        Zirconium Pyrithione 착물의 항균력

        Zr, Cu, Zn and Fe-pyrithione complexes were prepared in aqueous medium and their antimicrobial activities were evaluated by MIC and cylinder plate methods against Bacillus subtilis, Sarcina lutea, Staphylococcus aureus, Pseudomonas aeruginosa, Klebsiella pneumoniae, Trichophyton mentagrophytes, Candida albicans, Asperillus niger and Saccharomyces cerevisiae. Zr-pyrithione possessed most potent activities among the metal complexes against a wide range of microorganisms, especially Gram positive, B. subtilis, S. aureus and fungus, C. albicans. And all of the metal complexes synthesized were more active than pyrithione base. Fe(III)-pyrithione complex has an equal potency to Zn-pyrithione in general but it showed potent activity against B. subtilis microorganism than Zn-pyrithione.

        Zirconinm pyrithione complex was prepared by reaction of sodium-pyrithione solution and zirconyl chloride solution. The physico-chemical properties of the complex was examined by means of 1R, XRD DSC and NMR. And the stability of Zr-complex was investigated on the basis of accelerated stability analysis under conditions of temp, elevation, UV radiation and pH dependence. The result indicates that the ratio of the ligand to metal in Zr-pyrithione complex was determined 4:1, and its stability constant was 4.643×10 exp(4). The rate order of decomposition of the complex was apparent first-order reaction of which rate constant and the decomposition rate was not only accelerated by effect of heat and UV radiation but was catalyzed by specific acid-base catalysis considered the pH dependence for the hydrolysis of the complex and the suspension was most stable over the range pH 4-8 indicating that solvent catalysis is the primary made of reaction in this region.

      • Zr-pyrithione과 cellulose의 결합

        Zr-pyrithion complex was prepared by reaction of sod-pyrithione soln and zirconyl chloride soln and the acute toxicity, eye irritation and a primary skin irritation tests were performed. The binding of Zr-pyrithione to the cellulose was studied. The acute oral LD_50 of Zr-pyrithion complex in the mouse was found to be 5,000㎎/㎏. An eye irritation in rabbits with a 0.1㎖ of 10%-suspension of Zr-pyrithione was applied to the rabbit cornea and conjunctives and did not produce any signs of irritation. Zr-pyrithione was made on the shaved skin of raddits with a 0.5g single applications. No signs of erythema were found in any of the test areas. Cotton fabric, cellulose, by a two-bath process with zirconyl acetate and sod-pyrithion retained excellent bacteriostatic activity after eighteen launderings. One-bath treatments with a Zr-pyrithion complex produced antimicrobial activity that was initially extremely high but was less durable to laundering. The two-bath process appears to be a general method for durably binding small amounts of a compound to cellulose.

        The activity of LDH and the variation of LDH isozyme pattern in the serum and muscle of snakehead which keeped in the water polluted by HgCl₂ had been studied by the method of colorimetry and cellulose acetate electrophoresis gram and compared with each pattern of LDH isozyme. The activity of LDH was gradually increased in the serum of snakehead which keeped in the water polluted with 0.01ppm and 0.05ppm of HgCl₂ through the experimental period. LDH activity in the serum of snakehead which keeped in the water with 0.05ppm of HgCl₂ was increased more superos than the others. The LDH activity in the intramuscular of snakehead which keeped in the water with 0.01ppm of HgCl₂ was gradually decreased after 5 and 10 days. And also decreased the LDH activity in it which keeped in the water with 0.05ppm of HgCl₂. However it more decreased 5 pattern of LDH isozymes were separated in serum and muscle of snakehead which keeped in the both water with 0.01 and 0.05ppm HgCl₂. In the both of the snakehead which keeped with 0.05 and 0.01ppm it show a tendency that the percentage of LDH in serum and muscle was similary decreased and on the other hands the percentage of LDH_5 in serum and muscle was also similary increased in the whole experiment period.

        Norfloxacin 과 β - Cyclodextrin 간의 Inclusion Complex 에 관한 약제학적 연구

        To increase the bioavailability of norfloxacin, inclusion complex of antimicrobial agent norfloxacin with β-cyclodextrin was prepared and studied by the solubility method, spectrophotometric methods (UV, IR, ¹H-NMR), differential thermal analysis, powder X-ray diffractometry, the physical properties, the antimicrobial activity, DNA binding and in situ recirculation technique. The conclusions are summerized as following: 1) The inclusion complexation was identified by means of solubility, spectrophotometry (UV, IR, NMR), DTA and X-ray diffraction. 2) The molar ratio of norfloxacin-β-cyclodextrin complex was 1 : 1.3) The stability constant of norfloxacin-β-cyclodextrin complex was 21.5M^(-1), and both true and apparent partition coefficients of the inclusion complex were larger than those of norfloxacin. 4) The time required to dissolve 60% (T_(60)%) of the inclusion complex was 120 min in distilled water and in the artificial intestinal juice, while norfloxacin did not reach to 60% dissolution within 120 min. 5) The antimicrobial activity of the inclusion complex against Pseudomonas aeruginosa, Klebsiella pneumoniae and Staphylococcus aureus showed no significant difference compared to that of norfloxacin alone. 6) Studies on binding properties between the inclusion complex and norfloxacin alone to DNA according to equilibrium dialysis showed no significant differency. 7) In situ absorption rates (Ka) of inclusion complex and norfloxacin alone were 0.229 and 0.102 hr^(-1), respectively.
