's theory also shows that through interaction between the family members, attitude and principles of the members can change. For instance, Jungjo, son of Prince Sado, had a mother, Lady Hong at Hye Kyung palace, who had well-differentiated "self." With much help from his mother, Jungjo was able to evade many problems, such as emotional reactivity, compulsion, paranoia and mental, that ran in the family since Sukjong. Unlike previous kings, Jungjo was able to achieve well-differentiated "self" which allowed him to rule the country well. Typically, a member of family has much impact from another member in the family. Yeongjo had his only son, Sado, at the age of forty two. His ambition to raise his son well hasmade him to meddle in and scold on everything Sado was doing. He wanted Sado to only follow and honor his will. But Sado's compassion toward previous King, Kyungjong, made Yeongjo unrest. Closeness of father-son relationship in the royal family was tied to life and death as such relationship was considered very important in Chosun dynasty. Yeongjo considered Prince Sado as a part of him. Like him, Yeongjo wanted his is son to be very through and minute. Much demands from Yeongjo made Sado to become mentally ill. As Sado became a disappointment, Keongjong ordered Sado to be sealed alive in a large rice chest to die. The tragedy of Prince Sado makes many suggestions to current family lives. In a competitive society, it’s natural for parents wanting their children to live a better life than they have. This makes parents, unknowingly, push their children over the limit or even make their issues children's. This dissertation showed that it is possible to see various family problems in our society from afar by using Bowen's theory on Prince Sado's family.

      • 나팔꽃의 감성적 표현 연구 : 연구자의 작품을 중심으로

        예술은 자연과 인간을 연결해 주는 매개의 역할을 한다. 영국의 대표적 철학자 허버트 리드(Herbert Read)는 '자연과의 진정한 조화를 이룬다는 것은 하나의 생명을 얻는 것과도 같다.'고 말하였다. 이와 같이 예술은 진정한 자연과의 교감이 이루어 질 때 미의식이 충만해지며 자아를 만나게 되고 정신적인 만족감을 느끼게 된다는 것을 알 수 있다. 따라서 예술가는 어떤 재료를 사용하여 오로지 재현하는 것이 아니라 그가 받은 느낌이나 감동, 상상적인 반응을 표현하도록 노력을 해야 한다. 본 연구자는 자연의 수많은 대상 중 우리 주변에서 흔히 볼 수 있고 쉽게 접할 수 있는 나팔꽃을 소재로 정하여 보았다. 꽃이라는 대상으로부터 전해지는 새로운 이미지를 통해 본인의 내면세계를 솔직하게 만나보고 인간의 참된 모습을 찾아보고자 하였다. 본인은 나팔꽃을 직접 심어서 물도 주며 자라는 과정을 관찰해봄으로 몸소 체험하는 경험도 가져 보았다. 꽃봉오리의 성장 과정과 만개하여 피어나는 꽃 그리고 꽃의 시들음 속에서 인간이 태어나고 성장해 가며 죽음으로 소멸해 가는 과정이 인간의 삶과 비슷하게 느껴져 서로 유기적인 관계에 있다는 점을 착안하여 본 논문을 쓰게 되었다. 작품의 정신적 배경으로 동양의 사상을 살펴보고 연구해 보니 동양의 자연관은 자연과의 조화 겉으로 드러난 자연현상 뿐 만 아니라 자연의 의미 속에 존재하는 순화되고 동화되며 인간의 정신적 염원도 담고 있다는 것을 알 수 있었다. 화면 속에는 여러 가지 형상들이 존재하는데 연구자는 나팔꽃 속에 숨겨진 점·선·면 그리고 공간적 여백과 같은 순수 조형성을 발견하여 표현 가능성을 열어 보고자 하였다. 그 꽃의 형상을 그대로 복사하는 것이 아니라 본인이 느끼고 감동한 바를 자신의 예술 의지대로 자유롭게 표현하고자 하였다. 작업과정에서는 다양한 재료와 표현 방법을 시도하는 것이 조금 이른 느낌이 들어 전통적 회화방법을 먼저 연구하고 재해석하여 표현해 보았다. 장지에 색감이 우려 나오는 효과를 얻기 위해 아교포수를 하였는데 아교와 물의 비율은 계절과 온도 그리고 습도에 따라 많은 차이가 났으며 잘못된 사용으로 그림이 부패되고 손상되므로 자신만의 정확한 량을 만들어 작업하는 것이 중요하다고 사료된다. 채색은 일정기간 변하지 않으며 다작할 수 있는 접시물감과 튜브물감을 사용하였고 깊고 오묘한 맛을 내기 위해 서로 섞지 않고 색조를 한 단계 낮춰 칠하였다. 산만해 지기 쉬운 분위기는 전체적인 흐름을 보며 자연스럽게 마무리 해나갔다. 구성적 요소로 동양화의 특징인 여백을 적극 활용해 보았다. '그림은 다 그렸어도 뜻은 남아 있다.'라는 말이 암시하듯 표현 할 수 없는 뜻을 여백으로 남겨두었다. 작품초기에는 왕성한 의욕으로 무엇인가 채우고 싶은 마음에 수많은 고민을 하였는데 지금 와서 생각해보니 성급히 표시하지 않는 정적인 아름다움이 있는 여백으로 남기지 않았더라면 화면이 답답하며 막혀있는 그림이 되었을 것이다. 그래서 본인 작품에 있어 여백은 무한한 상상의 세계를 암시하는 공간으로 감성적 이미지를 부여하는 중요한 구성요소가 되었다. 연구 결과, 대상에서 느끼는 감성은 개인마다 다르고 시시각각 변하는데 이러한 감성적 느낌을 보다 직접적으로 전달하기 위해서 음악적 요소나 언어를 구체화 시킨 시 그리고 선, 형, 색채 등 조형적 요소를 가지고 있는 회화적 공간을 적극적으로 활용한다면 본인의 자율적인 내면의 심상을 좀 더 가깝게 전달할 수 있다는 것을 알게 되었다. 