Heme oxygenase-1 induced by desoxo-narchinol-A attenuated the severity of acute pancreatitis via blockade of neutrophil infiltration

<P><B>Abstract</B></P> <P>Heme oxygenase-1 (HO-1) has an anti-inflammatory action in acute pancreatitis (AP). However, its mechanism of action and natural compounds/drugs to induce HO-1 in pancreas are not well understood. In this study, we investigated the regulatory mechanisms of HO-1 during AP using desoxo-narchinol-A (DN), the natural compound inducing HO-1 in the pancreas. Female C57/BL6 Mice were intraperitoneally injected with supramaximal concentrations of cerulein (50 μg/kg) hourly for 6 h to induce AP. DMSO or DN was administered intraperitoneally, then mice were sacrificed 6 h after the final cerulein injection. Administration of DN increased pancreatic HO-1 expression through activation of activating protein-1, mediated by mitogen-activated protein kinases. Furthermore, DN treatment reduced the pancreatic weight-to-body weight ratio as well as production of digestive enzymes and pro-inflammatory cytokines. Inhibition of HO-1 by tin protoporphyrin IX abolished the protective effects of DN on pancreatic damage. Additionally, DN treatment inhibited neutrophil infiltration into the pancreas via regulation of chemokine (C-X-C motif) ligand 2 (CXCL2) by HO-1. Effects of 17β-estradiol on colorectal cancer development after azoxymethane/dextran sulfate sodium treatment of ovariectomized mice

<P><B>Abstract</B></P> <P>Estrogen is known to have a protective effect in colorectal cancer (CRC) development. Previously, we reported the anti-inflammatory and antitumorigenic effects of 17β-estradiol (E2) in azoxymethane (AOM)/dextran sulfate sodium (DSS)-treated male mice. The aim of this study was to investigate whether ovariectomy in a female AOM/DSS mouse model increases colorectal tumorigenesis and whether tumorigenesis is reduced by estrogen supplementation after ovariectomy. Clinical symptoms and histological severity of colitis and the levels of inflammatory mediators were evaluated in the colon of AOM/DSS-treated ovariectomized (OVX) mice. The levels of E2, myeloperoxidase (MPO), and NF-κB-dependent cytokines (interleukin (IL)-1β and IL-6) were measured by ELISA. Furthermore, quantitative real-time (qRT) PCR and Western blot analysis were performed. Ovariectomy did not aggravate AOM/DSS-induced colitis at 2 weeks. At weeks 10 and 16, ovariectomy significantly increased tumor number and incidence rate in only the proximal colon after AOM/DSS treatment (F_AOM/DSS vs OVX_AOM/DSS), and these increases were significantly reduced by E2 supplementation (OVX_AOM/DSS vs OVX_AOM/DSS/E2). However, ovariectomy did not affect CRC development in the distal colon (F_AOM/DSS vs OVX_AOM/DSS). At week 2, E2 administration to AOM/DSS-treated OVX mice attenuated the histological severity of colitis by decreasing the protein and/or mRNA levels of estrogen receptor alpha (ERα) and NF-κB-related mediators (i.e., COX-2, TNF-α, and IL-6) and by enhancing estrogen receptor beta (ERβ) and nuclear Nrf2 protein expression and the mRNA expression of related antioxidant enzyme genes (i.e., HO-1, GCLC, GCLM, and NQO1). Endogenous estrogen in females protects against the development of proximal colon cancer, and exogenous E2 replacement in OVX female mice showed protective effects against AOM/DSS-induced colitis and carcinogenesis. This study was investigated the inhibitory activity of RADIX ASARI (RA) on type I immediate hypersensitivity of the anaphylactic type. RA inhibited compound 48/80-induced anaphylaxis 100% with a dose of 0.5 g/㎏ body weigh at 1 h before or 5 and 10 min after injection of compound 48/80. RA also exhibited the dose-dependent inhibitory effect on compound 48/80-induced, Substance P-induced and anti-DNP IgE-induced histamine release from rat peritoneal mast cells. These results suggest the possibility that RA induced inhibition of type I allergic reaction is regulated by histamine.

To evaluate the in-vivo effects of mitomycin-C-polyglycolic acid composite(MMC-PGA) the disk: shaped composite were implanted into the peritoneal cavity of the Sprague-Dawley rats. The pathologic changes were examined at various time points up to 12 weeks. Initially the soft tissue around the inserted disks showed capillary congestion at 3 days. Inflammatory infiltrates with foreign body giant cells appeared from the 2nd week and reached peak response at 6-8 weeks. These reaction diminished prominently at 12 weeks. No specific pathologic change was found in the liver, the kidneys, and the heart. The above results suggest that the MMC-PGA composite can serve as a new device for intraperitoneal chemotherapy of various types of cancers.

This paper proposes a neural network approach to solve the hand-written digit recognition problem. The proposed approach is based on the graph matching, a form of elastic matching, and a mean field annealing algorithm. In general, a conventional recognition model uses a preprocessor to remove meaningless variations and to capture meaningful features. But this approach can recognize digits without any preprocessing such as thinning, scaling and segmentation.

We studies inhibitation effect of IgE release in the human B cell line(U266B1) and in the serum and spleen cell of mouse that has been immunized by antigen in order to find antiallergic effect of Fructus Xanthii (FX). We have some findings as below. 1. IgE release in serum was inhibited dose dependently in case of administering FX orally in the mouse immunized. 2. IgE release in serum was inhibited dose dependently in case of injecting FX into abdomen in the mouse immunized. 3. When we take LPS+IL-4 way in the mouse spleen cell, IgE release was less than LPS way. 4. When we take LPS+IL-4 way in the human B cell line (U266B1), IgE release was less than LPS way.

Glucose Oxidase가 척수지각신경절세포에 미치는 영향과 천마의 효과에 관한 연구

Objectives : To clarify the cytotoxic effect of glucose oxidase(GO) and protective effect of gastrodiae Rhizoma (GR) on spinal sensory ganglion(DRG) neurons, neurotoxicity mediated by GO was measured by MTT assay and neurofilament enzymeimmuno assay(EIA). Methods : DRG neurons were cultured in the media containing various concentrations of GO for 3 hours. In addition, neuroprotective effects of GR, on GO-induced neurotoxicity in DRG neurons were examined after DRG neurons were preincubated with various concentrations of GR for 2 hours before 15mU/ml GO for 3 hours. Results : GO decreased remarkably cell viability in dependently in these cultures, and also RG increased cell viability and amount of neurofilament in DRG neurons damaged by GO. Conclusion : It is suggested that GO has toxic effect in cultured mouse DRG neurons, and also RG was effective in the protection of GO-induced neurotoxicity in these cultures.

정사각형 용기의 Deep Drawing 성형성 실험을 다양한 작업조건하에서 수행하였다. 본 연구에서는 재료의 유동상태와 판두께의 국부적 변화량을 성형성의 척도로 사용하였으며, Photo-etching 방법을 이용하여 이들을 측정하였다. Draw Bead의 크기, 블랭크 홀더력의 크기 및 윤활상태의 변화에 따른 정사각형 용기의 둘레방향, 대각선방향 및 반경방향으로의 판두께 변화량과 재료의 유동상태를 실험적으로 분석하였다. Experiments on the drawability of squared boxes have been performed under various working conditions. In the present work, the pattern of material flow and the local change in the thickness of shell are used as the measures of drawabilitiy and they are measured using the photo-etching technique. The size of draw bead, the magnitude of blank holding force and the condition of lubrication have been changed and their effects on the pattern of material flow and the thickness of shell along the circumferential, diagonal and radial directions of the squared boxes have been analyzed experimentally.
