        攝食 및 絶食狀態가 放射性醋酸의 脂肪組織에의 編入에 미치는 影響

        放射性醋酸의 脂肪組織의 脂質에의 編入에 대하여 攝食 또는 絶食狀態에 따라 어떻게 變動하는가를 調査하여 다음과 같은 結果를 얻었다. 放射性醋酸의 脂質에의 編入樣相은 腸間膜脂肪組職에서나 副睾丸脂肪造直에서 同一하였다. 脂肪組織의 脂質에의 放射性醋散의 編入速度는 攝食 또는 絶食狀態에 相關없이 注入 후 3時間째에 最高値에 達하였다. 絶食狀態에서의 放射性醋酸의 脂肪組織에의 編入速度는 正常 攝食狀態에서의 그것에 比하여 約 4分의 1에 해당되는 적은 量이었다. 以上의 結果는 脂質合成 速度는 脂肪組織의 所在에 關係없이 同一하며 饑餓狀態에서는 脂肪合成이 顯著하게 減少됨을 示唆한다고 하겠다. To examine the effect of starvation on the incorporation of acetate-1-???C into adipose tissue, male rats weighing approximately 250 gm were divided into two groups; the control group was fed commercial diet ad libitum and the starved group was fasted for 24 hours before the adminis-tration of acetate-1???C. A single dose of 4 μCi of acetate-1-???C per 100 gm body weight was injected abdominally in both fed and fasted rats. To determine the rate of incorporation of acetate-1-???C into lipid, rats were killed at 1.5, 3.0 and 4.5 hours after the injection, and the radioactivity of labeled lipid was counted with gas flow counter. The incorporation of acetate-1-???C into lipid was the same rate in both mesenteric and the epididy-mal adipose tissue. The rate of acetate-1-???C incorporation into adipose tissue was reached to the peak at three hours after the injection of the acetate regardless of feeding or fasting state. The peak of the acetate incorporation into adipose tissue of starved rate was only one fourth compared with that of normal fed group. The above reaults indicate that lipogenesis in the adipose tissue is remarkablly inhibited by the starvation.

      • 肝吸蟲症患者에 있어서 血淸 Ceruloplasmin과 L-Phenylalanine Inhibited Alkaline Phosphatase 値에 關하여

        肝吸蟲症患者에서 肝의 細胞損傷, 蛋白合成低下 및 膽汁排泄障碍를 檢査하는 한편 血淸 ceruloplasmin과 血淸 total 및 L-phenylalanine inhibited alkaline phosphatase(SPIAP)를 測定하여 血淸 ceruloplasmin 과 이들 phosphatase와의 相互關係를 檢討하여 다음과 같은 結果를 얻었다. 1. 健康韓國人 男子 43名의 血淸 ceruloplasmin値는 平均 29.1±1.0(Mean±SE)mg% 였으며 이에 比해 肝 吸蟲症 28例에서는 平均 63.7±4.1(Mean±SE)mg%로 顯著한 增加를 보임과 同時에 total bilirubin, SAP 및 SPIAP도 有意한 增加를 보였다. 2. 肝吸蟲症患者에서 血淸 ceruloplasmin과 各血液化學檢査値와의 相關關係를 보면 다음과 같다. 血淸 ceruloplasmin과 血淸 total bilirubin과의 相關關係는 +0.695, SAP와는 +0.801,SPIAP와는 +0.842로 有 意義 (P<0.001)한 相關關係를 보였으나 SGPT와 血淸總蛋白 및 各蛋白分劃間에는 意義있는 相關關係를 볼 수 없었다. 血淸 ceruloplasmin의 增加가 肝外에서 生成되며 膽道를 通해 排泄되는 SPIAP의 增加와 相關關係가 높음 을 보아 肝吸蟲症患者에서의 血淸 ceruloplasmin의 增加는 膽汁排泄 障碍가 主要因이 될 것으로 생각된 다. The behaviour of serum ceruloplasmin was investigated in clonorchiasis. The serum concentration of ceruloplasmin in 28 cases of clonorchiasis patients and 43 normal subjects were estimated. At the same time, the values of several liver function tests and L-phenylalanine inhibited alkaline phosphatase (SPIAP) were examined of the same patients. In normal subjects, the mean value for serum ceruloplasmin was 29.1±1.0(Mean+SE)㎎/100㎖. The serum ceruloplasmin concenrtation in clonorchiasis patient was 63.7±4.1㎎/100㎖. Increased levels of ceruloplasmin were found in 20 clonorchiasis patients. Serum ceruloplasmin showed a significant correlation with total bilirubin (r=+0.801, P<0.001), serum total alkaline phosphatase (r=+0.801, P<0.001) and SPIAP (r=+0.842, P<0.001). No significant correlation was observed with albumin, alpha 2-globulin, gamma-globulin and A/G ratio.

