FSCB phosphorylation in mouse spermatozoa capacitation

It is generally accepted that spermatozoa capacitation is associated with protein kinase A-mediated tyrosine phosphorylation. In our previous study, we identified the fibrous sheath CABYR binding protein (FSCB), which was phosphorylated by PKA. However, the phosphorylation status of FSCB protein during spermatozoa capacitation should be further investigated. To this aim, in this study, we found that phosphorylation of this 270-kDa protein occurred as early as 1 min after mouse spermatozoa capacitation, which increased over time and remained stable after 60 min. Immunoprecipitation assays demonstrated that the tyrosine and Ser/Thr phosphorylation of FSCB occurred during spermatozoa capacitation. The extent of phosphorylation and was closely associated with the PKA activity and spermatozoa motility characteristics. FSCB phosphorylation could be induced by PKA agonist DB-cAMP, but was blocked by PKA antagonist H-89.Therefore, FSCB contributes to spermatozoa capacitation in a tyrosine-phosphorylated format, which may help in further elucidating the molecular mechanism of spermatozoa capacitation. This study presents the shear capacity of eco-friendly high-strength concrete (HSC) beams reinforced by 0.75% (by volume) of hooked steel fibers or shear stirrups with various spacings. Five large-scale HSC beams with steel fibers or stirrups were tested and investigated with respect to shear capacity, failure mode, and crack patterns. The test results indicate that all five tested beams finally failed by a shear-critical failure mode and that the use of 0.75 vol% of steel fibers significantly improved the shear strength although it decreased the diagonal crack angle of HSC beam in the case of without stirrups. There are some different properties compared with ACI 318-19 recommendation, but the use of 0.75 vol% steel fibers exhibited approximately 13.2% higher shear strength when compared with that of minimum shear reinforcement for the HSC beam. Furthermore, the shear capacity of steel fibers calculated by experimental data, and it was compared with several prediction equations. The prediction models containing fiber factor were more closely agreeable with test results with a minimum of 10.9% difference.

Purpose: To investigate the prevalence of spina bifida occulta (SBO) and its relationship with the presence of overactive bladder (OAB) in middle-aged and elderly people in China. Methods: A cross-sectional community-based survey was carried out at 7 communities in Zhengzhou City, China from December 15, 2013 to June 10, 2014, where residents aged over 40 years were randomly selected to participate. All of the participants underwent lumbosacral radiographic analysis and relevant laboratory tests. A questionnaire including basic information, past medical history and present illness, and the OAB symptom score was filled out by all participants. Chi-square tests and logistic regression were used for data analysis with a P-value of <0.05 denoting statistical significance. Results: A total of 1,061 subjects were qualified for the final statistical analysis (58.8±11.7 years; male, 471 [44.4%]; female, 590 [55.6%]). The overall prevalence of SBO was 15.1% (160 of 1,061): 18.3% (86 of 471) in men and 12.5% (74 of 590) in women. Among these subjects, 13.7% (145 of 1,061) had OAB: 13.2% (62 of 471) in men and 14.1% (83 of 590) in women. The results of logistic regression showed that age, SBO, history of cerebral infarction (HCI), and constipation were risk factors for OAB (P<0.05), while sex, history of childhood enuresis (HCE), body mass index (BMI), and diabetes mellitus (DM) were not (P>0.05). In men, age, SBO, and constipation were risk factors for OAB (P<0.05), while HCE, BMI, DM, HCI, and benign prostate hyperplasia were not (P>0.05). In women, age, SBO, and HCI were risk factors for OAB (P<0.05), while HCE, BMI, DM, vaginal delivery, and constipation were not (P>0.05). Conclusions: The prevalence of SBO is high and it is related to OAB in middle-aged and elderly people in China.

Objective: The purpose of this study was to evaluate computed tomography (CT) virtual non-contrast (VNC) spectral imaging for gastric carcinoma. Materials and Methods: Fifty-two patients with histologically proven gastric carcinomas underwent gemstone spectral imaging (GSI) including non-contrast and contrast-enhanced hepatic arterial, portal venous, and equilibrium phase acquisitions prior to surgery. VNC arterial phase (VNCa), VNC venous phase (VNCv), and VNC equilibrium phase (VNCe) images were obtained by subtracting iodine from iodine/water images. Images were analyzed with respect to image quality, gastric carcinoma-intragastric water contrast-to-noise ratio (CNR), gastric carcinoma-perigastric fat CNR, serosal invasion, and enlarged lymph nodes around the lesions. Results: Carcinoma-water CNR values were significantly higher in VNCa, VNCv, and VNCe images than in normal CT images (2.72, 2.60, 2.61, respectively, vs 2.35, $p{\leq}0.008$). Carcinoma-perigastric fat CNR values were significantly lower in VNCa, VNCv, and VNCe images than in normal CT images (7.63, 7.49, 7.32, respectively, vs 8.48, p< 0.001). There were no significant differences of carcinoma-water CNR and carcinoma-perigastric fat CNR among VNCa, VNCv, and VNCe images. There was no difference in the determination of invasion or enlarged lymph nodes between normal CT and VNCa images. Conclusions: VNC arterial phase images may be a surrogate for conventional non-contrast CT images in gastric carcinoma evaluation.

As a kind of reactive power compensation equipment with control flexibility and high reliability, magnetically controlled reactor (MCR) is widely used in reactive power compensation, over-voltage limitation and other aspects. However, low responding speed has severely limited the application of MCR. Especially when applied to suppress voltage flicker and arc suppression coil, the slow response of MCR will decrease the stability of control system, even causing the system shock. To improve the response performance of MCR, in this paper, the working principle of MCR has been analyzed, and a fast response structure of MCR has been designed with the novel fast response structure, fast excitation and rapid demagnetization can be achieved. According to the simulation and experiment results, the effectiveness of the proposed structure is verified by limiting the response time in 30ms.

Circular honeycombs are extensively used to absorb impact energy considering its good energy absorption properties. Rather than focusing on the existing perfect honeycomb, effects of concentrated filling inclusions on the in-plane dynamic behavior of circular honeycomb structure are studied in detail by using finite element simulations. The crushing model of honeycomb under in-plane impact is established which then is used to investigate the influences of packing ratio, defect distribution area and impact velocity on the deformation mode and the plateau stress. Investigation results show that the deformation model of imperfect honeycomb can still be classified into three types: Quasi-static mode, transition mode and dynamic mode; However, the value of packing ratio and defect location may seriously influence localized deformed band of transition mode; The plateau stress of the honeycomb relies on the defect location except for the packing ratio; The filling inclusions defect concentrated in sub-region 2 is more beneficial to improve the energy-absorption capacity; Especially under middle or low impact velocities, it displays higher sensitivity; The crushing plateau stress of honeycomb with a = 0.36 and defect concentrated in sub-region 2 is even improved about 56.9 % under impact velocity 100 m/s. These results can provide valuable suggestions in the study and design of other honeycomb structures.
