        Three sesquiterpene lactones suppress lung adenocarcinoma by blocking TMEM16A-mediated Ca2+-activated Cl− channels

        Transmembrane protein TMEM16A, which encodes calcium-activated chloride channel has been implicated in tumorigenesis. Overexpression of TMEM16A is associated with poor prognosis and low overall survival in multiple cancers including lung adenocarcinoma, making it a promising biomarker and therapeutic target. In this study, three structure-related sesquiterpene lactones (mecheliolide, costunolide and dehydrocostus lactone) were extracted from the traditional Chinese medicine Aucklandiae Radix and identified as novel TMEM16A inhibitors with comparable inhibitory effects. Their effects on the proliferation and migration of lung adenocarcinoma cells were examined. Whole-cell patch clamp experiments showed that these sesquiterpene lactones potently inhibited recombinant TMEM16A currents in a concentration- dependent manner. The half-maximal concentration (IC50) values for three tested sesquiterpene lactones were 29.9 ± 1.1 μM, 19.7 ± 0.4 μM, and 24.5 ± 2.1 μM, while the maximal effect (Emax) values were 100.0% ± 2.8%, 85.8% ± 0.9%, and 88.3% ± 4.6%, respectively. These sesquiterpene lactones also significantly inhibited the endogenous TMEM16A currents and proliferation, and migration of LA795 lung cancer cells. These results demonstrate that mecheliolide, costunolide and dehydrocostus lactone are novel TMEM16A inhibitors and potential candidates for lung adenocarcinoma therapy.

      • Potential Therapeutic Targets for the Primary Gallbladder Carcinoma: Estrogen Receptors

        Gallbladder carcinoma, the most frequent malignant neoplasm of the biliary tract system, has always been considered to feature late clinical presentation and diagnosis, limited treatment options and an extremely poor prognosis. In recent years, while the incidence of gallbladder cancer has appeared to be on the increase, the available treatment methods have not greatly improved survival of the affected patients. Thus, exploring new therapeutic targets for this devastating disease is an urgent matter at present. Epidemical studies have demonstrated that the incidence of gallbladder carcinoma exhibits a distinct gender bias, affecting females two to three times more than males, pointing to crucial roles of estrogen. It is well known that estrogen acts on target tissues by binding to estrogen receptors (ERs), which are mainly divided into three subtypes, $ER{\alpha}$, $ER{\beta}$ and $ER{\gamma}$. $ER{\alpha}$ and $ER{\beta}$ appear to have overlapping but also unique even opposite biological effects. As important pathogenic mediators, ERs have been considered to relate to several kinds of tumors. In gallbladder carcinoma tissue, ERs have been shown to be positively expressed, and ERs expression levels are associated with differentiation and prognosis of this cancer. Nevertheless, the exact mechanisms of estrogen inducing growth of gallbladder carcinoma remain poorly understood. On the base of the current investigations, we deduce that estrogen participates in promotion of gallbladder carcinoma by influencing the formation of gallstones, stimulating angiogenesis, and promoting abnormal proliferation. Since ERs mediate the carcinogenic actions of estrogen in gallbladder, and therapy targeting ERs may provide new directions for gallbladder carcinoma. Therefore, it should be stressed that ERs are potential therapeutic targets for gallbladder carcinoma.

        Meal Assistance System Operated by Electromyogram (EMG) Signals: Movement Onset Detection with Adaptive Threshold

        The objective of this study is to design a meal assistance system operated by electromyogram (EMG) signals. The system consists of four parts, EMG data acquisition, onset analysis, control circuit and operation of meal assistance robot. An algorithm based on EMG power with adaptive threshold is proposed for onset analysis, and applied to the operation of robot via control circuit. The performance of EMG-control system is tested in rate of success and comfort by both subjective and objective indices. The results show EMG signals acquired from active muscles in a human body can be used as control signals for the apparatus, by the proposed onset detection algorithm with adaptive threshold. The high rate of success and comfort of meal assistance system operated by EMG signals guarantee its use for patients with disabled upper limbs.

      • Expression of Tumor Necrosis Factor Receptor-associated Factor 6 in Lung Cancer Tissues

        Background: Tumor necrosis factor (TNF) receptor-associated factor 6 (TRAF6) has been reported to be associated with the development of various cancers. However, the role of TRAF6 in lung cancer remains unclear. Objective: To explore the expression and clinicopathological significance of TRAF6 protein in lung cancer tissues. Materials and Methods: Three hundred and sixty-five lung cancer samples and thirty normal lung tissues were constructed into 3 microarrays. The expression of TRAF6 protein was determined using immunohistochemistry (IHC). Furthermore, correlations between the expression of TRAF6 and clinicopathological parameters were investigated. Results: The expression of TRAF6 in total lung cancer tissues (365 cases), as well as in small cell lung cancer (SCLC, 26 cases) and non-small cell lung cancer (NSCLC, 339 cases) was significantly higher compared with that in normal lung tissues. The ROC curve showed that the area under curve of TRAF6 was 0.663 (95%CI 0.570~0.756) for lung cancer. The diagnostic sensitivity and specificity of TRAF6 were 52.6% and 80%, respectively. In addition, the expression of TRAF6 was correlated with clinical TNM stage, tumor size and lymph node metastasis in all lung cancers. Consistent correlations were also observed for NSCLCs. Conclusions: TRAF6 might be an oncogene and the expression of TRAF6 protein is related to the progression of lung cancer. Thus, TRAF6 might become a target for diagnosis and gene therapy for lung cancer patients.