본 논문을 통하여 꽃이 갖고 있는 색과 형태에서 오는 단순한 이미지가 아닌 나팔꽃을 보면서 느껴지는 삶의 이야기로 되돌려 보았고 또한 나의 내면의 감정을 자유롭게 표출할 수 있는 모체가 되어준 나팔꽃을 통하여 예술의 의미를 확인하는 계기가 되었다. 또한 본인 작품 세계에 좀 더 솔직하지 못하고 체계적이지 못했던 자신에게 유익한 시간이 되었으며 앞으로는 눈으로 흥미 있는 그림보다는 본인의 주관에 의해 다시 구성된 새로운 조형 시각을 형상화 하고자 노력하겠다. Fine arts work as a midway connecting the nature and human beings. Sir. Herbert Read, a well-known British philosopher said, 'being truly harmonious with the nature gains you another life.' Spiritual satisfaction along with aesthetic consciousness are realized, like this, when we are in true harmony with the nature. Therefore, artists should be focused to realize and express the feelings, impressions and imaginary reactions, not just reproduce with certain materials. The researcher then picked up morning glory, one that we can commonly find in our everyday life and we are familiar with, among numerous natural objects. This is to find the new images from a flower and to further seek the deeper understanding of my own inner self. The researcher herself raised a morning glory to observe how it grows. Focusing on the point that the growth and fall of the blossom are synchronized with the birth and death of human beings, I came to write this thesis. Studying oriental ideas as a spiritual background of works, it is found that the oriental view of the nature understands the human spiritual desires under the meanings and ideas of the nature as well as seeking for harmony with the nature in the surface. There exist various shapes in the screen, and the researcher explored to further express ideas by finding points, lines and faces hidden in a morning glory as well as pure plasticity such as spatial blanks. Not just copying the shapes of the flower, but expressing the feelings and impressions I had was the goal of the exploration. During the work process, I decided to first study and reinterpret the traditional painting methods, thinking that it is early to try various materials and methods now. To smoothly color the work, I glued the paper and found out that the ratio between glue and water differ a lot according to seasons, temperatures and humidity. It is also considered very important to find the exact proper amount, as inappropriate gluing damages and decays the paints. Dish colors and tube colors were used to avoid distortion and for diversification. For deeper coloring was realized with leveling coloring. Focused composition was also realized with overall harmony. For compositional completeness, spatial margins and blanks were actively utilized.'Even done, there left meanings'implies that unexpressable meanings are realized with blanks. In the early days, I had too many ideas to fill the screen, but now I realized that unless long-lasting motionless beauty was left with blanks, it must have become a blocked painting with stuffy screen. In my own works, blanks are important compositional objects to imply unlimited imaginary world. As a result of the study, it is understood that impressions from objects depend on individual feelings and are always subject to change, thus language-shaped poems, musical elements, and imagery components like lines, shapes and colors for active utilization further delivers the inner impressions. With this thesis, a flower’s colors and shapes are transferred into life stories, not just simple images from morning glories, and further I redefine the meanings of artworks for myself by understanding the free expressions of inner impressions through morning glories. Also, the study was very meaningful to make myself be more straight-forward and systematic for my own works. I will further make my best efforts to realize and embody the newer compositional views I make with my own view.

