      • Quality Characteristics of Jeju Beets Treated by Various Drying Conditions

        Beets (Beta vulgaris) contain a lot of betalains, a type of natural pigment, so they are known to be red and have strong antioxidant abilities. This study was conducted to compare the physicochemical properties of Jeju beets processed by hot air drying and combined drying. Beets were sliced to 3 mm and dried by hot air drying (13 hours at 50°C, 6 hours at 60°C, or 4.5 hours at 70°C) or combined drying (hot air and infrared radiation). Appearance, water activity, moisture content, pH, color, rehydration ratio (RR), total polyphenol content (TPC), FRAP value, DPPH radical scavenging ability, and ABTS radical scavenging ability were determined. In hot air drying, as the drying temperature increased, water activity, moisture content, and b* value were decreased while L*, a* value, RR, TPC, FRAP value, and ABTS radical scavenging ability were increased. The combined drying had lower water activity and pH value; moreover, it had a higher L*, a* value, RR, TPC, FRAP value, DPPH radical scavenging ability, and ABTS radical scavenging ability. In this study, it was found that the combined drying was more suitable than hot air drying to maintain the antioxidant ability of beet.

        열풍 및 복합건조된 제주산 비트의 이화학적 특성과 항산화 활성

        본 연구에서는 기존 비트의 한계점을 극복하고 고품질의 건조 비트를 제조하기 위한 기초 자료를 제시하고자 제주산 비트를 다양한 온도 조건에서 열풍 및 적외선 복합건조 하였고, 이에 따른 품질변화를 비교하여 건조 조건이 비트의 이화학적 특성과 항산화 활성에 미치는 영향에 대해서 조사하였다. 3 ㎜ 두께로 비트를 절단한 후 70℃에서 4.5시간, 60℃에서 6시간, 50℃에서 13시간 열풍건조 하거나 100℃에서 1시간 복합건조 하였다. 열풍건조한 비트는 건조 온도가 높아질수록 수분함량, 수분활성도, b<SUP>*</SUP>값(황색도), betanin 함량이 감소했고 건조수율과 pH에는 변화가 없었으며, L<SUP>*</SUP> 값(명도), a<SUP>*</SUP> 값(적색도), ΔE 값(건조 전 원물과의 색차), 재복원율, vulgaxanthin-Ⅰ 함량, 총 폴리페놀 함량, DPPH 라디칼 소거능, ABTS⁺ 라디칼 소거능, FRAP 활성은 증가하는 경향을 보였다. 그중에서 수분활성도, a<SUP>*</SUP>값, ΔE 값, FRAP 활성은 건조 온도가 높아짐에 따라 유의적으로 변화하였다. 복합건조한 비트는 열풍건조한 비트보다 L<SUP>*</SUP>값, a<SUP>*</SUP> 값, ΔE 값, 재복원율, 총 폴리페놀 함량, DPPH 라디칼 소거능, ABTS⁺ 라디칼 소거능, FRAP 활성이 유의적으로 높았고 pH는 유의적으로 낮았으며, 수분함량, 수분활성도, b<SUP>*</SUP>값, betanin 함량, vulgaxanthin-Ⅰ 함량에서는 열풍건조한 비트와 유의적인 차이를 나타내지 않았다. 이상의 결과에 따르면 복합건조한 비트가 다른 조건에서 건조를 진행한 비트에 비해 소요되는 시간이 매우 짧고 pH가 낮으며, 재복원율, 총 폴리페놀 함량, 항산화 활성이 높아 고품질의 건조 비트를 제조하기 위해서는 열풍뿐만이 아니라 적외선도 적절히 병용하는 것이 적합할 것으로 보인다. Beet (Beta vulgaris L.) is a healthy food that has recently attracted the consumer’s attention. However, beet has low storage stability due to the high moisture content. Therefore, a drying process is necessary to improve the beet quality. In this study, Jeju beet was dried by applying two processes: hot air drying (4.5 hours at 70℃, 6 hours at 60℃, 13 hours at 50℃) and combined drying (hot air and infrared radiation, 1 hour at 100℃). The effect of both processes was investigated on the physicochemical properties and antioxidant activity was investigated. The dried beets were compared for their appearance, drying yield, moisture content, water activity, pH value, color, rehydration ratio, betalain pigment content, total polyphenol content, DPPH radical scavenging activity, ABTS⁺ radical scavenging activity, and FRAP assay. The beet subjected to combined drying was determined to have high storage properties due to its low moisture content and pH value. In addition, the combined dried beet showed the highest rehydration ratio and antioxidant activity. Taken together, our results indicate that an appropriate combination of hot air and infrared radiation is suitable to produce high-quality dried beet.

