Backgrounds Arctigenin derived from the seeds of Arctium lappa Linnaeus is known as an anticancer drug candidate by targeting various pathways involved in anticancer therapy. Methods Using 2D monolayer and 3D spheroid culture systems in nasal septum carcinoma RPMI-2650 cells, the effects of arctigenin and dexamethasone on cell viability, ROS levels, ATP level, mitochondrial function, apoptosis and necroptosis were examined. Results The combination treatment of both compounds induced strong cytotoxicity, accompanied by increases of sub-G0/ G1 peak, annexin V-PE-positive cells, and ROS levels, loss of mitochondrial membrane potential, and decrease of cellular ATP content. These changes were observed as simultaneous induction of DNA damage, apoptosis, and necroptosis. A series of changes by arctigenin and dexamethasone were efficiently restored by decreasing ROS levels or supplementing ATP. Treatment of 3D spheroids with arctigenin and dexamethasone decreased cell viability in the spheroids, but it was slightly resistant than cells under 2D conditions. In addition, this phenomenon was accompanied by an increase in mediators for both apoptosis and necroptosis. Conclusion Results of this study suggest that the apoptosis and necroptosis-inducing effects of arctigenin are associated with ATP depletion due to oxidative mitochondrial dysfunction.

Objective:Pterional craniotomy (PC) using myocutaneous (MC) flap is a simple and efficient technique; however, due to subsequent inferior displacement (ID) of the temporalis muscle, it can cause postoperative deformities of the muscle such as depression along the inferior margin of the temporal line of the frontal bone (DTL) and muscular protrusion at the inferior portion of the temporal fossa (PITF). Herein, we introduce a simple method for reconstruction of the temporalis muscle using a contourable strut plate (CSP) and evaluate its efficacy. Materials and Methods:Patients at follow-ups between January 2014 and October 2014 after PCs were enrolled in this study. Their postoperative deformities of the temporalis muscle including ID, DTL, and PITF were evaluated. These PC cases using MC flap were classified according to two groups; one with conventional technique without CSP (MC Only) and another with reconstruction of the temporalis muscle using CSP (MC + CSP). Statistical analyses were performed for comparison between the two groups. Results:Lower incidences of ID of the muscle (p < 0.001), DTL (p < 0.001), and PITF (p = 0.001) were observed in the MC + CSP than in the MC Only group. The incidence of acceptable outcome was markedly higher in the MC + CSP group (p < 0.001). ID was regarded as a causative factor for DTL and PITF (p < 0.001 in both). Conclusion:Reconstruction of the temporalis muscle using CSP after MC flap is a simple and efficient technique, which provides an outstanding outcome in terms of anatomical restoration of the temporalis muscle.
The Design of an Optimal Demand Response Controller Under Real Time Electricity Pricing
Jin, Young Gyu,Choi, Tae-Seop,Park, Sung Chan,Yoon, Yong Tae The Korean Institute of Electrical Engineers 2013 Journal of Electrical Engineering & Technology Vol.8 No.3
The use of a demand response controller is necessary for electric devices to effectively respond to time varying price signals and to achieve the benefits of cost reduction. This paper describes a new formulation with the form of constrained optimization for designing an optimal demand response controller. It is demonstrated that constrained optimization is a better approach for the demand response controller, in terms of the ambiguity of device operation and the practicality of implementation of the optimal control law. This paper also proposes a design scheme to construct a demand response controller that is useful when a system controller is already adapted or optimized for the system. The design separates the demand response function from the original system control function while leaving the system control law unchanged. The proposed formulation is simulated and compared to the system with simple dynamics. The effects of the constraints, the system characteristics and the electricity price are examined further.
