<P>Copper oxide nanoparticles (CuONPs), metal oxide nanoparticles were used in multiple applications including wood preservation, antimicrobial textiles, catalysts for carbon monoxide oxidation and heat transfer fluid in machines. We investigated the effects of CuONPs on the respiratory system in Balb/c mice. In addition, to investigate the effects of CuONPs on asthma development, we used a murine model of ovalbumin (OVA)-induced asthma. CuONPs markedly increased airway hyper-responsiveness (AHR), inflammatory cell counts, proinflammatory cytokines and reactive oxygen species (ROS). CuONPs induced airway inflammation and mucus secretion with increases in phosphorylation of the MAPKs (Erk, JNK and p38). In the OVA-induced asthma model, CuONPs aggravated the increased AHR, inflammatory cell count, proinflammatory cytokines, ROS and immunoglobulin E induced by OVA exposure. In addition, CuONPs markedly increased inflammatory cell infiltration into the lung and mucus secretions, and MAPK phosphorylation was elevated compared to OVA-induced asthmatic mice. Taken together, CuONPs exhibited toxicity on the respiratory system, which was associated with the MAPK phosphorylation. In addition, CuONPs exposure aggravated the development of asthma. We conclude that CuONPs exposure has a potential toxicity in humans with respiratory disease.</P>

급성 전골수구성 백혈병의 변이형과 감별이 어려운 골수성/Natural Killer 세포 급성 백혈병

연구배경: 골수성/NK 세포 급성 백혈병은 형태학적 및 면역표지자 검사에서 급성 전골수구성 백혈병의 미세과립성 변이형(M3v)과 유사하여 흔히 M3v로 오진할 가능성이 높은 새로운 백혈병 아형 중의 하나로 알려져 있다. 이에 저자들은 M3와 감별이 어려웠던 골수성/NK 세포급성 백혈병의 임상적 및 혈액학적 특징을 분석하여 보았다. 방법: 전남대학교병원 혈액·종양내과에서 FAB 분류상 골수성/NK 세포 급성 백혈병에 해당되는 환자 3예를 대상으로 세포 형태학적 특성, 면역표지자검사. 염색체 핵형분석, FISH 분석 및 임상적인 특징 등을 후향적으로 알아보았다. 결과: 평균 연령은 37.7세(범위, 18-58) 였으며, 여자가 2예였다. 혈액응고검사는 모두 정상이었고, 1예에서 경막에 과립구성 육종이 동반되었다. 형태학적으로 골수모구는 깊이 함몰된 핵막과 미세한 아주르친화성 과립을 보이면서 소량의 세포질을 보였고, myeloperoxidase와 sudanblack-B에 강양성을 보였다. 면역표지자 검사에서 CD13+, CD56+, HLA-DR-, CD14-, CD19-,을 보였다. 염색체 핵형 분석상 2예에서 t(8;21)에 양성을 보였으며, t(15;17)는 모두 음성이었고, PML-RARα 는 모두 음성이엇다. 3예중 2예에서 완전 관해에 도달하였으며, 이들 중 1예는 12개월만에 재발하였다. 결론: 골수성/NK 세포급성 백혈병은 M3v와는 달리 출혈성 경향이 동반되니 않고, 면역표지자 검사에서 CD56+을 보이며, t(15;17) 및 PML-RARα는 음성을 보이기 때문에 감별 진단이 가능하였다. 향후 이러한 NK 세포 아형들의 명확한 정의와 임상적인 의의가 규명되어야 할것으로 사료된다. Background: The myeloid/Natural Killer (NK) cell acute leukemia is a unique subtype of acute leukemia potentially misdiagnosed as the microgranular variant of acute promyelocytic leukemia (M3v) by morphologic and immunophenotypic finding. In this study, we have conducted a retrospective analysis to evaluate clinical and hematological characteristics of myeloid/NK cellacute leukemia patients who were diagnosed in Chonnam University Hospital. Methods: Three patients with myeloid/NK cell acute leukemia were reviewed by cytomotphology and cytochemical reactions, immunophenotype, cytogentic analysis, fluorescence in situ hydridization (FISH) analysis and clinical characteristics. Results: There was no evidence of disseminated intravascular coagulopathy in there patients. One patient had granulocytic sarcoma in meninges. The blasts on marrow had deply invaginated nuclear membranes and scanty cytoplasms with fine azurophilic granules and high positivity for myeloperoxidase and sudan black-B. Immumophenotypic analysis showed HLA-DR, CD13+, CD56+, CD2-, CD5-, and CD19-. In chromosomal analyses, 2 cases had t (8;21) and 1 case had normal karyotype. However, all patients did not show both t (15;17) and PML-RARα gene rearrangement. Of 3 patients, 2 patients achieved complete remission and one patient relapsed after 12 months. Conclusion: Although the morphology of myeloid/NK cell acute leukemia were similar to that of M3v, myeloid/NK cell acute leukemia showed distinct clinical and hematological findings.

<P>Human adipose-derived stem cells (hADSCs) can differentiate into various cell types depending on chemical and topographical cues. One topographical cue recently noted to be successful in inducing differentiation is the nanoengineered polystyrene surface containing nanopore array-patterned substrate (NP substrate), which is designed to mimic the nanoscale topographical features of the extracellular matrix. In this study, efficacies of NP and flat substrates in inducing neural differentiation of hADSCs were examined by comparing their substrate-cell adhesion rates, filopodia growth, nuclei elongation, and expression of neural-specific markers. The polystyrene nano Petri dishes containing NP substrates were fabricated by a nano injection molding process using a nickel electroformed nano-mold insert (Diameter: 200nm. Depth of pore: 500nm. Center-to-center distance: 500nm). Cytoskeleton and filopodia structures were observed by scanning electron microscopy and F-actin staining, while cell adhesion was tested by vinculin staining after 24 and 48h of seeding. Expression of neural specific markers was examined by real-time quantitative polymerase chain reaction and immunocytochemistry. Results showed that NP substrates lead to greater substrate-cell adhesion, filopodia growth, nuclei elongation, and expression of neural specific markers compared to flat substrates. These results not only show the advantages of NP substrates, but they also suggest that further study into cell-substrate interactions may yield great benefits for biomaterial engineering.</P>
Scanning Electron Microscopy of Thelazia callipaeda Railliet and Henry, 1910 in the Eye of a Dog

저자들은 1999년 5월에 경기도 남양주군 군부대에서 사육하고 있는 한 마리의 군견 오른쪽 안부위에서 7마리 (암놈 4마리, 수놈 3마리)의 동양안충을 검출하였다. 이 중 일부는 고정과 lactophenol 액으로 봉입한 후 광학현미경으로 관찰하였으며 일부는 glutaraldehyde액에 고정하여 주사전자현미경으로 관찰하였다. 충체의 암놈 및 수놈의 두부와 꼬리부위의 형태 특징 그리고 충체 표피의 각질, 암놈 음문의 위치 등을 관찰한 결과 동양안충으로 동정하였다. We isolated 7 oriental eye worms, Thelazia callipaeda, from right eye of a military dog that had been reared in the military base of Namyangju-Gun, Kyungki province in May, 1999. Some of them were observed with light microscope after fixation and mounting with lactophenol, the others with scanning electron microscope after fixation with glutaraldehyde. As the result, the morphological differences of head part of females and males, tail part of females and males, cuticular characteristics of the surface and location of vaginal opening, we identified of these worms with T. callipaeda.
