        For the isolation of mycobacteria in sputum specimens, it is essential to use the most gentle procedure of digestion system for the survival of organisms. The present studies were undertaken to evaluate and compare the effect of various sputum digestants on the death of Mycobacterium fortuitum and Mycobacterium tuberculosis. The results were summarized as follows: 1. Death rates of M. tuberculosis in a solution of 2% NaOH, 3% NaOH and 4% NaOH digestants after 30 minutes of contact time were 97.1%, 97.6% and 97.9%, respectively. In the digestants of 2% NaOH, 3% NaOH and 4% NaOH solution after 1 hour of contact time, survival rates were less than 2%. 2. Death rates of M. tuberculosis in Zephiran-N-acetyl-L-cysteine digestant after 30 and 60 minutes of contact time were 28.5% and 39.5%, while those of M. fortuitum after the contact time were 74.1% and 81.2%, respectively. 3. Death rates of M. tuberculosis in Zephiran-trisodium phosphate digestant after 30 and 60 minutes of contact time were 36.1% and 40.3%, and those of M. fortuitum after the contact time were 71.6% and 74.2%, respectively. 4. Death rates of M. tuberculosis and M. fortuitum in N-acetyl-L-cysteine-NaOH (2%) digestant were similar to the digestant of 2% NaOH solution.

      • 결핵균체 단백항원에 대한 항결핵균 IgG 항체에 관한 연구

        In order to study the level of IgG antibody binding to sonicate antigen of Mycobacteriumn tuberculosis H_34Rv for ammonium sulfate 30∼40 fraction in sera isolated from 209 patients and. 135 controls were measured by plastic tube enzyme-linked immunosorbent assay (ELISA). The levels of IgG antibody binding to Mycobacterium tuberculosis H_37Rv antigen in sera from all patients and controls were measured after absorption of 1 : 400 diluted sera with ,Wycobacteriuez bovis and dMlycobacterium Kansasii for ammonium sulfate 30∼40 fraction. The authors conducted the survey from August 1984 to January 1985. The results obtained were as follows : 1. At the serum dilution of 1:400, the mean ELISA values of IgG antibody to Mycobacterium tuberculosis H_37Rv after absorption with Mycobacterium kansasii antigen from far advanced patients and control were 0.8666±0.4903 and 0.2058±0.1353, respectively. 2. The mean ELISA value of IgG antibody to Mycobacterium tuberculosis H_37Rv afer absorption with Mycobacterium kansasii antigen from moderate patients was 0.6488±0.3918, the ratio of ammonium sulfate fraction 30∼40 in sera from patients and controls was 3.153. 3. The mean ELISA value of IgG antibody to Mycobacterium tuberculosis H_37Rv after absorption with Mycobacterium kansasii antigen from minimal patients was 0.4205±0.3155, the ratio of ammonium sulfate fraction 30∼40 in sera from patients and controls was 2.043.

      • 폐결핵 환자로부터 분리된 결핵균의 각종 항결핵제에 대한 감수성에 관한 연구

        Drug resistance, whether acquired as a result of inappropriate or inadequate chemotherapy or from an individual whose organism was drug resistant, may present serious obstacles to the eradication of tuberculosis. The authors attempted direct drug susceptibility test to various primary and second line antitubereulosis drugs on 139 isolates from pulmonary tuberculosis patients who have been hospitalized at National Kongju Hospital for tuberculosis. The results were summarized as follows; 1. Out of 101 tuberculosis patients, male patients were slightly more than female as a ratio of 1.59: 1 and the average age of onset was 30 years. 2. The number of patients who have been suffered from pulmonary tuberculosis more than 6 years was 61 (60.4%). 3. A11 of 139 isolated tubercle bacilii were resistant to one or more antituberculosis drugs. 4. Out of 139 isolates, 94.2% was resistant to INH, 89.2% to EMB, 75.5% to SM, 63.3% to PZA, 56.1% to RMP,47.5% to KM, 42.4% to PAS, 27.3% to CS, 16.5% to PTA and 2.9% to TUM. 5. Resistances to INH-SM-PAS, INH-SM-EMB, INH-SM-RMP, INH-EMB-RMP, PTA-PZA-CS and INH-SM-TUM were 36.0%, 69.1%, 41.0%, 46.8%, 7.2% and 2.9%, respectively. 6. Resistances to INH-EMB-RMP-PZA and INH-SM-RMP-PZA were 31.7% and 27.3%, respectively.

      • 임상실습에 임하는 학생들의 비강 및 피부에 분포된 포도구균과 환자병소에서 분리된 포도구균에 대한 병원성의 연관성에 관한 연구

        The authors have studied the pathogenecity of staphylococci isolated from 96 patients in Chungnam National University Hospital and Staphylococci isolated from nose and axilla of 50 medical and 50 nursing students in Chungnam National University. The authors conducted the survey from January 1986 to June 1986. The results obtained were as follows : 1. Out of staphylococci isolated from 96 patients, Staphylococcus aureus and Staphylococcus epidermidis were 59.4%, and 32.3%, respectively. In staphylococci isolated from nose of medical and nursing students, Staphylococcus aureus was 13.6^(male), 17.2%(female) and Staphylococcus epidermidis was 61.0%(male), 75.9%(female), respectively. 2. In the biotype of Staphylococcus epidermidis isolated from patients, the rate of biotype 1 was 93.5%. In the staphylococcus epidermidis isolated from nose of medical and nursing students, the rate of biotype 1 was 55.6%(male), 31.8%(female). In the axilla, the rate of biotype 1 was 56.5%(male), 52.6%(female), respectively. 3. In the biotype 1 of Staphylococcus epidermidis isolated from patients and control groups, enzymatic activities were more active than other biotypes. 4. Crystal violet ring test of Staphylococcus epidermidis isolated from patients, biotype 1 was WR type 22.6%, SR type 67.7% and DR type 3.2%. 5. To the antibiotic activities of staphylococci isolated from patients, Staphylococcus aureus was resistant to gentamycin(96.5%), penicillin(78.9%), lincomycin(47.4%) and erythromycine(35.1%). In the nose of medical students, Staphylococcus aureus was resistant to penicillin(11.9%), gentamycin(11.9%), lincomycin(6.8%) and tetracycline(5.1%). 6. In the multiple antibiotic resistance of staphylococci isolated from patients, Staphylococcus aureus was resistant to 2 antibiotics(35.1%), 3 antibiotics(17.5%), 5 antibiotics(15.8%) and, Staphylococcus epidermidis was resistant to 3 antibiotics(32.3%), 4 antibiotics(19.4%) and 5 antibiotics(16.1%).
