        유통 음료, 액상차 및 액상커피의 다량무기질 함량

        The aims of this study were to investigate some macrominerals (Na, Ca, P, K, Mg) in 207 beverages, 19 liquid teas, and 24 liquid coffees. The samples were digested by microwave and determinations of macrominerals were carried out by an Inductively Coupled Plasma Optical Emission Spectrometer (ICP-OES). The elements, listed in order of mean value of macromineral content, were potassium 208.4±298.2 mg/L (72.2±169.8 mg/container)> calcium 89.0±161.0 mg/L (26.0±57.7 mg/container)> sodium 71.2±75.0 mg/L (20.9±27.9 mg/container)> phosphorus 55.6±91.9 mg/L (17.9±33.8 mg/container)> magnesium 6.1±18.4 mg/L (2.4±10.1 mg/container). All 250 samples contained sodium and potassium, and the detection rate of calcium, phosphorus and magnesium was 88.4%, 93.2%, and 20.4%. The mean ratio of phosphorus to calcium in beverages, liquid teas, and liquid coffees was 4.2±16.0 (ND~164.4), and sports drinks showed the highest mean ratio (48.5±75.6) significantly (p<0.05). In case of sodium, detected content exceeding labeling regulations (less then 120%) was observed in 12 samples (5.5%).

        어린이와 청소년의 비알콜성음료 섭취에 따른 다량무기질 섭취량 평가

        The aims of this study were to estimate daily intake of macrominerals from beverages, liquid teas, and liquid coffees and to evaluate their potential health risks for Korean children and adolescents (1-to 19 years old). Assessment of dietary intake was conducted using the actual level of sodium, calcium, phosphorus, potassium, and magnesium in non-alcoholic beverages and (207 beverages, 19 liquid teas, and 24 liquid coffees) the food consumption amount drawn from “The Fourth Korea National Health and Nutrition Examination Survey (2007-2009)”. To estimate the dietary intake of nonalcoholic beverages, 6,082 children and adolescents (Scenario I) were compared with 1,704 non-alcoholic beverage consumption subjects among them (Scenario II). Calculation of the estimated daily intake of macrominerals was based on point estimates and probabilistic estimates. The values of probabilistic macromineral intake, which is a Monte-Carlo approach considering probabilistic density functions of variables, were presented using the probabilistic model. The level of safety for macrominerals was evaluated by comparison with population nutrient intake goal (Goal, 2.0 g/day) for sodium, tolerable upper intake level (UL) for calcium (2,500 mg/day) and phosphorus (3,000-3,500 mg/day) set by the Korean Nutrition Society (Dietary Reference Intakes for Koreans, KDRI). For total children and adolescents (Scenario I), mean daily intake of sodium, calcium, phosphorus, potassium, and magnesium estimated by probabilistic estimates using Monte Carlo simulation was, respectively, 7.93, 10.92, 6.73, 23.41, and 1.11, and 95th percentile daily intake of those was, respectively, 28.02, 44.86, 27.43, 98.14, and 3.87 mg/day. For consumers-only (Scenario II), mean daily intake of sodium, calcium, phosphorus, potassium, and magnesium estimated by probabilistic estimates using Monte Carlo simulation was, respectively, 19.10, 25.77, 15.83, 56.56, and 2.86 mg/day, and 95th percentile daily intake of those was, respectively, 62.67, 101.95, 62.09, 227.92, and 8.67 mg/day. For Scenarios I·II, sodium, calcium, and phosphorus did not have a mean an 95th percentile intake that met or exceeded the 5% of Goal and UL. (Korean J Nutr 2013; 46(1): 50 ~ 60)

        GC-tandem mass spectrometry를 이용한 분쇄원두커피 중 PAHs(polycyclic aromatic hydrocarbons) 분석법 연구 및 인체노출량 평가

