        Impaired Spindle Checkpoint Response of Brca1-deficient Mouse Embryonic Fibroblasts (MEFs) to Nocodazole Treatment

        항암유전자 Brca1의 변이는 유방암 및 난소암에 대한 감수성을 증가시키며, Brca1은 DNA손상신호후 세포주기 조절에 필수적인 역할을 한다. 연구결과, Brca1이 세포주기 S기와 G2/M 조절점에서 중요한 역할을 담당함이 밝혀졌다. 그러나, Brca1의 spindle checkpoint 관여여부는 알려져 있지 않다. 본 연구에서는 spindle checkpoint를 활성화시키는 nocodazole를 처리하여 야생형, $p53^{-/-}$ 그리고 $p53^{-/-}\;Brca1^{-/-}$ 세포주의 세포주기 변화를 조사하였다. 야생형과 $p53^{-/-}$ 세포주는 신속한 mitosis기 정지가 나타난 반면, $p53^{-/-}\;Brca1^{-/-}$ 세포주의 경우 모든 세포가 M기에서 정지하지 않았다. Double-thymidine block 기법에 의한 세포주기 동조화후 nocodazole 처리시에도 $p53^{-/-}\;Brca1^{-/-}$ 세포주에서는 일부세포가 M기 조절점을 통과하여 계속 G1기로 진행하였다. 형태학적 분석에서도 nocodazole 함유배지에서 계속 증식하는 세포형태가 관찰되었다. 이와 같은 결과들은 Brca1이 spindle checkpoint가 정상적으로 작동하는데 중요한 역할을 담당한다는 것을 의미하고 있다. Genetic alternation of Brca1 predispose of breast and ovarian cancer. Brca1 plays critical role in cell cycle regulation following DNA damage. Previous studies revealed that Brca1 plays an important role in S phase and G2/M checkpoint regulation. However, whether Brca1 involves in spindle checkpoint is unclear. In this study, the role of Brca1 in cell cycle response following nocodazole, which is a reagent that depolymerizes microtubules and activates the spindle checkpoint, has been examined using wild type $p53^{-/-}\;and\;p53^{-/-}Brca1^{-/-}$ mouse embryonic fibroblasts (MEFs). While wild type and Brca1-proficient MEFs showed an acute mitotic arrest, Brca1-deficient MEFs failed to arrest at mitotic phase in response to nocodazole treatment. In double-thymidine block and nocodazole treatment experiment, a portion of $p53^{-/-}\;Brca1^{-/-}$ MEFs were clearly by-passed nocodazole induced mitotic arrest. Consistent with this, in morphologic analysis, $p53^{-/-}\;Brca1^{-/-}$ MEFs showed growing cell morphology after nocodazole treatment. Taken together, these results suggest that Brca1 protein is an important component for normal induction of spindlecheckpoint and impairment of Brca1 function could induce dysregulation of mitotic cell cycle that ultimately results in genomic instability.

