      • Roles of PTEN (Phosphatase and Tensin Homolog) in Gastric Cancer Development and Progression

        Gastric cancer is highly invasive, aggressively malignant, and amongst the most prevalent of all forms of cancer. Despite improved management strategies, early stage diagnosis of gastric cancer and accurate prognostic assessment is still lacking. Several recent reports have indicated that the pathogenesis of gastric cancer involves complex molecular mechanisms and multiple genetic and epigenetic alterations in oncogenes and tumor suppressor genes. Functional inactivation of the tumor suppressor protein PTEN (Phosphatase and Tensin Homolog) has been detected in multiple cases of gastric cancer, and already shown to be closely linked to the development, progression and prognosis of the disease. Inactivation of PTEN can be attributed to gene mutation, loss of heterozygosity, promoter hypermethylation, microRNA- mediated regulation of gene expression, and post-translational phosphorylation. PTEN is also involved in mechanisms regulating tumor resistance to chemotherapy. This review provides a comprehensive analysis of PTEN and its roles in gastric cancer, and emphasizes its potential benefits in early diagnosis and gene therapy-based treatment strategies.

      • Platycodin D Induces Apoptosis, and Inhibits Adhesion, Migration and Invasion in HepG2 Hepatocellular Carcinoma Cells

        Background: Platycodin D (PD), a triterpenoid saponin isolated from the Chinese medicinal herb Platycodonis radix, possesses anti-cancer effects in several cancer cell lines. The aim of this study was to evaluate its anticancer activities in hepatocellular carcinoma cells. Materials and Methods: MTT and colony formation assays were performed to evaluate cell proliferation, along with flow cytometry and Western blotting for apoptosis. Cell adhesion was tested by observing cellular morphology under a microscope, while the transwell assay was employed to investigate the cell migration and invasion. Results: PD concentration-dependently inhibited cell proliferation in both HepG2 and Hep3B cells, and significantly suppressed colony formation and induced apoptosis in HepG2 cells. The protein levels of cleaved poly ADP-ribose polymerase (PARP) and Bax were up-regulated while that of survivin was down-regulated after treatment with PD. Moreover, PD not only obviously suppressed the adhesion of HepG2 cells to Matrigel, but also remarkably depressed their migration and invasion induced by 12-O-tetradecanoylphorbol 13-acetate (TPA). Conclusions: PD presents anti-cancer potential in hepatocellular carcinoma cells via inducing apoptosis, and inhibiting cell adhesion, migration and invasion, indicating promising features as a lead compound for anti-cancer agent development.

      • SMC Reliable Design for T-S Model-Based Systems

        This paper studies the robust reliable control issues based on the Takagi-Sugeno (T-S) fuzzy system modeling method and the sliding mode control (SMC) technique. The combined scheme is shown to have the merits of both approaches. It not only alleviates the on-line computational burden by using the T-S fuzzy system model to approximate the original nonlinear one (since most of the system parameters of the T-S model can be computed off-line) but it also preserves the advantages of rapid response and robustness of the SMC schemes. Moreover, the combined scheme does not require on-line computation of any nonlinear term of the original dynamics and the increase in the partition number of the region of premise variables does not create extra on-line computational burdens for the scheme. Under the design, the control mission can continue safely without prompt external support even when the susceptible actuators fail to operate. The proposed analytical results are also applied to the attitude control of a spacecraft. Simulation results demonstrate the benefits of the proposed scheme.

        An Investigation Into the Relationship Between Metabolic Responses and Energy Regulation in Antibody-Producing Cell

        Energy-efficient metabolic responses were often noted in high-productive cultures. To better understand these metabolic responses, an investigation into the relationship between metabolic responses and energy regulation was conducted via a comparative analysis among cultures with different energy source supplies. Both glycolysis and glutaminolysis were studied through the kinetic analyses of major extracellular metabolites concerning the fast and slow cell growth stages, respectively, as well as the time-course profiles of intracellular metabolites. In three cultures showing distinct antibody productivities, the amino acid metabolism and energy state were further examined. Both the transition of lactate from production to consumption and steady intracellular pools of pyruvate and lactate were observed to be correlated with efficient energy regulation. In addition, an efficient utilization of amino acids as the replenishment for the TCA cycle was also found in the cultures with upregulated energy metabolism. It was further revealed that the inefficient energy regulation would cause low cell productivity based on the comparative analysis of cell growth and productivity in cultures having distinct energy regulation.

        Home-based traditional Chinese medicine nursing interventions for discharged patients with COVID-19: a rapid review of Chinese guidelines

        Background: The aim of this review was to comprehensively summarize and analyze the current guidelines on home-based traditional Chinese medicine (TCM) nursing interventions for discharged patients with COVID-19. Methods: Eight data sources were searched until June 28, 2020. The frequency of home-based TCM nursing interventions and the use of specific acupuncture points recommended in Chinese guidelines for discharged COVID-19 patients were computed and analyzed. Results: In total, we identified 5 Chinese guidelines that provide for home-based TCM nursing interventions for discharged patients with COVID-19. Moxibustion and acupressure were singled out as the most frequently used intervention of the 11 home-based TCM nursing interventions recommended by these guidelines. RN12 and ST36 were the 2 most promoted acupuncture points for moxibustion and acupressure interventions for these patients. Conclusions: The present review showed the important role of home-based TCM nursing interventions for discharged COVID-19 patients. However, direct evidence of their efficacy is still insufficient.