      • Elevated Serum Ferritin Levels in Patients with Hematologic Malignancies

        Purpose: To retrospectively analyze variability and clinical significance of serum ferritin levels in Chinese patients with hematologic malignancies. Materials and Methods: Serum ferritin were measured by radioimmunoassay, using a kit produced by the Beijing Institute of Atomic Energy. Patients with hematologic malignancies, and treated in the Department of Hematology in Nanjing First Hospital and fulfilled study criteria were recruited. Results: Of 473 patients with hematologic malignancies, 262 patients were diagnosed with acute leukemia, 131 with lymphoma and 80 with multiple myeloma. Serum ferritin levels of newly diagnosed and recurrent patients were significantly higher than those entering complete remission stage or in the control group (p<0.001). Conclusions: Serum ferritin lever in patients with hematologic malignancies at early stage and recurrent stage are significantly increased, so that detection and surveillance of changes of serum ferritin could be helpful in assessing conditions and prognosis of this patient cohort.

      • Polypropylene–waste ground rubber tire powder microcellular composites: Effect of processing variables on morphology and physico-mechanical properties

        <P>Microcellular composites made from polypropylene and waste ground rubber tire powder using azodicarbonamide as chemical blowing agent have been investigated. A small quantity of SEBS-g-MA has been added as compatibilizer. The effect of variation in processing variables such as pressure, heating time, and operating variables such as blowing agent and crosslinker concentrations have been investigated to establish a relationship between the structure and morphology of the microcellular composite and ultimate performance characteristics such as tensile strength, tear strength, and elongation at break. The results indicate that both processing and operating variables have tremendous effect on the morphological features such as average, minimum, and maximum cell size as observed from scanning electron microscopy.         Surface coordination and excellent anticorrosion performance of strontiumapatite nanocomposite

        A mass-produced, simple and scalable self-heating-based method for the manufacture of surfacecoordination strontiumapatite (SP) nanocomposite with excellent corrosion resistance is proposed. Thereaction heat released from the formation of SP material increases the solubility of benzotriazole (BTA)which achieves reasonable distribution of system energy. The BTA isfirmly adhered on the SP by Sr Ncoordination bond, which acts as barrier impeding on the penetration of the corrosive medium. Scanelectron microscopy (SEM) and X-ray diffraction (XRD) were employed to characterize coatingmorphology and chemical composition. Meanwhile, electrochemical impedance spectroscopy (EIS) datashow that the resultant resistance of strontiumapatite/benzotriazole (SPB) nanocomposite coating was3.02 × 104V cm2, which was improved by 601.8% compared with SP material. The improvement ofcorrosion performance is attributed to the synergistic anticorrosion mechanism that includes theshielding effect, extension of charge transfer paths and formation of multiple passivationfilms. Themethod for organic and inorganic synergistic material provides ideas for the design and synthesis ofanticorrosive materials.

      • Influence of regenerated polyurethane on the mechanical properties and thermal stability of polypropylene during multiple processing cycles

        <P>This work mainly introduces the recyclability of polypropylene (PP) modified by regenerated polyurethane (PU) that was obtained after partial thermochemical decomposition of waste PU. The influence of regenerated PU on the mechanical properties and thermal stability of PP during multiple processing cycles was evaluated by a universal testing machine, differential scanning calorimetry, and thermogravimetric analyses. The data showed that the mechanical properties of the modified PP were dependent on the content of regenerated PU with the increase in the number of processing cycles, whereas the thermal stability of the modified PP was almost independent of the content of regenerated PU.         Suppression of ICI and MAI in SC-FDMA communication system with carrier frequency offsets

        <P>Similar to other orthogonal frequency division multiplexing (OFDM)-based systems, carrier frequency offset (CFO) is a challenging problem in uplink communications of single carrier frequency division multiple access (SC-FDMA) system. It must be noticed that CFO, which is mainly due to oscillator instability and/or Doppler shift, would generate inter carrier interference (ICI) as well as multi-access interference (MAI) to disturb the received signals and seriously degrade system performance. Frequency synchronization in uplink communications is difficult because different users always experience different CFOs and one user¿s CFO correction would misalign other users. In this paper, we proposed a suppression method to overcome the multi CFOs problem. To implement this algorithm, block type pilots would be exploited, which is also utilized in LTE uplink standard. The proposed algorithm is based on the following two assumptions. Firstly the proposed algorithm is applied in this scenario, where different users start to communicate with the base station at different symbol periods, and secondly, during the pilot block and the following data blocks, the CFO of each user is quasi static, which is feasible since CFO is slow varying. Compared with other interference suppression methods, the proposed method could directly estimate the interference components from the inverse pilot matrix, thus it does not need to do the CFO estimation. Further more, since block type pilots is a common pilot pattern in wireless communication system, this algorithm can be easily extended to other communication system. Simulation results show that the proposed suppression algorithm can significantly improved system performance.</P>