      • 다중지능을 활용한 수준별 수업이 학업성취도와 태도에 미치는 효과

        The purpose of this study was to investigate the effects of multiple intelligence-based differentiated instruction on the academic achievement and attitude of the 4th grade elementary school students. The specific research problems of interest in this study were as follows: First, what is the effect of multiple intelligence-based differentiated instruction in the 4th grade elementary Korean class on the academic achievement? Second, what is the effect of multiple intelligence-based differentiated instruction in the 4th grade elementary Korean class Korean on the attitude? The main hypotheses tested were as follows: (1) The group of multiple intelligence-based differentiated instruction would be higher than the group of traditional instruction on the academic achievement in Korean. (2) The group of multiple intelligence-based differentiated instruction would be higher than the group of traditional instruction on the attitude. The subjects of this study were 60(2 classes) 4th grade students of B elementary school in K city, Gyeongsangbuk province. Each class was composed of 30 students and assigned to the experimental and control group. Those two groups were turned out as homogeneous at the academic achievement and attitude on the pretest. The experimental group was provided multiple intelligence-based differentiated instruction by the unit of ‘Like This If I Am’ and ‘Helping Each Other’ in Korean, while the control group was provided traditional type of instruction. The experimental group participated in the differentiated instruction based on multiple intelligence theory for four weeks. After the experimental treatment, the posttest of academic achievement and attitude were administered to verify the effect of treatment, and the data collected were analyzed with T-test methods by using SPSS 12.0 for Windows. The significant level was 5%. And the data of student's reaction and teacher's observation through the differentiated instruction based on multiple intelligence theory were collected and analyzed. The main findings obtained from the results and discussion of this study were as follows: (1) There was no significant difference on the academic achievement between the group of multiple intelligence-based differentiated instruction and that of traditional instruction in Korean. But it was revealed that many students were interested in multiple intelligence-based differentiated instruction and preferred this type of instruction. Therefore, if the study were carry out during the long period, multiple intelligence-based differentiated instruction would have a positive effects on the academic achievement in Korean. (2) There was a significant difference on the attitude between the group of multiple intelligence-based differentiated instruction and that of traditional instruction in Korean. The group of multiple intelligence-based differentiated instruction was higher than that of traditional instruction on the attitude in Korean. The results of this study suggest that multiple intelligence-based differentiated instruction have a positive effects on the attitude in Korean. In this study, quantitative data analysis were performed mainly and then qualitative analysis were used partly. In the following study, the various types of qualitative research or mixed methods of quantitative and qualitative research need to be used to investigate the relevant themes more thoroughly.