      • 총수담관을 결찰한 흰쥐의 혈청 및 간장의 Leucine Aminopeptidase에 미치는 Actinomycin D의 영향

        간담도질환시 중가되는 혈청 leucine aminopepItidase외 증가기전을 알아보기 위하여 흰쥐의 총수 담관을 결찰하고 경시적으로 혈청파 간장의 leuline aminopeptidase의 활성을 측정하는 한편 간장 및 혈청의 alkaline phosphatase, 단백합성능과 감세포의 RNA합성능등을 관찰하였으며 아울러 이들에 대한 actinomycin D의 영향도 관찰하여 상호 비교하였다. 혈청 leneine aminopeptidase는 총수담관결찰후 6시간부터 그활성이 증가되며 이후 계속 증가되어 3일후에는 가수술군에 비해 약 2.5배나 증가되었다. 그리고 간장의 leucine aminopeptidase외 활성치는 혈청의 치보다 약 17배나 현저히 높은치를 나타내었으며, 역시 담관결찰후 증가되었으나 그 증가율은 혈청보다는 다소 낮은 경향이었다. 한편 이효소활성의 증가는 actinomycin D를 수술전후 투여함으로써 혈청이나 간장이 다같이 저해되어 정상치의 수준으로 저하되었다. 총수담관결찰시 간장 및 혈청의 alkaline phosphatase는 leucine aminopeptidase와 마찬가지로 담관결찰후 현저히 증가되었는데 그 증가율은 leucine aminopeptidase보다 더욱 현저하였다. 그리고 이효소도 역시 actinomycin D투여로써 그 활성이 현저히 억제되었다. 총수담관결찰후 1일째의 amino acid mixture(U)^14C의 혈청 및 간단백에의 편입속도와 orotic acid-(6)^14C의 간장의 총 및 핵RNA에의 편입속도는 모두 높은증가를 보였는데 그증가의 정도는 단백보다는 RNA가 훨씬 현저하였다. 그리고 이들 또한 actinomycin D의 투여로써 그편입율이 현저히 억제되었는데 그억제의 정도는 단백보다는 RNA쪽이 더현저하였고 또 RNA에서는 핵RNA쪽이 더현저하였다. 이상의 성적으로 보아 담즙울체로 간장손상이 야기되면 간장 및 혈청의 leucine aminopeptidase는 간장에서 유도가되어 증가될 것으로 생각되며 이증가는 효소단백생합성의 항진에 의할것이라 사료된다. This study was intended to observe the changes in serum and hepatic leucine aminopeptidase activities after the ligation of common bile duct in rats. Activities of serum and hepatic alkaline phosphatase, incorporation rates of amino acid mixture-(U)-^14C into liver, serum protein and orotic acid-(6)-^14C into liver RNA are compared with the changes observed. Then the effects of actinomycin D adminstered rats. treated in the same method, were also observed for comparison. Common bile duct ligation in the rats brought about a rapid increase in hepatic leucine aminopeptidase activity and more rapid of serum leucine aminopeptidase. Serum leucine aminopeptidase showed an increase of 1.5, 1.9 and 2.5 fold at the 1st, 2nd and 3rd day respectively following the bile duct ligation. But the increase was observed markedly inhibited in the case of adminstration of actinomycin D (50㎍/l00g of body weight) one hour prior to and eighteen hours alter the ligation of the common bile duct in rats. After the ligation of common bile duct, the levels of hepatic and serum alkaline phosphatase increased rapidly, hepatic alkaline phosphatase more so. These levels continued to be high throughout the experiments. The increase of hepatic and serum alkaline phosphatase was significantly diminished with additional actinomycin D treatment. Incorporation rate of orotic acid-(6)-^14C into both hepatic total and nuclear RNA showed a remarkable increase after the common bile duct ligation and reverse effect when actinomycin D treatment was practiced. Incorporation rate of amino acid mixture-(U)-^14C into hepatic and serum protein showed a similar increase. Actinomycine D inhibited incorporation of amino acid mixture(U)-^14C into hepatic and serum protein showed a far less degree than orotic acid-(6)-^14C incorporation into hepaticRNA.