      • Matrix Metalloproteinase-8 Plays a Pivotal Role in Neuroinflammation by Modulating TNF-α Activation

        <P>Matrix metalloproteinases (MMPs) play important roles in normal brain development and synaptic plasticity, although aberrant expression of MMPs leads to brain damage, including blood–brain barrier disruption, inflammation, demyelination, and neuronal cell death. In this article, we report that MMP-8 is upregulated in LPS-stimulated BV2 microglial cells and primary cultured microglia, and treatment of MMP-8 inhibitor (M8I) or MMP-8 short hairpin RNA suppresses proinflammatory molecules, particularly TNF-α secretion. Subsequent experiments showed that MMP-8 exhibits TNF-α–converting enzyme (TACE) activity by cleaving the prodomain of TNF-α (A<SUP>74</SUP>/Q<SUP>75</SUP>, A<SUP>76</SUP>/V<SUP>77</SUP> residues) and, furthermore, that M8I inhibits TACE activity more efficiently than TAPI-0, a general TACE inhibitor. Biochemical analysis of the underlying anti-inflammatory mechanisms of M8I revealed that it inhibits MAPK phosphorylation, NF-κB/AP-1 activity, and reactive oxygen species production. Further support for the proinflammatory role of microglial MMP-8 was obtained from an in vivo animal model of neuroinflammatory disorder. MMP-8 is upregulated in septic conditions, particularly in microglia. Administration of M8I or MMP-8 short hairpin RNA significantly inhibits microglial activation and expression/secretion of TNF-α in brain tissue, serum, and cerebrospinal fluid of LPS-induced septic mice. These results demonstrate that MMP-8 critically mediates microglial activation by modulating TNF-α activity, which may explain neuroinflammation in septic mouse brain.</P>

        Manifestation and treatment in a cleidocranial dysplasia patient with a RUNX2 (T420I) mutation

        Cleidocranial dysplasia is an autosomal dominant heritable skeletal disorder. The characteristic features of cleidocranial dysplasia (CCD) may include hypoplasia of the clavicle, delayed closure of frontanelles, late tooth eruption, and other skeletal disorders. This case report describes clinical and radiographic manifestations at the age of 11 and 29 of a CCD patient, investigates the mutation of core-binding factor A1 (CBFA1) based on gene analysis, and illustrates successful oral reconstruction with fixed prosthesis and dental implant after the extraction of multiple teeth.

        Original Article : Evaluation of the Biological Activities of Marine Bacteria Collected from Jeju Island, Korea, and Isolation of Active Compounds from their Secondary Metabolites

        To explore marine microorganisms with medical potential, we isolated and identified marine bacteria from floats, marine algae, animals, and sponges collected from Jeju Island, Korea. We isolated and identified 21 different strains from the marine samples by 16S rRNA analysis, cultured them in marine broth, and extracted them with ethyl acetate (EtOAc) to collect secondary metabolite fractions. Next, we evaluated their anti-oxidative and anti-inflammatory effects. Among the 21 strains, the secondary metabolite fraction of Bacillus badius had both strong antioxidant and anti-inflammatory activity, and thus was selected for further experiments. An antioxidant compound detected from the secondary metabolite fraction of B. badius was purified by preparative centrifugal partition chromatography (n-hexane:EtOAc:methanol:water, 4:6:4:6, v/v), and identified as diolmycin A2. Additionally, diolmycin A2 strongly inhibited nitric oxide production. Thus, we successfully identified a significant bioactive compound from B. badius among the bacterial strains collected from Jeju Island.