        식품 중 존재하는 PAHs를 분석하기 위해 GC-MS/MS를 이용하여 분쇄원두커피를 대상으로 15종의 PAHs를 동시분석 하였다. 전처리법은 soxhlet법, 비누화분해 후 액체-액체 추출법, 초음파 후 고체-액체추출법으로 비교실험하여 재현성과 불순물제거효율이 가장 좋은 초음파 후 고체-액체 추출법을 선택하여 실험하였다. 실험법의 회수율은 PAHs종에 따라 52.6-93.3%, 검출한계는 0.002-0.1 μg/kg, 정량한계는 0.006-0.2 μg/kg로 PAHs분석에 가장 좋은 감도를 나타내는 HPLC-FLD나 GC-MS-SIM과 비슷하거나 낮은 수준의 검출한계를 나타내었다. 적합한 전처리 방법을 선택한 후 시중 유통되는 원두커피를 대상으로 15종의 PAHs 함량을 조사하였을 때 46종의 총 PAHs농도는 불검출-5.988 μg/kg이었다. 분쇄원두커피를 커피메이커로 추출하여 PAHs의 이행계수를 계산 한 후 WHO와 U.S.EPA에서 설정한 상대독성계수(TEF)를 근거로 음용되는 원두커피의 인체노출량(mg/kg b.w./day)을 구한 결과 5.24×10<sup>-8</sup> mg/kg b.w./day였다. The purpose of this study was to develop an analytical method for determining 15 polycyclic aromatic hydrocarbons (PAHs) of EU priority using gas chromatography (GC)-tandem mass spectrometry (MS). The PAHs in ground coffee were analyzed after being extracted using methods such as saponification-liquid-liquid extraction, Soxhlet extraction, and solid-liquid extraction. The solid-liquid extraction method showed the greatest repeatability and most efficient reduction of the matrix effect. GC-tandem MS for the quantification of the 15 PAHs showed better resolution and lower limit of detections (LODs) than GC-MS-selected ion monitoring (SIM) and high performance liquid chromatography with fluorescence detector. LODs of this method for the ground coffee types were 0.002-0.1 μg/kg and limit of quantifications (LOQs) were 0.006-0.2 μg/kg The recoveries ranged from 52.6 to 93.3%. Forty-six commercial types of ground coffee were analyzed to determine their PAHs contamination levels. PAHs concentration ranged from ND to 5.988 μg/kg. This study was conducted with toxicity equivalence factors, the U.S. EPA recommendation to identify dietary risks for PAHs in different types of coffee. The estimated average daily dose of PAHs was 5.24×10<sup>-8</sup> mg/kg body weight/day.

        RRLC-MS/MS를 이용한 견과류 및 그 가공품과 건조과실류의 아플라톡신 분석

        국내 유통되는 견과류 및 그 가공품, 건조과실류 총 225건을 대상으로 면역친화성칼럼으로 정제한 후 RRLC-MS/MS을 이용하여 아플라톡신에 대한 오염도를 조사하였다. 보다 효율적인 아플라톡신 분석을 위해 RRLC-MS/MS를 이용하여 직선성, 정확성, 정밀성, 검출한계 및 정량한계로 분석법의 유효성 검증을 하였다. 상관계수(R²)는 0.9999 이상의 우수한 직선성을 보였고, 아플라톡신의 회수율은 71.1-97.2%였고, RSD는 0.25-4.50%로 우수한 재현성을 나타냈다. 검출한계는 0.02-0.05 ㎍/㎏였고, 정량한계는 0.05-0.10 ㎍/㎏였다. 확립된 분석법에 의해 분석한 결과 그 오염수준은 B₁은 0.10-9.94 ㎍/㎏, B₂는 0.08-1.54 ㎍/㎏, G₁은 0.04-3.21 ㎍/㎏, G₂는 0.06-0.14 ㎍/㎏였다. 본 연구결과에서 나타난 오염수준은 국내규격 기준 이하였으나, 곰팡이독소의 생성 특이성을 고려한다면 다양한 식품 유형에 대한 지속적인 아플라톡신 함량조사와 함께 다른 곰팡이독소에 대한 연구도 필요하다고 판단된다. The occurrence of aflatoxins B₁, B₂, G₁ and G₂ in nuts, their products and dried fruits was investigated. Samples were collected from local markets in Seoul and analyzed by rapid resolution liquid chromatography coupled with tandem mass spectrometry using an immunoaffinity column. The chromatography method was validated for assay of aflatoxins using linearity, accuracy, precision and limit of detection and quantification. The linearity in the concentration ranged from 0.10 to 20 ㎍/㎏ with R²>0.9999. Sample recoveries ranged from 71.1 to 97.2% with relative standard deviations below 4.5% for spiking levels from 1 to 10 ㎍/㎏. The limits of detection ranged between 0.02 and 0.05 ㎍/㎏ and the limits of quantification ranged between 0.05 and 0.10 ㎍/㎏. The levels of aflatoxin B₁, B₂, G₁ and G₂ in nuts, their products and dried fruits were B₁ 0.10 to 9.94 ㎍/㎏, B₂ 0.08 to 1.54 ㎍/㎏, G₁ 0.04 to 3.21 ㎍/㎏ and G₂ 0.06 to 0.14 ㎍/㎏.