        총담관을 결찰한 흰쥐 혈청 및 Isocitrate Dehydrogenase간의 활성도와 총담즙산의 농도

        A study was made on the changes of NADP+- and NAD+ dependent isocitrate dehydrogenase (NADP+-ICD, NAD+-ICD) activities in cholestatic rat liver. The cytosolic and mitochondrial NADP+-ICD and mitochondrial NAD+-ICD activities were determined in the cholestatic rat liver induced by common bile duct ligation over a period of forty two days. And the activities of NADP+- and NAD+-ICD in the serum were measured. The concentrations of total bile acids in their serum and liver cytosolic fraction were also measured. The cytosolic and mitochondrial NADP+-ICD activities of the cholestatic rat liver each showed a marked decrease between the 1st and the 42nd day and the 12th hour and the 42nd day respectively after the ligation. However, the mitochondrial NAD+-ICD activity in the cholestatic liver markedly increased between the 2nd and the 42nd day after the ligation. On the other hand, the NADP+-ICD activity of the serum markedly increased between the 12th hour and 3rd day after the ligation. That of the serum NAD+-ICD also showed a marked increase between the 1st and the 7th day after the ligation. The concentrations of total bile acids in the cholestatic liver cytosolic fraction showed a marked increase between the 12th hour and the 7th day after the ligation. That of total bile acids in the serum showed a marked increase throughout the experiments. Viewed from above results, the NADP+-ICD in the liver with cholestasis seems to be the decreasing enzyme. But the NAD+-ICD in the cholestatic liver thought to be the increasing enzyme in its activity. Especially, when the cholestasis occurred, the activities of these enzymes of serum are higher than those of the controls because of increased permeability secondary to liver cell necrosis, which causes the enzymes to leak into the blood in great quantity.

        Effect of Acute Ethanol Intoxication on Hepatic Rhodanese Activity in Rats with Extrahepatic Cholestasis

        Liver and serum rhodanese activities were determined in acute ethanol intoxicated rats with extrahepatic cholestasis induced by common bile duct ligation (CBD) to manifest the biochemical background of alcohol drinking hazard under the hepatobiliary disease. Liver cytosolic and microsomal rhodanese activities and these Vmax values in CBD ligated rats with acute ethanol intoxication were found to be decreased much more than that in CBD ligation alone. However, the difference of Km value on above hepatic enzyme was not found between the experimental groups. On the other hand, serum rhodanese activity in CBD ligated rats with acute ethanol intoxication was greater increased more than that in CBD ligation alone. These results indicate that the biosynthesis of the hepatic rhodanese decreases and the serum rhodanese activity increases in cholestasis combined with acute ethanol intoxication. reflecting damage of aggravated hapatocytic membrane. Accordingly, the resulting data supported the fact that alcoholic drinks were enzymologically harmful to the hepatobiliary disease.

        Catechol-O-Methyltransferase Activity from Regenerating Liver after Partial Hepatectomy in Rats

        The change of catechol-O-methyltransferase (COMT) activity during regeneration of rat liver was studied. Cytosolic, mitochondrial and microsomal COMTs activities were estimated in regenerating rat livers over a period of ten days after 70% (median and left lateral lobes) partial hepatectomy. The values of Km and Vmax in the hepatic enzymes were also measured. The activities of cytosolic and microsomal COMTs in regenerating rat liver after partial hepatectomy were found to be significantly increased between the second and the third day. Whereas the mitochondrial COMT activity did not change. The Vmax values of the cytosolic and microsomal COMTs in the regenerating rat liver were significantly increased at the second day after partial hepatectomy, however, the Km values of the above hepatic enzymes did not vary in all the experimental groups. Therefore, the results suggest that the biosynthesis of COMT was increased during the regeneration of rat liver.

