        Melatonin Attenuates Noise Stress-induced Gastrointestinal Motility Disorder and Gastric Stress Ulcer: Role of Gastrointestinal Hormones and Oxidative Stress in Rats

        Background/Aims There are increasing evidences for gastrointestinal motility disorder (GIMD) and gastric stress ulcer induced by noise stress. The present study was to investigate the reversed effect of melatonin on GIMD and gastric stress ulcer induced by noise stress and potential mechanism. Methods Noise stress was induced on rats, and melatonin (15 mg/kg) was administered to rats by intraperitoneal injection. Differences were assessed in gastric residual rate (GRR), small intestine propulsion rate (SPR), Guth injury score, cortisol, gastrointestinal hormones (calcitonin-gene-related peptide and motilin) and oxidative stress markers (superoxide dismutase and malondialde hyde) in blood plasma as well as gastric mucosa homogenate with or without melatonin. The pathological examination of gastric mucosa was also performed. Results The GRR and SPR were improved by noise stress compared with control (P < 0.05). The pathological examination and Guth injury score revealed gastric stress ulcer. Moreover, the levels of cortisol, motilin and malondialdehyde in blood plasma and malondialdehyde in gastric mucosa homogenate were increased by noise stress (P < 0.05). CGRP and superoxide dismutase activity in both of blood plasma and gastric mucosa homogenate were significantly decreased (P< 0.05). Furthermore, melatonin reversed changes in GRR, SPR, pathological examination, Guth injury score, cortisol, motilin, CGRP, superoxide dismutase activity and malondialdehyde (P < 0.05). Conclusions Melatonin is effective in reversing the GIMD and gastric stress ulcer induced by noise stress. The underlying mechanism may be involved in oxidative stress and gastrointestinal hormones.       • Advanced features of whole body sectioned images: Virtual Chinese Human

        <P>Serially sectioned images of whole cadavers have become available through the work of four projects: the Visible Human Project, the Visible Korean, the Chinese Visible Human, and the Virtual Chinese Human (VCH). For the VCH, new techniques and equipment were developed and applied to two female and two male cadavers to overcome some of the limitations noted in previous studies. In this article on the VCH, the procedures described are those used on a male cadaver. The cadaver was young with little to no pathology; there were no flat back artifacts because the cadaver was frozen and embedded in the upright position. Sectioned images (intervals, 0.2 mm) were of exceptional quality and resolution (0.1 mm-sized pixels). Several specific structures were outlined (intervals, 0.6 mm) to acquire segmented images, from which surface models were constructed. The VCH data are to be distributed worldwide and are expected to encourage other investigators to produce useful three-dimensional images and develop interactive simulation programs for clinical practice.         Multi-path Routing Policy for Content Distribution in Content Network

        Content distribution technology, which routes content to the cache servers, is considered as an effective method to reduce the response time of the user requests. However, due to the exponential increases of content traffic, traditional content routing methods suffer from high delay and consequent inefficient delivery. In this paper, a content selection policy is proposed, which combines the histories of cache hit and cache hit rate to collaboratively determine the content popularity. Specifically, the CGM policy promotes the probability of possible superior paths considering the storage cost and transmission cost of content network. Then, the content routing table is updated with the proportion of the distribution on the paths. Extensive simulation results show that our proposed scheme improves the content routing and outperforms existing routing schemes in terms of Internet traffic and access latency.

        Research on Preparation of Ni/SiO2 Optical Attenuation Slice by Magnetron Sputtering

        In order to find new methods for preparing and improving the performance of optical attenuation slice. Vacuum magnetron sputtering method was used to prepare Ni/SiO2 composite film optical attenuation slices, with vacuum magnetron sputtering apparatus, at 5,10,15,20,25min sputtering time, 0.2,0.4,0.6,0.8,1.0Pa sputtering pressure and sputtering power 300~1200W. The XRD, SEM, EDS, AFM and 722 spectrophotometer also were used to study the effects of different sputtering time, sputtering pressure and sputtering power on the film structure, surface morphology, composition, three-dimensional structure, surface roughness and light attenuation rate of optical attenuation slice samples. The results indicated that: Ni/SiO2 composite films were formed, instead of simple physical adsorption between Ni film and SiO2 substrates with magnetron sputtering by XRD analysis and calculation, with uniform grain size of 25.96, 32.38, 32.29,26.95, 25.92nm, respectively. The main component element was Ni, but there were few impurities deposited on the substrates. Impurities were reduced gradually with the increase of sputtering time, mainly resulted from two sputtering of Ni atoms by EDS; The film surface was smooth and dense, flatness and organizational structure of Ni film were better, surface roughness was 1.267nm at 25min sputtering time, 0.4Pa pressure and 400W power with SEM and AFM. Light attenuation rate of optical attenuation slices was different in different process parameters, and the maximum reached 0.52, film flatness, impurities and defects were the main reason by the light attenuation performance analysis.