      • Development of simultaneous analytical method for quantitation of polysaccharide in meningococcal vaccine

        Meningococcus is classified into 13 serogroups according to the specific sugar structure in the capsule, and the meningococcal vaccine is made based on the specific polysaccharides of the serogroup that causes invasive diseases. Therefore, WHO recommends measuring the polysaccharide content of meningococcal as a method to evaluate the quality, stability and effectiveness of meningococcal vaccine. In the case of meningococcal vaccine, as a biological product, it must undergo a national lot release before distribution in Korea. As a test item for national lot release, quantitation of free and total polysaccharide in vaccine is being performed, but until now, there is no official test method for national lot release. So the national lot release has been based on the manufacturer's test method. In the case of (A, C, W135, Y)-CRM197 protein conjugate vaccine, one of the domestically approved meningococcal vaccine, an analysis method using HPAEC-PAD is used to measure the polysaccharide quantity. However, the existing test method of (A, C, W135, Y)-CRM197 protein conjugate vaccine has disadvantages that it is difficult to manipulate and maintain sample pretreatment and analysis equipment, and there is no internal standard. For these reasons, the test result for quantitation of polysaccharide has low reproducibility. In addition, since the tests for serogroup C, W135, Y are performed separately, the test takes a long time. Therefore, this study was carried out with the aim of developing a simultaneous analytical method that can shorten the period of total polysaccharide content test for serogroup C, W135, Y and increase the reproducibility of the test result. In order to develop a simultaneous analytical method, research was carried out in two stages. First, a simultaneous analytical method for quantitation of polysaccharide of serogroups C, W135, Y was designed to complement the problems of the existing test methods. For this, internal standards and quantitative standards were selected and a new standard curve was set using quantitative standards. In addition, based on the existing literature, a hydrolysis condition for three serogroups was set and this can have a recovery close to that of a positive control standard. And SOP(draft) was produced based on this hydrolysis condition. Next, validation of the developed simultaneous analytical test method was performed to examine the feasibility of the simultaneous analytical method. As a result of the study, Fucose was selected as an internal standard, and a quantitative standard for each serogroup was prepared to set a new standard curve for the simultaneous analytical method. In addition, the common hydrolysis condition for the three serogroups was found (2 M, 90 ℃, 2h 30 min). The developed simultaneous analytical method was evaluated by six validation parameters of accuracy, linearity, specificity, precision, range, and robustness in accordance with guidelines for validation of drug testing methods, internationally harmonized with the ICH. As a result, all criteria of validation parameters were satisfied. Accordingly, the simultaneous analytical method developed in this study effectively shortened the test time for quantitation of total polysaccharide and improved the reproducibility of the test. Ultimately, the developed simultaneous analytical method can be applied to lot release for the (A, C, W135, Y)-CRM197 protein conjugate vaccine. 