      • Possible Epithelial Ovarian Cancer Association with HPV18 or HPV33 Infection

        The present study was conducted to investigate the prevalence of HPV infection in epithelial ovarian cancer (EOC) in Hunan province. DNA samples were collected from paraffin embedded ovarian tissue from 322 patients with EOC, 99 with ovarian benign tumors and 199 normal persons. The polymerase chain reaction and direct sequencing were used to identify the HPV types in the samples. The relationship between the infection of human papillomavirus (HPV) and the epithelial ovarian carcinoma (EOC) was investigated combined with clinical data. The prevalence of HPV18 and HPV33 in EOC group and benign group was higher than in the normal group. HPV18 and HPV33 may play a role in the development of both EOC and ovarian benign tumor and may participate in the development of EOC with traditional risk factors, family history and abortion, possibly exerting synergistic effects.

        Effects of roasting on composition of chili seed and storage stability of chili seed oil

        This work focused on how roasting changed the chemical components of chili (Capsicum annuum L.) seeds and how it affected the stability of chili seed oils during storage. The oils from chili seeds before and after roasting treatments were stored at 63 °C for 30 days and then analyzed. Results showed that roasting changed the main sugars compositions and amino acid compositions (total content decreased from 15.9 to 7.4%), which confirmed that it could form brown pigments and volatile flavor compounds in pepper seeds after roasting. Compared with oil from unroasted seeds, oils from roasted seeds had greater oxidative stability and maintained greater antioxidant capacity during storage. These effects were possibly due to the synergistic of the neo-formed products by Maillard reaction, vitamin E, and other bioactive components. This investigation showed that roasting treatment could be considered as an appropriate method for extending the storage stability of chili seed oils.

        Thruster fault diagnosis method based on Gaussian particle filter for autonomous underwater vehicles

        Autonomous Underwater Vehicles (AUVs) generally work in complex marine environments. Any fault in AUVs may cause significant losses. Thus, system reliability and automatic fault diagnosis are important. To address the actuator failure of AUVs, a fault diagnosis method based on the Gaussian particle filter is proposed in this study. Six free-space motion equation mathematical models are established in accordance with the actuator configuration of AUVs. The value of the control (moment) loss parameter is adopted on the basis of these models to represent underwater vehicle malfunction, and an actuator failure model is established. An improved Gaussian particle filtering algorithm is proposed and is used to estimate the AUV failure model and motion state. Bayes algorithm is employed to perform robot fault detection. The sliding window method is adopted for fault magnitude estimation. The feasibility and validity of the proposed method are verified through simulation experiments and experimental data.

        Gut microbiota metabolic characteristics in coronary artery disease patients with hyperhomocysteine

        Hyperhomocysteine (HHcy) is known as a risk factor for coronary artery disease (CAD). Despite the knowledge that gut microbiota related metabolism pathway shares metabolites with that of Hcy, little has been shown concerning the association between HHcy and gut microbiota. To explore their relationship in the context of CAD, 105 patients and 14 healthy controls were recruited from one single medical center located in Beijing, China. Their serum and fecal samples were collected, with multi-omics analyses performed via LC/MS/ MS and 16S rRNA gene V3-V4 region sequencing, respectively. Participants from the prospective cohort were divided into CAD, CAD & HHcy and healthy controls (HC) groups based on the diagnosis and serum Hcy concentration. The results revealed significant different metabolic signatures between CAD and CAD & HHcy groups. CAD patients with HHcy suffered a heavier atherosclerotic burden compared to CAD patients, and the difference was closely associated to betaine-homocysteine S-methyltransferase (BHMT)-related metabolites and trimethylamine N-oxide (TMAO)-related metabolites. Dimethylglycine (DMG) exhibited a strong positive correlation with serum total Hcy (tHcy), and TMAO and trimethylysine (TML) were associated with heavier atherosclerotic burden. Multiple other metabolites were also identified to be related to distinct cardiovascular risk factors. Additionally, Clostridium cluster IV and Butyricimonas were enriched in CAD patients with elevated tHcy. Our study suggested that CAD patients with elevated tHcy were correlated with higher atherosclerotic burden, and the impaired Hcy metabolism and cardiovascular risk were closely associated with BHMT-related metabolites, TMAO-related metabolites and impaired gut microbiota homeostasis.

        Seismic Response of Two Site Models and Their Effects on the Railway Cable-Stayed Bridge

        In order to accurately analyze the seismic response of the site and their effects on a railway cable-stayed bridge, the site condition of the railway cable-stayed bridge was surveyed, and geotechnical exploration holes ZK1 and ZK2 were set near the left and right tower of the bridge to gain the detailed geological data, then 32 representative ground motions records were selected corresponding to the site characteristics, and the equivalent linear and nonlinear model were established to analyze the site seismic response of 32 representative ground motions based on the Deepsoil Software. Next, to investigate the influence of site seismic response on the long-span railway cable-stayed bridge, the seismic response of the cable-stayed bridge considering site effect are calculated and evaluated via time history analysis under one-dimensional and multi-point excitation based on the ANSYS platform. At last, several critical and meaningful conclusions are drawn.

