      • The clinical characteristics of the sensitive skin Department of Dermatology, Kangnam Sacred Heart Hospital, Hallym University, Seoul, Korea

        Background: Sensitive skin exhibits various clinical characteristics. Baumann classified sensitive skin into the four categories; stinging, allergic, acne and rosacea types Objectives: We aimed to investigate the clinical manifestations of sensitive skin according to the classification of Baumann Methods: With the sensitive skin scale-10, a questionnaire for measuring severity, the patients whose scores are over 50 and less than 20 were defined as sensitive skin group (n=37) and control group (n=21), respectively. Characteristics and objective measurements (transepidermal water loss [TEWL], epidermal moisture content, Von Frey filament test, demodex density) are recruited and sensitive skin group were divided according to the classification of Baumann Results: Most patients with sensitive skin were female and the mean age were 36.24 ± 11.90. The most common subtype was rosacea type followed by allergic type. Overall, the average of the TEWL in patient group was 23.21 ± 13.28, higher than control group (8.90 ± 3.09) and the reference value (14.0 ± 8.80). The epidermal moisture content of patient group was statistically lower than control group. There is no significant difference in TEWL and epidermal moisture content among subtypes. The demodex density of rosacea type is statistically higher than that of acne and stinging type Conclusion: There are distinct clinical characteristics in the four subtypes in sensitive skin. It will help to approach patients with sensitive skin

      • Hsc70B, An Antiviral Gene in Anopheles gambiae

        Mosquito-borne viral pathogens infect millions of people worldwide, often resulting in fatal infections. Our research interests in mosquito vector biology focuses on understanding of the molecular and genetic basis of mosquito resistance to arbovirus infections. Unlike mammalian cells including humans, for instance, mosquito cells do not show pathologic symptoms when infected by arboviruses. This observation led us to embarking a microarray study to investigate mosquito-virus interactions using Anopheles gambiae and o’nyong-nyong virus (ONNV) as a model system. As a result, we found that transcription of the hsc70B gene is increased about 2.6-fold in ONNV-infected An. gambiae compared to non-infected controls. Subsequently, in vivo RNAi silencing of the hsc70B transcript caused enhanced ONNV replication in female mosquitoes. Therefore, these results suggest that the hsc70B protein has an inhibitory effect on ONNV replication. A promoter analysis of the hsc70B locus further demonstrated that the hsc70B promoter is able to induce transcription of hsc70B in response to ONNV infection. In addition, hsc70B transcription was also induced by West Nile or La Crosse virus infection. Collectively, our findings indicate that hsc70B plays a role in suppressing virus replication as a general antiviral mechanism. Implications of hsc70B research and our other research endeavors toward the control of mosquito-borne infectious diseases will be discussed.

      • Clinical manifestations and patch-test results in facial contact dermatitis induced by disposable face masks

        Background: The patients with facial contact dermatitis by wearing a disposable mask were rapidly increased due to the outbreak of COVID-19 Objectives: The aim of this study was to investigate the clinical manifestation and results of allergy patch test for the patients with facial contact dermatitis induced by disposable mask. Methods: The clinical manifestations, and patch-test (T.R.U.E. TEST®) results of 26 patients with contact dermatitis induced by the disposable mask (mask group) and 70 patients by other cause (control group) are investigated as a control group. Results: The skin lesion was more common in centrofacial area (69.23%) than peripheral area (30.76%). Hyperkeratosis (23.07%) and xerosis (11.53%) were more frequent in mask group. Patients in mask group complained mostly itching (57.69%) and flushing (23.07%). Patients in mask group show more positive reaction of Potassium dichromate (26.92%, [mask group] vs 10.00%, [control group]) and 4-tert-butylphenol-formaldehyde resin (15.38%, [mask group] vs 2.85%, [control group]) than control group in patch-test results. Eleven patients in control group (15.71%) were negative reaction of all items in patch-test result, in contrast only one patient showed negative reaction in mask group (3.84%). The patients of mask group showed clinical improvement after changing to cloth mask Conclusion: Patients with the facial contact dermatitis induced by disposable mask presented different contact sensitization profiles compared to control group

