Identification of PhoB Binding Sites of the yibD and ytfK Promoter Regions in Escherichia coli

By using a lacZ operon fusion genomic library of the Escherichia coli O157:H7 Sakai, we identified phosphatestarvation-inducible (psi) promoters located upstream of the yibD and ytfK genes. They have been previously proposed to belong to the phosphate regulon (pho regulon) by Beak and Lee (2006), based on the DNA array and in vivo transcriptional experiments. However, the direct interaction of these promoters with the activator protein of the pho regulon, PhoB, has not been determined. We determined the binding regions of PhoB in these promoter regions by DNase I footprinting. Both regions contained two pho boxes similar to the consensus sequence for PhoB binding.

Multi-cell inverter include an array of single-phase inverters capable of handling higher voltage ratings with lower rating power electronics devices. In this paper, a deadbeat control combined with a multi-sampling compensation method is proposed to control a Variable Frequency Drive (VFD) fed by a multi-cell inverter and realize real time digital feedback control using FPGA based hardware controller. This digital control is characterized by a very fast transient response, low switching losses, and a proper compensation for load disturbances and cyclical fluctuations.

In this paper, the use of a seawater desalination system as a controllable load is proposed to compensate the power fluctuation of renewable energy sources and control methods are discussed. On/Off control tests were conducted with the actual desalination system without a power converter and variable pump speed tests were conducted with a experimental desalination system with a power converter. The On/Off control turned out to be not so useful for power quality enhancement. With a power converter, the power consumption of the system can be controlled with enough accuracy and response speed for the suppression of the power fluctuations caused by wind turbines and photovoltaic cells.

Purpose To reveal the outcomes of partial medial meniscus posterior root tears following transtibial pullout repair compared with the outcomes of complete radial meniscus posterior root tears. Materials and methods We retrospectively evaluated 15 consecutive patients (male/female, 5/10; average age, 64.4 years) who underwent transtibial pullout repair for partial medial meniscus posterior root tears and compared their results with those of 86 consecutive patients who underwent the same surgery for complete medial meniscus posterior root tears. All patients underwent second-look arthroscopy on average 1 year postoperatively, and a semiquantitative meniscal healing score (anteroposterior width, stability, and synovial coverage, total 10 points) was evaluated. Medial meniscus extrusion was evaluated preoperatively and at second-look arthroscopy. Results Postoperative clinical scores were not significantly different in the short term. However, second-look arthroscopy revealed a significant difference in repaired meniscal stability (partial tear; 3.3 points, complete tear; 2.3 points, p < 0.001) and total meniscal healing scores (partial tear; 8.3 points, complete tear; 7.1 points, p < 0.001). Medial meniscus extrusion progression was significantly different (partial tear; 0.4 mm, complete tear; 1.0 mm, p < 0.001). Conclusion Partial medial meniscus posterior root tears showed better meniscal healing and less medial meniscus extrusion progression following pullout repair than complete medial meniscus posterior root tears.

This paper presents the effectiveness evaluation approach of demand side management (dema nd control) for frequency regulation in regional power networks (so called Cluster-oriented networks). The analytical model of the frequency fluctuation for a three-cluster interconnected network model was developed with MATLAB/SIMULINK. The control method is similar to the conventional TBC (Tie Line Bias Control) widely used in power grids. In order to maintain the quality of frequency, Battery Energy Storage Systems (BESS) and controllable loads such as heat pumps installed in the customer side will play an effective role in clusters. By the results of conducted simulations, it shows that the proposed new frequency regulation scheme by utilizing controllable loads such as battery charges and heat pumps retains effectiveness to suppress frequency deviation when a large amount of natural renewable energy is installed in the regional cluster-oriented networks.

Battery energy storage system (BESS) is attracting attention all over the world. BESS yields two types of benefits, i.e., economical benefit and environmental benefit. Economical benefit is yielded by reducing the cost of power plants that is composed by the operation cost and the construction cost. Recent serious concern against environment will be more important in the near future and it might be treated as penalty and/or constraints. The installation merit of BESS stands out when load factor of demand is low. If base generators composed of inexpensive and clean sources, and if peak generators composed of expensive and dirty sources, the benefit against operation cost becomes more remarkable. Besides, BESS can contribute to developing countries by reducing construction cost of generation plants. In this paper, the numerical analysis of this benefit is presented. The optimal amount of BESS is searched by evaluating CO2-reductions and cost benefits.