        Circulating Low Absolute CD4+ T Cell Counts May Predict Poor Prognosis in Extranodal NK/T-Cell Lymphoma Patients Treating with Pegaspargase-Based Chemotherapy

        Purpose Extranodal natural killer/T-cell lymphoma, nasal type (ENKTL) is a rare subtype of non- Hodgkin lymphoma, and asparaginase-based regimens are the best first-line treatments. Data on the role of specific circulating lymphocyte subsets in the progression of ENKTL are limited. The aim of this study was to investigate the clinical correlation and distribution of circulating absolute CD4+ T-cell counts (ACD4Cs) in ENKTL. Materials and Methods We retrospectively searched medical records for 70 newly diagnosed ENKTL patients treated with pegaspargase-based regimens. Comparison of ACD4Cs as a continuous parameter in different groups was calculated. Univariate and multivariate analyses were used to assess prognostic factors for overall survival (OS) and progression-free survival (PFS). Results Stage III/IV, B symptoms, elevated lactate dehydrogenase, monocytopenia, high-intermediate and high risk International Prognostic Index (IPI) and Korean Prognostic Index (KPI), high risk Prognostic Index of Natural Killer Lymphoma (PINK), and lower lymphocytes were significantly associated with low ACD4C at diagnosis. With a median follow-up time of 32 months, patients who had an ACD4C < 0.30109/L had a worse OS. Median OS was 11 months and median PFS was 5 months in the low ACD4C cohort. There were significant differences in both OS and PFS between the two cohorts. Moreover, multivariate Cox analysis identified ACD4Cs as an independent predictor for OS and PFS. Conclusion Low ACD4Cs were associated with poorer survival and could act as a negative predictor for ENKTL patients treated with asparaginase-based regimens.

      • EKI1 (Epha2 Kinase Inhibitor 1) Suppresses Tumor Growth in Hepatocellular Carcinoma and Cholangiocarcinoma by Inducing Autophage and Apoptosis

        Aims: Erythropoietin-producing hepatocellular receptor tyrosine kinase subtype A2 (EphA2) is an attractive therapeutic target for suppressing tumor progression. EKI1 is coordination complex of zinc and known for its use in treating dandruff and seborrhoeic dermatitis. The aim of this study is to discover novel small molecules to inhibit EphA2 for the treatment of hepatocellular carcinoma and cholangiocarcinoma. Methods: To discover novel and potent EphA2 inhibitors, we performed HTRF (Homogeneous time resolved fluorescence) kinase assay using the chemical library of Korea Chemical Bank and primary screened novel hit compounds. The enhancement of EKI1-mediated apoptosis and autophage were examined using immunoblotting and FACS analysis. Additionally, the antitumor effect of EKI1 was assessed using a mouse model. Results: Thirty-six compounds screened as EphA2 kinase inhibitor by HTRF assay. We validated these substances related to inhibit cell proliferation and cell death. We identified EKI1, a effective theranostics based small molecules. Human hepatocellular carcinoma cell line and cholangiocarcinoma cell lines were treated various concentration of EKI1 for 12h. At low concentration of EKI1, proliferation of these cells was inhibited. At high concentration of EKI1, cell death was induced in these cells. As quantitatively assessed by flow cytometry, apoptosis was induced by EKI1 in cells. We investigated apoptotic signaling by Western blot and observed cleavage or overexpression of pro-caspase-7 and PARP in EKI1 treated cells compared with vehicle. The anti-proliferation effect of EKI was due to an increased autophage, which was confirmed by up-regulation of Autophagy protein 5 (Atg5), BECN and LC3 (autophagosome marker). In addition, EKI1 induced reactive oxygen species (ROS) in JCK and Huh7. We examined EKI1 up-or down-signal transduction pathways to use therapeutic target for HCC and CC. In vivo mouse model, tumor growth was suppressed in EKI1 injected mouse group compared with control group. Conclusions: Our results revealed that autophage and apoptosis are involved in EKI1-mediated tumor cell death. Therefore, the EphA2 kinase inhibitor EKI1 is therapeutic target for hepatocellular carcinoma and cholangiocarcinoma.

        MicroRNA-375 is a therapeutic target for castration-resistant prostate cancer through the PTPN4/STAT3 axis

        The functional role of microRNA-375 (miR-375) in the development of prostate cancer (PCa) remains controversial. Previously, we found that plasma exosomal miR-375 is significantly elevated in castration-resistant PCa (CRPC) patients compared with castration-sensitive PCa patients. Here, we aimed to determine how miR-375 modulates CRPC progression and thereafter to evaluate the therapeutic potential of human umbilical cord mesenchymal stem cell (hucMSC)-derived exosomes loaded with miR-375 antisense oligonucleotides (e-375i). We used miRNA in situ hybridization technique to evaluate miR-375 expression in PCa tissues, gain- and loss-of-function experiments to determine miR-375 function, and bioinformatic methods, dual-luciferase reporter assay, qPCR, IHC and western blotting to determine and validate the target as well as the effects of miR-375 at the molecular level. Then, e-375i complexes were assessed for their antagonizing effects against miR-375. We found that the expression of miR-375 was elevated in PCa tissues and cancer exosomes, correlating with the Gleason score. Forced expression of miR-375 enhanced the expression of EMT markers and AR but suppressed apoptosis markers, leading to enhanced proliferation, migration, invasion, and enzalutamide resistance and decreased apoptosis of PCa cells. These effects could be reversed by miR-375 silencing. Mechanistically, miR-375 directly interfered with the expression of phosphatase nonreceptor type 4 (PTPN4), which in turn stabilized phosphorylated STAT3. Application of e-375i could inhibit miR-375, upregulate PTPN4 and downregulate p-STAT3, eventually repressing the growth of PCa. Collectively, we identified a novel miR-375 target, PTPN4, that functions upstream of STAT3, and targeting miR-375 may be an alternative therapeutic for PCa, especially for CRPC with high AR levels.
