Aspergillus oryzae A-5로부터 Low Density Lipoprotein(LDL)의 산화에 대한 항산화 효과

Antioxidative activity of fraction extracted from cultivation of Aspergillus sp. A-5 against oxidation of human low density lipoprotein(LDL) was investigated. Fractions of Aspergillus sp. A-5 cultivation was sucessively purified with ethyl acetate and silica gel column chromatography. The concentration of fraction 4 inhibited Cu$^{2+}$-induced oxidation of LDL almost completely. Band 3 isolated by the further purification of fraction 4 was higher than that of same concentration of $\alpha$-tocopherol, BHA and BHT. The elcetrophoretic mobility of oxidized LDL by addition of Band 3 was faster than that of native LDL, but slower than that of oxidzed LDL. It is concluded that fraction of Aspergillus cultivation contained antioxidants with the capacity to inhibit oxidative modification of LDL.
회전원판 반응조를 이용한 Pseudomonas sp. KS-96에 의한 gallic acid로부터 Pyrogallol의 전환

Gallic acid로부터 pyrogallol로 전환하는 균주를 토양에서 분리 동정하여 얻는 Pseudomonas sp. KS-96의 균체를 회전원판 반응조에 고정화시킨 후 연속전환을 시도하여 다음과 같은 결과를 얻었다. Gallate decarboxylase의 활성은 배양 24시간에서 가장 좋았다. 반응조의 원판에 gallic acid가 함유된 액체 배양액을 시간당 20m${\ell}$씩 공급하면서 균체막의 두께가 steady state에 도달할때까지 공급하였다. 균체를 고정화 시킨 후 시간 경과에 따른 전환율은 18시간부터 36시간에서 전환율이 높았다. 균체 고정화 후에 gallic acid를 15g/${\ell}$ 함유하는 기질을 시간당 30m${\ell}$씩 주입했을 때 pyrogallol의 전환율이 가장 좋았으며 회전원판 반응조에 의한 연속적 전환은 6일에서 8일까지가 각각 81%, 80%로서 가장 높았다. In previous paper Pseudomonas sp. Ks-96 isolated from the bioconversion into pyrogallol from gallic acid . Continuous bioconversion of pyrogallol was carried out using rotatory disc contactor immobilized Pseudomonas sp. Ks-96 . Enzyme activity of gallate decarboxylase released from Pseudomonas sp. Ks-96 were shown at the highest activity on 24h incubation. Culture media containing gallic acid supplied on the flow rate of 20m${\ell}$/h until thickness of cells wall reached steady state. Bioconversion rate of pyrogallol from gallic acid showed at highest level ranging from 18hr to 36h according to time courses. Continuous bioconversion of pyrogallol using rotating disc contactor was about 82% and 80% between 6 and 8 days at the feeding rate of 300m${\ell}$ per hour in the medium containing 15g/${\ell}$ gallic acid.
반응시간지수(Response Time Index)를 이용한 국내 화재감지기 등급분류에 관한 연구

This paper presents classification of domestic fire detector using response time index. Response time is measured using fire detector distributed in Korea, and the response time index is estimated. Plunge test prescribed by FM is conducted to measure response time of fire detector. The detector used to test is fixed temperature type(thermistor and bimetal type) and rate of rise temperature type(thermistor and pneumatic type). The nominal operation temperature of fixed temperature type detector is 70℃ and rate of rise temperature is 15℃/min. The fixed temperature type is measured 7 products, and the rate of rise temperature type is measured 5 products. The results show that in case of fixed temperature type(thermistor) is classified “Quick” or “Standard” and fixed temperature type(bimetal) is not classified. The rate of rise temperature type(thermistor) is classified “Fast” or “Ultra Fast” and the rate of rise temperature type(pneumatic) is classified “Very Fast” or “Ultra Fast”. The pneumatic type shows more fast response than thermistor type. Also these results indicate the fixed temperature type(bimetal) is not suitable for early stage fire detection.

말똥 성게를 인공적으로 수정시킨 후, 칼럼 크로마토그래피법으로 분리 정제하여 리보뉴크레아제 $H_1$과 리보뉴크레아제 $H_2$를 얻었다. 리보뉴크레아제 $H_1$의 분자량은 약 68,000이었고, 리보뉴크레아제 $H_2$의 분자량은 약 44,000 이었다. 리보뉴크레아제 $H_1$는 약알카리성에서, 리보뉴크레아제 $H_2$는 알카리성에서 활성이 높았다. 이들 효소는 모두 2가이온인 마그네슘이온 ($Mg^{2+}$), 망간이온($Mn^{2+}$)과 그리고 염의 일정농도에서 활성을 나타내고 있으며, N-에칠 말례이마이드에 감수성이 높았다. 그리고, 리보뉴크레아제 $H_1$과 $H_2$는 단일가닥과 이중가닥의 활성은 없었으나, RNA-DNA hydrid는 급히 저하 시켰다. From the sea-urchin, Hemicentrotus pulcherrimus, we have purified by two column chromatographic steps two ribonuclease H (RNase H) activities of RNase $H_1$ and RNase $H_2$ in homogeneous form. RNase $H_1$ was molecular weight around 68,000 and RNase $H_2$ was around 44,000 by Sephadex G-75 gel filteration and SDS-polyacrylamid gel electrophoresis. RNase $H_1$ was optimal active as well as slightly alkaline pH and RNase $H_2$ was as alkaline pH. These enzymes shown to be a function of the divalent metal ion $Mg^{2+}$, $Mn^{2+}$ and salt employed as activator, also were sensitive -SH reagent N-ethylmaleimide. RNase $H_1$ and RNase $H_2$ were shown no activity with single or double stranded RNA but rapidly hydrolysis the RNA-DNA hybrid.
랙크식 물류창고 조기 화재감지를 위한 최적 화재감지기 설치방법에 관한 실험연구

This paper is an experimental study to find an optimal detection method for detecting fire early in a rack-type warehouse stored with goods. In this study, we constructed rack-type structure with the fourth floor of 13.5 m high and conducted fire experiments which were to measure flow of heat/smoke in rack-type structure and response time of fire detectors. The detectors used at experiments were fixed temperature type detectors, rate of rise detectors, photoelectric smoke detectors, air sampling smoke detectors and flame detectors. The used ignition sources are n-heptane fire for response of heat detection and cotton fire for response of smoke detection. The fixed temperature type detectors, rate of rise detectors and photoelectric detectors were installed to every rack level respectively. The results show that the rate of rise detector should be installed every 2 levels and photoelectric smoke detector should be installed every 4 levels for the early stage fire detection. Air sampling smoke detectors can detect fire early in response to control of sensitivity, but there is a problem in false alarm. The fixed temperature detector is not suitable for early stage fire detection in warehouse and flame detector not worked if flame is not visible, so it need to install combination with other detector.