        The Pattern of Time to Onset and Resolution of Immune-Related Adverse Events Caused by Immune Checkpoint Inhibitors in Cancer: A Pooled Analysis of 23 Clinical Trials and 8,436 Patients

        Purpose The occurrence pattern of immune-related adverse events (irAEs) induced by immune checkpoint inhibitor (ICI) in cancer treatment remains unclear. Materials and Methods Phase II-III clinical trials that evaluated ICI-based treatments in cancer and were published between January 2007 and December 2019 were retrieved from public electronic databases. The pooled median time to onset (PMT-O), resolution (PMT-R), and immune-modulation resolution (PMT-IMR) of irAEs were generated using the metamedian package of R software.Results Twenty-two eligible studies involving 23 clinical trials and 8,436 patients were included. The PMT-O of all-grade irAEs ranged from 2.2 to 14.8 weeks, with the longest in renal events. The PMT-O of grade ≥ 3 irAEs was significantly longer than that of all-grade irAEs induced by programmed cell death protein 1 (PD-1) and its ligand 1 (PD-L1) inhibitors (27.5 weeks vs. 8.4 weeks, p < 0.001) and treatment of nivolumab (NIV) plus ipilimumab (IPI) (7.9 weeks vs. 6.0 weeks, p < 0.001). The PMT-R of all-grade irAEs ranged from 0.1 to 54.3 weeks, with the shortest and longest in hypersensitivity/infusion reaction and endocrine events, respectively. The PMT-IMR of grade ≥ 3 irAEs was significantly shorter than that of all-grade irAEs caused by PD-1/PD-L1 blockade (6.9 weeks vs. 40.6 weeks, p=0.002) and NIV+IPI treatment (3.1 weeks vs. 5.9 weeks, p=0.031).Conclusion This study revealed the general and specific occurrence pattern of ICI-induced irAEs in pan-cancers, which was deemed to aid the comprehensive understanding, timely detection, and effective management of ICI-induced irAEs.

        Mesenchymal Stem Cells Modified with Stromal Cell-Derived Factor 1${\alpha}$ Improve Cardiac Remodeling via Paracrine Activation of Hepatocyte Growth Factor in a Rat Model of Myocardial Infarction

        Mesenchymal stem cells (MSCs) are a promising source for cell-based treatment of myocardial infarction (MI), but existing strategies are restricted by low cell survival and engraftment. We examined whether SDF-1 transfection improve MSC viability and paracrine action in infarcted hearts. We found SDF-1-modified MSCs effectively expressed SDF-1 for at least 21days after exposure to hypoxia. The apoptosis of Ad-SDF-1-MSCs was 42% of that seen in Ad-EGFP-MSCs and 53% of untreated MSCs. In the infarcted hearts, the number of DAPI-labeling cells in the Ad-SDF-1-MSC group was 5-fold that in the Ad-EGFP-MSC group. Importantly, expression of antifibrotic factor, HGF, was detected in cultured MSCs, and HGF expression levels were higher in Ad-SDF-MSC-treated hearts, compared with Ad-EGFP-MSC or control hearts. Compared with the control group, Ad-SDF-MSC transplantation significantly decreased the expression of collagens I and III and matrix metalloproteinase 2 and 9, but heart function was improved in d-SDF-MSC-treated animals. In conclusion, SDF-1-modified MSCs enhanced the tolerance of engrafted MSCs to hypoxic injury in vitro and improved their viability in infarcted hearts, thus helping preserve the contractile function and attenuate left ventricle (LV) remodeling, and this may be at least partly mediated by enhanced paracrine signaling from MSCs via antifibrotic factors such as HGF.

        Objective Quantitation of EGFR Protein Levels using Quantitative Dot Blot Method for the Prognosis of Gastric Cancer Patients

        Purpose: An underlying factor for the failure of several clinical trials of anti-epidermal growth factor receptor (EGFR) therapies is the lack of an effective method to identify patients who overexpress EGFR protein. The quantitative dot blot method (QDB) was used to measure EGFR protein levels objectively, absolutely, and quantitatively. Its feasibility was evaluated for the prognosis of overall survival (OS) of patients with gastric cancer. Materials and Methods: Slices of 2×5 μm from formalin-fixed paraffin-embedded gastric cancer specimens were used to extract total tissue lysates for QDB measurement. Absolutely quantitated EGFR protein levels were used for the Kaplan-Meier OS analysis. Results: EGFR protein levels ranged from 0 to 772.6 pmol/g (n=246) for all gastric cancer patients. A poor correlation was observed between quantitated EGFR levels and immunohistochemistry scores with ρ=0.024 and P=0.717 in Spearman's correlation analysis. EGFR was identified as an independent negative prognostic biomarker for gastric cancer patients only through absolute quantitation, with a hazard ratio of 1.92 (95% confidence interval, 1.05–3.53; P=0.034) in multivariate Cox regression OS analysis. A cutoff of 208 pmol/g was proposed to stratify patients with a 3-year survival probability of 44% for patients with EGFR levels above the cutoff versus 68% for those below the cutoff based on Kaplan-Meier OS analysis (log rank test, P=0.002). Conclusions: A QDB-based assay was developed for gastric cancer specimens to measure EGFR protein levels absolutely, quantitatively, and objectively. This assay should facilitate clinical trials aimed at evaluation of anti-EGFR therapies retrospectively and prospectively for gastric cancer.

        Analysis on an improved resistance tuning type multi-frequency piezoelectric spherical transducer

        The existing piezoelectric spherical transducers with fixed prescribed dynamic characteristics limit their application in scenarios with multi-frequency or frequency variation requirement. To address this issue, this work proposes an improved design of piezoelectric spherical transducers using the resistance tuning method. Two piezoceramic shells are the functional elements with one for actuation and the other for tuning through the variation of load resistance. The theoretical model of the proposed design is given based on our previous work. The effects of the resistance, the middle surface radius and the thickness of the epoxy adhesive layer on the dynamic characteristics of the transducer are explored by numerical analysis. The numerical results show that the multi-frequency characteristics of the transducer can be obtained by tuning the resistance, and its electromechanical coupling coefficient can be optimized by a matching resistance. The proposed design and derived theoretical solution are validated by comparing with the literature given special examples as well as an experimental study. The present study demonstrates the feasibility of using the proposed design to realize the multi-frequency characteristics, which is helpful to improve the performance of piezoelectric spherical transducers used in underwater acoustic detection, hydrophones, and the spherical smart aggregate (SSA) used in civil structural health monitoring, enhancing their operation at the multiple working frequencies to meet different application requirements.
