DNA Comet Assay를 이용한 콩류의 방사선 조사 확인

콩류의 방사선 조사 여부를 DNA comet assay로 확인 하였다. 대두, 강남콩, 그리고 팥을 각각 0.1, 0.3, 0.5, 0.7, 그리고 1.0 kGy의 저선량으로 조사하고 비조사 시료와 조사시료간의 DNA 손상정도를 현미경으로 육안 검사 및 comet의 tail 길이를 측정하여 상호 비교하였다. 모든 시료에서 비조사 시료보다 조사시료의 tail 길이가 더 길었으며 조사 선량이 증가할수록 tail 길이가 길게 나타났다. 비조사 시료에서도 손상된 세포의 comet 모양의 핵이 관찰되기는 하였으나 대부분 비손상된 세포의 원형모양의 핵이 관찰되었으며 조사된 시료에서는 comet 모양의 핵이 주로 관찰되는 경향이어서 비조사 시료와 조사시료간에 comet 양상을 비교할 수 있었다. 한편 100개의 세포를 무작위로 선택하여 comet tail 길이를 측정하고 통계분한 결과, 비조사 시료와 조사시료간의 유의적인 차이가 있었고 조사선량이 증가할수록 tail 길이가 유의적으로 증가함을 확인하였다. DNA comet assay로 간단하고 비교적 저렴한 비용으로 짧은 시간 내에 콩류의 방사선 조사 여부를 확인할 수 있었다. The single cell-gel electrophoresis assay (comet assay) was used to identify irradiated beans. Soy beans, kidney beans, and red beans were irradiated with (60)^Co gamma rays at 0.1, 0.3, 0.5, 0.7, and 1.0 kGy. Beans were peeled out, crushed lightly, and treated with phosphate-buffered saline (PBS) to extract cells. The extracted cell suspension was mixed with agarose gel solution and spread on an agarose precoated slide. After lysis of the cells, they were subjected to microgel electrophoresis for 2 minutes, and then silver-stained. We found that the DNA fragments of the irradiated samples were stretched, migrated out of the cells, and formed tails towards the anode giving the appearance of comets, while the unirradiated or the undamaged cells formed very short or no tails. The tail lengths of irradiated samples were significantly increased as irradiation dose increased at the above 0.3 kGy.

Objectives: This Study, designed as a retrospective cohort study, aims to study the epidemiology and characteristics of traumatic spinal cord injury (SCI) in Korean military. Methods; A cohort of 64 male veterans, with traumatic SCI which occurred between May, 2012 to April, 2016 who visited the armed forces military hospital, were assembled from the Defense Medical Statistical Information System. The records were verified by the New Defense Medical Information System and reviewed by a physical medicine & rehabilitation doctor. Results: Patients consisted of officers (11 persons), sergeants (21 persons), soldiers (28 persons), others (1 person) and unknown (3 persons) rank and were injured in their mid-twenties. The main causes or injury were military training (19 persons, 30%), sports including diving injury (12 persons, 19%), traffic accident (11 persons, 17%). There was a higher incidence or injury during sports in officers and training in sergeants. However, suicide injury was identified only in soldiers. The main causes of injury was carelessness in old age and training in younger age. The severity of injury was 10 persons (21.7%) with American spinal injury impairment scale A, 4 persons (6.3%) with B, 8 persons (12.5%) with C and 38 persons (59.4%) with D. Thirty one patients (48.5%) had continued military service and 26 patients (40.6%) had discharged after injury due to disability, and 48 patients (65.6%) among them had confirmed as official injury. Only 5 patients or z8 patients who received the phone call,were selected as veterans and were being treated at the Veterans hospital. Conclusions: This is the first study to investigate the epidemiology and characteristics of traumatic spinal cord injury in Korean military. More follow-up studies on the psychologic problems, quality of life and occupation are expected to be published through the cooperation with die Veterans hospitals.

우리는 보통 ‘성경(誠敬)의 도’라고 통용해서 사용하고 있지만, 성(誠)과 경(敬) 개념은 각기 자체만으로 수양공부론적 의미를 함의한다. 성경을 중심으로 수양공부를 제시한 인물은 정호(程顥)인데 정이(程頤)와 구별되는 자신만의 사상을 제시한다. 그에게 있어서 ‘성(誠)’은 믿을만한 행위로 이루어진 삶을 의미하며 이상적인 경계로까지 그 의미가 확대된다. ‘경(敬)’은 실제 삶에서의 행동지침으로 구체적인 수양방법이면서 성(誠)과 동일시된다. 성경(誠敬)은 본체공부로서의 특색을 드러내면서도 정호 사상체계를 전반적으로 파악할 수 있는 핵심 개념이다. 본 논문은 그가 제출한 성경(誠敬)의 의미를 성(誠)과 경(敬)을 합치한 성경(誠敬)의 도에 대한 통합적 고찰을 통해 정호가 제시하는 공부론의 특색을 드러내는 방식으로 전개한다. 이는 정호 사상 전반을 파악하는 유의미한 작업이 될 것이다. Sincerity and Piety are usually combined together to define justice as the word Justice of Sincerity and Piety. However, the concepts of Sincerity and Piety imply the meaning of self-discipline study theory respectively. It was Cheng Hao who suggested the study of self-discipline focused on Sincerity and Piety, which was distinguished from Cheong Yi’s theory. He thoughts that Sincerity meant the life that consisted of reliable behaviors, which would extend its meaning up to the ideal level. Piety is not only a clear self-discipline method that plays a behavior standard to everyday life but also considered as same as Sincerity. The concept of Sincerity and Piety is essential in grasping Cheng Hao’s idea system while exposing its attribute as a study of the existence root. This study is developed in the way to expose the attribute of the study theory suggested by Cheng Hao through a convergent consideration on the Justice of Sincerity and Piety in which they are integrated into Justice in order to explain their meaning shown by him, which will be a meaningful work to understand Cheng Hao’s idea in overall.