        떫은감 농축액을 첨가한 인절미의 품질 특성

        본 연구에서는 떫은감을 효율적으로 활용하기 위해 떫은감을 가열 농축하여 일반성분과 특성을 분석하였으며 떪은감 농축액을 0%, 4%, 8%, 12%, 16%로 첨가하여 제조한 감인절미를 20℃에서 저장하면서 수분함량, 색도, 기계적 품질특성, 관능적 품질특성을 측정한 결과는 다음과 같다. 23시간을 가열하여 제조한 떫은감 농축액은 수분함량 36.57%, 조단백질 0.71%, 조섬유 2.39%, 조회분 2.14%, 비타민 C 1.09 mg%, 총페놀함량 57 mg%였고, 감농축액은 pH 4.71, 당도 63 Brix, 유리당은 fructose, glucose, sucrose, maltose가 각각 16.23, 31.07, 0.51, 0.50%였으며 색도는 L, a, b 값이 각각 21.50, 1.59, 0.98였다. 감농축액 0, 4, 8, 12, 16% 첨가군의 수분함량은 57~59% 범위였다. L값은 감농축액 첨가량이 증가할수록 유의적으로 감소하였고 a값은 감농축액 8% 첨가군이 가장 높았고 b값은 감농축액 첨가량이 증가할수록 감소하였다. 경도는 감농축액 0% 첨가군의 경우 저장 2일째부터 부패가 진행되면서 감소하였으나 감농축액 첨가군은 적은폭으로 증가하였다. 부착성은 제조직후와 저장기간동안 감농축액 0% 첨가군이 가장 낮았다. 응집성은 저장기간 동안 감농축액 16% 첨가군에서 가장 낮았으며, 탄력성은 저장, 2일째까지 감농축액 첨가량에 따른 시료간에 유의적인 차이가 없었다. 점착성은 저장 1일째 이후의 저장 기간에서는 시료간에 유의적인 차이가 없었다. 전반적으로 저장기간에 따른 기계적 품질특성의 변화가 적었다. 관능평가는 감농축액 첨가량이 증가할수록 색, 향, 단맛, 쓴맛, 떫은맛, 촉촉한 정도, 씹힘성을 강하게 평가하였다. 부드러운 정도는 8% 첨가군에서 가장 강하게 평가되었다. 전반적인 기호도는 8%>12%>4%>16%>0% 순으로 좋게 평가되었다. 결론적으로 감농축액의 첨가는 당과 탄닌성분을 함유함으로써 인절미의 노화를 지연시키고 저장성을 높이는데 효과가 있는 것으로 생각된다. 또한 인절미를 제조할 때 찹쌀가루에 8%의 감농축액을 첨가하는 것이 감인절미의 색, 향, 단맛, 씹힘성이 적당하고 쓴맛과 떫은맛은 약하게 느끼면서 촉촉한 정도와 부드러운 정도는 강하게 느낄수 있어서 전반적인 기호도에서 가장 적절한 방법으로 사료된다. The purpose of this study was to find the optimum addition amount of astringent persimmon paste to glutinous rice flour in the preparation of Gaminjeolmi (persimmon glutinous rice cake). The moisture contents of Gaminjeolmi with added astringent persimmon paste were 57~59%. With increasing addition of astringent persimmon paste, the L- and b-value were decreased. The a-value was the highest at the 8% level. In the mechanical evaluation of Gaminjeolmi, the hardness was the highest in the 0% astringent persimmon paste-Injeolmi but the 0% level was decomposed after 2 day of storage. The adhesiveness and cohesiveness were the lowest at the 0% and 16% levels. The springiness did not differ significantly with the addition of astringent persimmon paste for 2 days of storage. The gumminess was the highest at the 0% level for 1 day of storage, significantly. In the sensory evaluation of Gaminjeolmi, the acceptance of the softness characteristics was the best at the 8% level. Addition of astringent persimmon paste improve preservation of Injeolmi and the addition of 8% of astringent persimmon paste to glutinous rice flour was found to be the best recipe in terms of the sensory qualities of softness and overall acceptability.

        베트남인의 한국 의료관광 이용의도에 영향을 미치는 요인

        Purpose: The purpose of this study was to identify factors related to Vietnamese customers who use Korean medical and tourism services. The study was based on the Anderson Models Methods: Participants were 173 Vietnamese who were living in Ho Chi Minh and Hanoi in Vietnam. Data were analyzed using descriptive statistics, x², t-test and Multiple Logistic Regression. Results: Research necessary factors had the most significant effect among the preceding factors, possible factors and necessary factors. Human service and outstanding natural beauty of necessary factors had significant influence on customer intention to use. According to the result of the stepwise logistic regression analysis, the most important item in medical services was human service OR=1.89 (p=.014), and for tour services, outstanding natural beauty OR=4.30 (p=.033). The explained variance for customer intention to use was 91.9%. Conclusion: These results suggest the need to improve the human service and outstanding natural beauty to retain customers" intention to use.

        간호사의 이미지와 호감도 분석

        Purpose: The purpose of this study was to investigate nurse images and likability that were perceived by various groups in the general population. Methods: Using cross-sectional descriptive design, six groups of people were invited to participate in the survey. The participants included nurses, physicians, nurse aids/technicians, patients/caregivers, college students, and the general public. A total of 1,530 individuals were drawn from 42 hospitals nationwide. Bhargava`s likability questionnaires and the Ward Nurse Image tool were used. Results: Compared with other groups, patients and caregivers had higher scores for likability and better nurse image than other groups. Most respondents perceived nurses are clean and caring, but many individuals still had the image that nurses are submissive. Conclusion: To improve nurse image, perceptions from various groups of people need to be considered and programs develop to show the positive aspects of nursing and nurses.

        율피 추출물에 의한 Tyrosinase 활성억제 및 Melanin 생성억제 효과

        The inhibitory effects of chestnut bark extracts (CIS, CIS-gel, CIS jelly) were investigated in vitro and in B-16 mouse melanoma cells on melanin biosynthesis which is closely related to hyperpigmentation. These extracts inhibited tyrosinase activity which converts dopa to dopachrome in the biosynthetic process and the extracts inhibited the production of melanin from dopachrome by auto-oxidation. B-16 cells treated by chestnut bark extracts showed that the viability was over 85% in MTT assay. Melanin production was inhibited significantly by chestnut bark extracts in vitro and in B-16 cells. Especially CIS showed 15 times stronger inhibition of melanin production than kojic aicd in vitro. CIS-gel and CIS showed much stronger inhibition of melanin production under concentration of 4 ㎍/㎖ than kojic acid in B-16 cells. These results suggest that the chestnut bark extracts can be used for a whitening agent of the skin.
