Conjugated linoleic acid (CLA), when incorporated into mouse liver microsomal membranes, selectively inhibits the mutagenesis of 2-amino-3-methylimidazo[4,5-f]quinoline (IQ). Nine-week old female ICR mice were given (p.o.) 0.1 mL olive oil alone (control), 0.1 mL olive oil plus 0.1 mL linoleic acid, or 0.1 mL olive oil plus 0.1 mL CLA, twice weekly for four weeks. The animals were then sacrificed and liver S-9 fractions were prepared. Activation of IQ for mutagenesis by the liver S-9 from CLA-treated mice was significantly reduced in comparison with liver S-9 from control or linoleic acid-treated mice. By contrast, the activation of 7,12-dimethylbenz[a]anthracene (DMBA) and benzo[a]pyrene (BP) was unaffected. Hence, CLA incorporated into phospholipids may selectively affect cytochrome P450 isozymes responsible for activating IQ, but not those which activate BP or DMBA. The addition of free CLA or the methyl esters of CLA, linoleic acid, or oleic acid, to control S-9 inhibited the activation of all three mutagens (IQ, BP, and DMBA).
Directing the Distribution of Potassium Cations in Zeolite-LTL through Crown Ether Addition

<P>We discover that the crystal morphology of zeolite-LTL could be modified by crown ether (21-crown-7, CE), where CE decreases the aspect ratio of zeolite-LTL while increasing the nucleation of domains on the (0001) face and hindering their growth along the c-axes. Moreover, the study using scanning electron microscopy supports that the ratio between the rates for generation of cancrinite columns and bridging cancrinite columns on the {10 (1) over bar0} face remains constant among the LTL frameworks with different amounts of CE molecules. In addition, X-ray diffraction analysis shows that potassium cations redistribute into pore cavities (t-lil) from cancrinite cages (t-can) and t-ste cages by the strong interactions between potassium and CE as the amount of CE molecules is increased. Additionally, Monte Carlo simulations clarify that stabilization of the t-lil cage via the redistribution of potassium cations at high CE concentration is attributed to the dominant effect in the crystal morphology changes observed. To understand the catalytic and adsorption properties of zeolites, it is important to investigate their structure/property relationships. Especially, studying the morphology of an anisotropic zeolite crystals has been of great interest because of the strong influence on controlling its properties. Thus, morphological control of the material with a particular crystallographic direction is highly desirable to obtain maximum properties for applications.</P>

<B>ABSTRACT</B><P>Induction of mucosal immunity may be important for preventing SARS-CoV infections. For safe and effective delivery of viral antigens to the mucosal immune system, we have developed a novel surface antigen display system for lactic acid bacteria using the poly-γ-glutamic acid synthetase A protein (PgsA) of <I>Bacillus subtilis</I> as an anchoring matrix. Recombinant fusion proteins comprised of PgsA and the Spike (S) protein segments SA (residues 2 to 114) and SB (residues 264 to 596) were stably expressed in <I>Lactobacillus casei</I>. Surface localization of the fusion protein was verified by cellular fractionation analyses, immunofluorescence microscopy, and flow cytometry. Oral and nasal inoculations of recombinant <I>L. casei</I> into mice resulted in high levels of serum immunoglobulin G (IgG) and mucosal IgA, as demonstrated by enzyme-linked immunosorbent assays using S protein peptides. More importantly, these antibodies exhibited potent neutralizing activities against severe acute respiratory syndrome (SARS) pseudoviruses. Orally immunized mice mounted a greater neutralizing-antibody response than those immunized intranasally. Three new neutralizing epitopes were identified on the S protein using a peptide neutralization interference assay (residues 291 to 308, 520 to 529, and 564 to 581). These results indicate that mucosal immunization with recombinant <I>L. casei</I> expressing SARS-associated coronavirus S protein on its surface provides an effective means for eliciting protective immune response against the virus.</P>