        Plasticity in Bud Demography of a Rhizomatous Clonal Plant Leymus chinensis L. in Response to Soil Water Status

        We surveyed the bud demography of Leymus chinensis L. plants along a soil-moisture gradient that was caused by a flood in 1998 on the Song-nen Plain in northeastern China. The number of vegetative buds per ramet was influenced by soil water content, with regression curves being quadratic and the opening of the parabola pointing downward. In addition, the optimum regression models for the numbers of rhizomatous buds and tiller buds relative to soil water resulted in a quadratic parabola and exponential curve, respectively. Vegetative buds flourished between August and October, with plants producing more of those buds on flooded plots than on control sites. The number of rhizomatous buds per ramet was much higher than for tiller buds throughout most of the growing season, and production of the former was more apt to be affected by soil water status. This observed superiority of rhizomatous bud production was thought to be a consequence of the whole-plant adjustment that was stimulated by an abnormally high moisture content. It could also be interpreted as a strategy for escape from disadvantageous overly wet conditions. Moreover, the position-based preference for bud emergence along the ramets could be an underlying mechanism for selective ramet placement.

        Preparation and Characterization of Tributyrin Sub-micron Emulsion as Carrier for Paclitaxel

        Paclitaxel is a well known anticancer agent and has been a pharmaceutical challenge because of its extremely poor water-solubility and susceptibility to the p-glycoprotein (p-gp)-mediated efflux in multi-drug resistant (MDR) cancer cells. Tributyrin (TB), a triglyceride with relatively short fatty acid chains, was chosen as solubilizing vehicle for paclitaxel based on the solubility study (26.6 mg/mL). Tributyrin (10%) o/w emulsion containing paclitaxel (5%), egg phosphatidylcholine (5%) and pegylated phospholipid (0.5%) was prepared by high pressure homogenization to obtain submicron- sized emulsion. The mean particle size of the resultant TB emulsion was 395.5 nm. Paclitaxel in TB emulsion showed higher anticancer activity against human breast cancer cell line, MCF-7, than free form delivered in DMSO solution. On the other hand, its anticancer activity was significantly reduced in MCF-7/ADR, a MDR variant cancer cell line of MCF- 7, and recovered by the presence of verapamil, suggesting of the susceptibility to the p-gp mediated efflux even though paclitaxel was encapsulated into emulsion. The TB emulsion showed great potential as a promising vehicle for water-insoluble anticancer agent, paclitaxel.

        Correlation between population size of pear Psylla (Cacopsylla chinensis) and leaf structure features in different pear cultivars

        The feeding preference of pear psylla (Cacopsylla chinensis) varies among pear cultivars. To clarify whether leaf structural factors are related to C. chinensis resistance, eleven pear cultivars of three pear species and their hybrids in China were studied. The population size of C. chinensis in plots of different cultivars was investigated, and leaf paraffin sections of each pear cultivar were observed. Correlation analysis results showed that the thickness of palisade tissue (PT) and the thickness ratio (TR) of PT to leaf (L) were the main factors influencing resistance to C. chinensis. The leaves of cultivar Wujiuxiang and those species from France and the USA exhibited the thickest PT and the highest TR; these three cultivars demonstrated much higher resistance to C. chinensis than did the other cultivars. The cultivar Gold Nijisseiki, with a large C. chinensis population size, showed low PT and TR. The findings of this study may provide valuable information to pear growers, experts and scientists regarding plant protection, cultivation and breeding when selecting suitable varieties to culture from the perspective of pest control.

      • An improved algorithm for pile damage localization based on complex continuous wavelet transform

        Since the complex continuous wavelet transform (CCWT) based pile damage detection method is empirical and subjective, an improved algorithm for pile damage localization based on CCWT is proposed by introducing K-means clustering and fast Fourier transform (FFT). In this method, the K-means clustering algorithm is used to accurately calculate the time coordinates of two energy concentrating points caused by the incident and reflected waves, respectively. Meanwhile, FFT is employed to estimate the concerned frequency band of the response signal. Therefore, a specific region in the time frequency plane is defined objectively and it can be used to search the phase angle turning points and localize pile damage. The proposed method is verified by numerical examples of piles with single and multiple damage positions. A parameter analysis is also conducted to investigate how damage depth and damage degree in piles affect the accuracy and effectiveness of the proposed method. The results demonstrate that the proposed method is able to localize a pile with a damage at least 2.5 m away from the pile head when the damage degree is as less as 5%. After that, dynamic tests of an actual square reinforced concrete pile and an actual circular reinforced concrete pile are investigated to verify the application of the proposed method on practical engineering. Although the proposed method is capable of localizing actual piles more accurately than the CCWT method, the problem of interference points needs to be addressed by mutual verification with other pile damage localization methods.