      • 간엽을 부분 절제한 흰쥐의 혈청 및 재생간장의 Leucine Aminopeptidase의 활성도

        간장을 부분절제했을때의 재생간장 및 혈청의 leucine aminopeptodase의 변동을 알아보기 위하여 건강한 흰쥐를 사용하여 간장을 약 70% 부분절제하고 10일동안 경시적으로 재생간장 및 혈청중의 leucine aminopeptodase의 활성도를 측정하여 상호 비교 하였다. 재생간의 leucine aminopeptodase의 활성도는 간엽절제후 1일과 2일째의 재생간장에서만 다소증가된치(약1.2배, P<0.05)를 나타내었다. 그러나 혈청에서의 변동은 재생간장에서의 변동과는 다르게 12시간부터 증가되어 2일에는 약1.7배(P<0.01)로 증가되었다가 이후 감소되어 6일째부터 정상으로 회복되었다. The effect of partial hepatectomy (removal of 70% of the liver in rat) on the changes of leucine aminopeptodase(E.C. 3.4.1.1) in regenerating liver and serum was performed by comparing the activities of leucine aminopeptidase in regenerating liver and serum respectively for 10 days. After partial hepatectomy, the activity of leucine aminopeptidase of regenerating liver increased slightly about 1.2-fold (P<0.05) just on 1st and 2nd day, but that of serum stated to increase more radily from 12 hrs and up to 1.7-fold(P<0.01) times on 2nd day then declining gradually to normal on 6th day.

      • 實驗犬에 있어서 出血性Shock時의 血淸 및 胸管淋巴의 Ceruloplasmin 活性度의 變化에 關하여

        The pupose of this study was to investigate the changes of the activity of ceruloplasmin, transaminase, alkaline phosphatase and alpha 2-globulin concentration of serum and thoracic duct lymph in dogs induced hemorrhagic shock maintained blood pressure at level of 60-70mmHg. The results obtained were summerized as follows. 1. The activities of serum and thoracic duct lymph ceruloplasmin were significantly increased at 1 hour pothemorrhage and gradually decreased to prehemorrhagic levels after 3hours. Reinfusion of the blood resulted in an increase of the activities. 2. No significant change were observed in activities of alkaline phosphatase and α_2-globulin concentration after hemorrage or reinfusion of blood. 3. The activities of serum and thoracic duct lymph transaminase showed no significant changes during the course of hemorragic shock but the activities were markedly increased after reinfusion of blood.

        흰쥐 재생간의 5´-Nucleotidase 및 Gamma-Glutamyl Transpeptidase의 활성치

        A study was made on the changes in activities of the followings during 6 post-operative days : Plasma membrane, microsomal, ,nuclear and cytosolic 5'nucleotidase and gamma-glutamy1 transpeptidase of regenerating rat livers, ant rat serum 5'nucleotidase and gamma-glutamy1 transpeptidase after 70% (median and left lateral lobes) partial hepatectomy. The activities of alanine aminotransferase in both serum and cytosol of regenerating rat liver were also measured. The activities of 5'-nucleotidase in the normal rats hepatic subcellular fractions were " 59.29±11.35(n mol Pi/mg protein/min)in the plasma membrane ; 52.86±3.88 in the microsome ; 10.56±2.12 in the mitochondria ;11.48±3.62 in the nuclei, and 3.55±0.54 in the cytosol. Those of gamma-glutamy1 transpeptidase were: 1.96±0.46 (n mol p-nitroaniline/mg protein/min) in the plasma membrane ; 3.38±0.62 in the microsome ; 0.88±0.12 in the mitochondria ; 1.40±0.13 in the nuclei, and 0.76±0.12 in the cytosol. Normal activity of alanine aminotransferase was 330±56 Karmen unit/mg protien in the rat hepatic cytosol. After partial hepatectomy in the rats, activities of plasma membrane bound gamma-glutamy1 transpeptidase in regenerating livers significantly increased in the span between the second and the third days but plasma membrane bound 5'-nucleotidase showed no change. The activity of microsomal 5'- nucleotiadase in the regenerating liver tremendously from the second to the sixth days after partial hepatectomy. And the activity of hepatic microsomal gamma-glutamy1 transpeptidase also elevated from the second and the third days after partial hepatectomy. Tomy activity of mitochondrial 5'-nucleotidase in the regenerating liver decreased between the third and the sixth days after partial hepatectomy. And the activity of hepatic microsomal gamma-glutamy1 trsnspeptidase also elevated from the second and the third days after partial hepatectomy. The activity of mitochondrial 5'-nucreotidase in the regenerating liver decreased between the third and the sixth days of partial hepatectomy. The hepatic nuclear and cytosolic 5'-nucleotidase decreased in the regenerating stage. The decreased values were seem after two days in cytosolic 5'-nucleotidase and after three days in nuclear 5'-nucleotidase following the operations. However, no significant changes in hepatic mitochondrial, nuclear and cytosolic gramma-glutamy1 were ntoed through out the experiments. The activity of serum alanine aminotransferase markedly elevated from 12 hours to three days after partial hepatectomy, but serum 5'-nucleotidase and gamma-glutamy1 transpeptidase activities remained normal.
