        인간의 Jurkat T세포에서 프로스타글란딘 PGE<sub>2</sub>) (PGE<sub>2</sub>)의 cAMP 경로를 통한 인터페론 감마(INF--γ ) 유전자의 methylation

        We have examined the effects of S-nitroso-N-acetylpenicillamine (SNAP), prostaglandin $E_2$ (PG $E_2$) and dibutric cyclic AMP (dbcAMP) on the methylation of interferon- ${\gamma}$ (IFN- ${\gamma}$ ) gene in human Jurkat T cells. The CpG dinucleotide which is critical for promoter function of IFN- ${\gamma}$ gene was methylated by treatment with SNAP, PG $E_2$ and dbcAMP, respectively. The DNA methylation induced by PG $E_2$ was suppressed by the addition of 2',5'-dideoxyadenosine (DDA), an inhibitor of adenylyl cyclase, but the suppression was not observed in SNAP treated cells. The NO production was not enhanced in PG $E_2$ or dbcAMP treated cells. The methylation induced by PG $E_2$ and dbcAMP was not suppressed by the addition of $N^{G}$-methyl-L-arginine (L-NMMA), NO synthase inhibitor. In conclusion, the inhibition of INF- ${\gamma}$ gene expression by PG $E_2$ was associated with the methylation of INF- ${\gamma}$ gene by elevation of intracellular cAMP in human Jurkat T cells. However, the methylation induced by PG $E_2$ might not be mediated through the NO production.rough the NO production. 본 연구에서 인간의 백혈병세포주인 Jurkat T 세포에서 인터페론 감마(INF-${\gamma}$ 유전자의 methylation에 대한 S-nitroso-N-acetylpenicillamine (SNAP), 프로스타글란딘 $E_2$ (PG $E_2$) 그리고 dibutric cyclic AMP (dbcAMP)의 효과를 조사하였다. 인터페론 감마 유전자의 프로모터기능에 아주 중요한 디뉴클레오티드인 CpG는 SNAP, PG $E_2$, 그리고 dbcAMP를 각각 처리하였을 때 methylation되었다. PG $E_2$에 의해서 유도된 그 methylation은 아데닐산 사이클라제의 저해제의 하나인 2',5'-dideoxyadenosine (DDA)에 의해서 억제되었지만, SNAP에 의해서 유도된 methylation은 DDA에 의해서 억제되지 않았다. PG $E_2$나 dbcAMP를 처리한 세포에서 일산화질소(NO)의 생성의 증가는 나타나지 않았으며, PG $E_2$나 dbcAMP에 의해 유도된 인터페론 감마유전자의 methylation도 일산화질소합 성효소의 저해제인 $N^{G}$ -methyl-L-arginine (L-NMMA)에 의해서 억제되지 않았다. 따라서 인간의 Jurkat T 세포에서 PG $E_2$에 의한 인터페론 감마 유전자의 발현 억제는 세포내의 cAMP생성경로를 통한 인터페론 감마 유전자의 methylation과 연관되어있으나 일산화질소의 생성경로와는 무관한 것으로 보인다.화질소의 생성경로와는 무관한 것으로 보인다.

        As the regulation of total phosphorus (T-P) concentration in biologically treated sewage effluent is reduced to 0.2~2 mg/L, flocculation process is recommended to remove T-P. In this study, the performance of Tisalt coagulant was investigated in terms of dosage and pH in removing phosphorus and the collected sludge after Ti-salt flocculation was calcined to produce titania for effective sludge recycling. The flocculation performance was carried out using two methods: sedimentation and air floatation. Both methods were feasible to apply for Tisalt flocculation. Ti-salt flocculation was effective in reducing phosphorus concentration in sewage effluent, which showed similar performance of alum (Al2(SO4)3). The calcined sludge was recycled to titania which is the widely used metal oxide. Titania produed from Ti-salt sludge indicated similar charateristics of commercially-available P- 25 in regard to photocatalytic activity and surface area. Therefore, this can be easily adopted to titania application by replacing P-25.

        전립선암 노인 환자의 하부요로증상

        Purpose: The purpose of this study was to investigate the degree of LUTS (Lower Urinary Tract Symptoms) and factors affecting LUTS in patients with prostate cancer. Methods: Fifty-four patients with prostate cancer participated and they completed a structured questionnaire at a hospital in Seoul. The LUTS level was measured by using the International prostate symptom score (1992) and the data were analyzed by Wilcoxon rank sum test and Kurskal-Wallis test. Results: Most subjects obtained a higher than moderate LUTS score. The participants who had a lower perceived health status and did not drink alcohol were more likely to had a lower LUTS score than other patients who had higher perceived health status and drink alcohol. In addition, eye diseases were significantly associated with the LUTS score. In LUTS, all the patients received higher IPSS-QOL (International prostate symptom score quality of life) and frequency, nocturia. Conclusion: For proper nursing care related to LUTS, nurses should focus on the factors identified when performing nursing assessments and should develop nursing intervention programs for symptoms management.

        The alkylation of 2-isopropyl naphthalene with isopropyl alcohol was studied over heteropoly acid catalyst supported on mesoporous material. The physicochemical state of loaded heteropoly acid was investigated using XRD, nitrogen adsorption and FT-IR techniques. Heteropoly acid was highly dispersed on the wall of mesoporous material, and retained its Br□nsted acidity. The initial conversion of 2-isopropyl naphthalene was high over the heteropoly acid catalyst supported on mesoporous material, while that was negligible over bulk heteropoly acid. The high selectivity for β,β - diisopropyl naphthalene was explained by the uniform mesopore inducing transition-state shape selectivity.

      • 전침 자극에 의한 오디 괄약근 운동성 변화에 관한 연구

        Background/Aims: This study was designed to evaluated the effects of electroacupuncture on the sphincter of Oddi (SO) motility in humans and to correlate the manometric findings with plasma cholecystokinin (CCK) levels. Methods: Eleven patients (M: F= 5: 6) who had various kinds of biliary disorders were enrolled. SO motility was monitored with a conventional low-compliant continuously perfused technique using ERCP (n=9) or percutaneous transhepatic cholangioscope (n=2). After baseline monitoring for phasic wave contractions of SO, electroacupuncture was applied at a specific acupoint, GB 34, in these 11 patients. A nonspecific acupoint 5 cm away from GB 34 was selected as a control. Manometric parameters of SO were also checked in 6 subjects during stimulation of the control acupoint. CCK plasma levels were measured during the time course of electroacupuncture stimulation. Result: All the manometric parameters including the basal pressure of SO, amplitude, frequency and duration of phasic wave contraction of SO were significantly decreased (p< 0.05) during electroacupuncture stimulation. The inhibition of SO contractility was accompanied by increased CCK plasma levels. After removal of electroacupuncture stimulation, restoration of amplitude and duration to basal condition were noted. A tendency towards the return of SO contractility was also observed in basal pressure and frequency. Stimulation of the control acupoint, however, did not affect the SO contractility and plasma CCK levels. Conclusions: Electroacupuncture stimulation of acupoint GB 34 showed reversible inhibition of SO contraction in humans. We speculate that the response of SO to electroacupuncture stimulation may be mediated by some neurohormonal mechanisms including CCK release.