        잠재적 학업중단청소년을 위한 개입프로그램 개발

        본 연구는 schmidt(2007)의 학교상담 프로그램 개발 절차에 근거하여 잠재적 학업중단청소년을 위한 개입프로그램을 개발하고, 그 효과를 검증하였다. 학업중단 청소년에 대한 선행 연구를 고찰하고, 전문상담교사 11명의 전문가 집단을 대상으로 잠재적 학업중단 청소년에게 필요한 프로그램의 구성 요인에 대해서 확인한 후, 프로그램의 수혜자에 대한 요구분석의 결과를 바탕으로 모듈식 접근을 활용하여 프로그램의 내용을 구성하였다. 서울 및 경기 지역에 거주하는 잠재적 학업중단청소년 18명(남9명, 여9명)을 대상으로 전문상담교사 6명이 개발된 프로그램을 적용하여, 프로그램의 효과성을 평가하였다. 측정도구는 학업중단의도와 학교적응유연성척도를 사용하였다. 효과검증을 위한 방법으로는 프로그램 실시 전과 후에 학업중단의도척도와 학교적응유연성척도의 변화 정도를 검증하였고, 추후에 실제 학업지속 여부를 확인하고, 프로그램 만족도를 포함한 종합평가지를 실시하였다. 또한 프로그램의 평가의 타당성 확보를 위해서 프로그램에 참여한 전문상담교사와 잠재적 학업중단 청소년을 대상으로 평가회의와 상담회기평가지 작성을 실시하였다. 연구 결과, 프로그램 실시 이후에 학업중단의도척도와 학교적응유연성척도의 점수가 유의미하게 상승하였으며, 잠재적 학업중단 개입 프로그램은 잠재적 학업중단 청소년의 학업중단 예방에 효과적으로 작용한다. 잠재적 학업중단 청소년에 대해서 예방적 차원에서의 개입을 강화할 필요성이 있다는 점을 제언하였다. The purpose of this study was to develop an intervention program for adolescents who have a high dropout risk, and thus contribute to the prevention of adolescent school dropouts and to adolescents' sound development. In order to increase effectiveness and field applicability of the program, needs assessments were conducted with school counselors who are supposed to run the dropout prevention program for high risk youth at school. The program was devised with seven modules and the study was conducted with 18 adolescents who were categorized as having a high dropout risk at 6 middle and high schools in Seoul and the Gyeonggi area. Pre-tests and Post-tests were conducted to assess the effectiveness of the program. The thoughts and attitudes dropout scale together with a school adjustment resiliency scale were applied. A program evaluation form was also administered. Counselors were also asked to fill out the program evaluation form. As a result, after participating in this program, students' motivation to dropout of school decreased. School counselors evaluated the program positively in general. Specifically, they reported that the program is timely, practical, and could contribute to adolescents' wellbeing. As for the students' satisfaction of the program, overall the satisfaction score was lower than school counselors,' but the scores on the program's easiness, practicality, and utilization were relatively high. Therefore, for future study, it is recommended that an intervention program for adolescents with a high dropout risk who are currently attending school, is developed. In order to support high dropout risk students and to increase their school adjustment, it is necessary to analyze all aspects of student lives. A comprehensive treatment approach, considering all these environmental aspects for high dropout risk youth could lead them to successful school adjustment.

      • Homogeneity Evaluation of Geopolymer Waste Form Containing Radioactive Spent Ion Resins Using LIBS and Statistical Approach

        The homogeneity of radioactive spent ion exchange resins (IERs) distribution inside waste form is one of the important characteristics for acceptance of waste forms in long-term storage because heterogenous immobilization can lead to the poor structural stability of waste form. In this study, the homogeneity of metakaolin-based geopolymer waste form containing simulant IERs was evaluated using a laser-induced breakdown spectroscopy (LIBS) and statistical approach. The cation-anion mixed IERs (IRN150) were used to prepare the simulant spent IERs contaminated by non-radioactive Cs, Fe, Cr, Mn, Ni, Co, and Sr (0.44, 8.03, 6.22, 4.21, 4.66, 0.48, and 0.90 mg/g-dried IER, respectively). The K2SiO3 solution to metakaolin ratio was kept constant at 1.2 and spent IERs loading was 5wt%. For the synthesis of homogeneous geopolymer waste form, spent IERs were mixed with K2SiO3 solution and metakaolin first, and then the fresh mixture slurry was poured into plastic molds (diameter: 2.9 cm and height: 6.0 cm). The heterogeneous geopolymer waste form was also fabricated by stacking two kinds of mixtures (8wt% IERs loading in bottom and 2wt% in top) in one mold. Geopolymers were cured for 7d (1d at room temperature and 6d at 60°C). The hardened geopolymers were cut into top, middle, and bottom parts. The LIBS spectra and intensities for Cs were obtained from the top and bottom of each part. Cs was selected for target nuclide because of its good sensitivity for measurement. Shapiro-Wilk test was performed to determine the normality of LIBS data, and it revealed that data from the homogeneous sample is normal distribution (p-value = 0.9246, if p-value is higher than 0.05, it is considered as normal distribution). However, data from the heterogeneous sample showed abnormal distribution (p-value = 7.765×10-8). The coefficient of variation (CoV) was also calculated to examine the dispersion of data. It was 31.3% and 51.8% from homogeneous and heterogeneous samples, respectively. These results suggest that LIBS analysis and statistical approaches can be used to evaluate the homogeneity of waste forms for the acceptance criterion in repositories.

      • Effects of the aryl hydrocarbon receptor and autophagy on skin barrier protein expression

        Background: Autophagy and the aryl hydrocarbon receptor (AHR) have been reported as important players in the development and differentiation of the skin barrier. Objectives: We aimed to evaluate the effects of autophagy and AHR on epidermal barrier protein expression and crosstalk between them. Methods: We stimulated HaCaT cells and psoriasis skin tissue with 2,3,7,8- tetrachlorodibenzo-p-dioxin (TCDD), chloroquine (CQ), rapamycin, and a siRNA treatment before performing quantitative reverse transcriptase polymerase chain reaction and western blot for epidermal barrier genes and proteins. A proximity ligation assay was employed to explore the effects on the relationship between AHR and LC3B in HaCaT cells and normal human epidermal keratinocytes (NHEKs). Results: TCDD increased the expression of S100A7 and S100A8. In contrast, AHR knockdown downregulated S100A7 and S100A8 expression. Autophagy inhibition by CQ suppressed filaggrin, hornerin, involucrin, and keratin 10 expression, whereas autophagy induction by rapamycin upregulated these proteins. CQ led to a significant increase in AHR-LC3 interaction and AHR promoter activity. TCDD-induced effects on epidermal protein were repressed by CQ, whereas CQ increased AHR expression and the phosphorylation of p38 and p65. Conclusion: Autophagy inhibition suppressed the TCDD-induced upregulation of epidermal barrier proteins via AHR/ p38MAPK/ p65NF-kB signaling. Our findings provide new insights into the crosstalk between autophagy and AHR signaling.