      • 청각 이미지와 의복 이미지의 관련성에 대한 탐색 :성격, 성별,의복관심의 영향을 중심으로

        This study exploratory examined the relationship between auditory images and clothing style images of young consumers by their personality types, sex and clothing interest. A questionnaire was developed to measure auditory images, clothing style images, personality types, clothing interest and demographic variables. Scale items were adopted from prior studies on clothing self images and Myers-Briggs Type Indicator (MBTI) known to measure psychological types. 120 undergraduate students with diverse majors were the respondents of the empirical study. MBTI categorized respondents into one of 3 personality types, IFT (Introversion and Thinking or Feeling, n=23), EF (Extroversion and Feeling, n=73) and ET (Extroversion and Thinking, n=24). ANOVA results indicated that, among 13 auditory images measured in the study, rational, self-confident and social images were significantly different across three personality type groups. Overall, ET group showed higher level of the the auditory images. Among clothing style images, social images were significantly different. IET group showed significantly lower level of social images. In addition, respodents' age and level of clothing interest influenced auditory and clothing style images. Correlation coefficients indicated that there were significant relationships between most auditory and clothing style images included in the study except for the self-confident image.

        열처리 및 해동방법에 따른 냉동 감자와 당근의 품질 변화

        본 연구에서는 냉동 감자와 당근의 전처리 및 해동 방법에 따른 변화를 관찰하기 위해 hot water blanching 및 steam blanching 처리한 것과 전처리하지 않은 대조구를 개별 급속냉동 하였고, 이후 상온해동, 유수해동, 전자레인지 해동을 진행하여 이화학적, 물리적, 영양학적 변화를 분석하였다. 해동속도는 상온해동, 유수해동, 전자레인지 해동 순으로 빨라졌으며, 전자레인지 해동 방법이 약 24~70배 정도 빠르게 해동되었다. Blanching 처리구의 경우 낮은 해동감량을 나타내었으며 이에 따라 수분함량이 증가하는 경향을 나타내었다. 감자의 색도는 가열 전처리하였을 때 대조구에 비하여 L<SUP>*</SUP>값과 a<SUP>*</SUP> 값이 유의적으로 차이를 나타내어 blanching 처리가 감자의 갈변현상을 억제하는 것으로 판단된다. 조직감은 전처리의 영향을 받아 blanching 처리를 함에 따라 조직이 연화되어 경도가 감소하는 경향을 나타내었다. pH와 총 페놀 함량은 전처리와 해동 공정에 따른 유의적 차이가 나타나지 않았다. DPPH 라디칼 소거능과 비타민 C함량은 steam blanching 처리했을 때 모든 해동 방법에서 높은 수치를 나타냈기 때문에 해동 후 감자와 당근의 항산화 효과를 높게 유지하기 위해서는 steam blanching 처리가 적절할 것으로 판단된다. 이상의 결과를 종합했을 때 감자와 당근의 해동 후 품질 변화는 해동 방법보다 blanching 처리에 따른 것으로 추측되며, 냉동 전 blanching 처리는 해동 시 발생하는 품질 변화를 억제할 수 있고, 특히 steam blanching 처리 후 유수해동 하는 것이 영양성분을 높게 유지해 주는 것으로 판단된다. This study investigates the effects of various blanching methods (hot water blanching (HB) or steam blanching (SB)) and thawing methods (room temperature thawing (RT), running water thawing (WT), and microwave thawing (MT)) on the quality attributes of frozen potatoes and carrots. Blanched potatoes and carrots were frozen using individual quick freezing (−35℃), followed by applying various thawing methods. We observed increased thawing speed in the order of RT, WT, and MT. The MT method was about 24 to 70 times shorter than the other methods. The thawing losses were influenced by the blanching methods which affect the drip loss and moisture contents of potatoes and carrots. The blanching process reduced the browning of potatoes but did not affect color of the carrots. No change was obtained in the pH value when considering different blanching or thawing methods. The hardness of potatoes and carrots tended to be lowered by blanching treatment. SB treated potatoes and carrots had high DPPH radical scavenging ability and vitamin C content. Especially, the WT treated potatoes and carrots had the highest total polyphenol content. Our results confirmed that SB is an excellent pre-treatment for potatoes and carrots from the nutritional aspect, and blanching pre-treatment before the freezing process could suppress the quality changes that occur during the thawing processes. Furthermore, we propose that RT after SB treatment for potatoes and carrots is probably the most suitable method to maintain their nutritional quality.