수막구균은 캡슐에 있는 특이적 당 구조에 따라 13 가지 혈청군으로 분류되며, 수막구균 백신은 침습적 질환을 일으키는 혈청군의 특이적인 다당류들을 기반하여 만들어진다. 따라서 WHO는 수막구균 백신의 품질, 안정성, 유효성을 평가하기 위한 방법으로 수막구균의 다당류 함량 측정을 권고하고 있다. 수막구균 백신의 경우, 생물학적제제로써 국내 유통 전 국가출하승인 검정시험을 거쳐야 하며 국가출하승인 검정시험 항목으로 유리 다당류 와 총 다당류에 대한 다당류 함량 시험이 수행되고 있으나, 현재까지 국가출하승인 공식시험법이 없어 제조사의 시험법에 따라 시험을 진행하고 있다. 국내 허가된 수막구균 백신 (A, C, W135, Y)-CRM197단백접합백신의 경우, 다당류 함량 측정을 위해 HPAEC-PAD를 이용한 분석법이 사용된다. 그러나 (A, C, W135, Y)-CRM197단백접합백신의 기존 시험법은, 시료의 전처리 및 분석 장비의 조작 및 유지가 까다롭고, 내부표준품이 없어 시험결과의 재현성이 낮은 단점이 있다. 또한 혈청군 C형과 혈청군 W135, Y형 다당류 분석이 따로 이루어지므로 시험에 장시간이 소요된다는 단점이 있다. 따라서 본 연구는 해당 백신의 혈청군 C, W135, Y형의 총 다당류 함량시험에 대한 기간을 단축하고 시험의 재현성을 높일 수 있는 동시분석법을 개발하여 국가출하승인검정 시험에 활용하는 것을 목표로 수행되었다. 동시분석법을 개발하기 위하여 크게 2 단계로 연구를 수행하였다. 우선, 기존 시험법의 문제점을 보완하여 혈청군 C, W135, Y에 대한 총 다당류 함량 동시분석법을 설계하였다. 이를 위해 내부표준품 및 정량표준품을 선정하고 이를 이용해 새로운 표준곡선을 세팅하였다. 또한 기존 문헌을 바탕으로 세 가지 혈청군에 대하여 동시에 양성대조표준품에 가까운 회수율을 가질 수 있는 가수분해 조건을 설정하고 이를 바탕으로 SOP(안) 등을 제작하였다. 다음으로, 개발된 동시분석시험법에 대한 밸리데이션을 수행하여 동시분석법의 타당성을 검증하였다. 연구 결과, 내부 표준품으로 Fucose를 선정하고 각 혈청군에 대한 정량 표준품을 제조하여 동시분석법을 위한 새로운 표준곡선을 세팅하였다. 또한 세 가지 혈청군에 대한 공통 가수분해 조건으로 (2 M, 90 ℃, 2h 30 min)을 찾았다. 개발된 동시분석법은 ICH 가이드라인과 국제조화된 ‘의약품 등 시험방법 밸리데이션 가이드라인’에 따라 정확성, 직선성, 특이성, 정밀성, 범위, 완건성의 6 가지 파라미터를 통해 평가되었으며, 모든 항목에서 기준을 만족하였다. 이로써, 다당류 함량 측정을 위해 본 연구에서 개발된 동시분석법은 해당 백신에 대한 시험 시간을 효과적으로 단축하고 시험의 재현성을 높였음을 확인할 수 있었다. 궁극적으로, 개발된 동시분석법을 (A, C, W135, Y)-CRM197단백접합백신의 국가검정시험에 적용할 수 있음을 확인하였다.

      • 加熱處理 우유배지에서 Lactobacillus bulgaricus의 形態變化에 關한 硏究

        This study was carried out to investigate morphology and activity variation of Lac. bulgaricus, which is used as a starter of fermented milk, in different heat treatment of milk medium and addition of various minerals. The results could be summerized as follows: 1. In the heat treated-milk medium at 121℃ for 15 min, the acid production was higher and the cell length was shorter than in that of below 100℃. 2. When the heat treated-milk medium at 100℃ for 15 min was incubated for 12 hours, pH, titeatable acidity and the number of viable cells were 4.4, 0.65%, 3×10^(8)/ml, respectively. 3. When the heat treated-milk medium at 121℃ for 15 min was incubated for 12 hours, pH, titratable acidity and number of viable cells were 3.9, 0.9%, 2×10^(9)/ml, respectively. 4. In the heat treated-milk medium at 100℃ for 15 min, the cell was elongated, septum was not formed and nuclear was normal. 5. When formic acid added to the heat treated-milk medium at 100℃ for 15 min, the number of viable cell was higher than no addition of formic acid. 6. Regardless of heat treatment condition, when heat treated-milk medium with 2% phosphate was contents incubated for 8 hours, the number of viable cell was decreased from 1×10^(8)/ml to 2×10^(5)/ml and cell was elongated. 7. In case of the addition of 0.01 - 1mM Ca++ to filamentous cell of Lac. bulgaricus, the cell recovered a normal rod cells from the filamentous cells.
