      • 조직배양에서 얻어진 쪽파의 유전적 변이체들이 보여주는 단백질과 Isozyme의 유형분석

        Some genetic variants of Amphihaploid, Hypoamphidiploid, Amphidiploid are regenerated from tissue culture in Allium wakegi. They were applied to investigate the effects of the chromosomal differences on the gene expression of some isozymes. The electrophoretic migration patterns of total proteins were different at the position of MW5×10^5 Dalton among the genetic variants. Three different isozymes were characterized from the migration patterns of isozymes in the discontinuous polyacrylamide gel electrophoresis. Alcohol dehydrogenase(ADH) appeared 3 bands in the gel electrophoresis corresponding to 3 isozymes, that is allozymes distributed as single isozyme in each individual. Isocitric acid dehydrogenase(ICDH) appeared 5 bands in the gel and corresponds to 5 isozymes as allozymes which distributed single isozyme in each individual. Phosphoglucoisomerase(PGI) appeared 8 bands in electrophoresis and to 8 isozymes which originated from 2 genes and distributed more than two isozumes in each individual. According to the isozume expression in the genetic variants of Allium wagegi, the amplification of chromosomal numbers can affect neither to increase the isozyme numbers and concentration nor to express heterogeneous types in the genetic variants.

        Trichloroethylen의 직업적 노출에 의한 독성간염 및 박탈성 피부염 1례

        28세 남자가 trichloroethylene을 이용한 세척 및 탈지작업에 종사한지 한 달만에 발열, 오한 및 홍반을 동반한 피부발진 등의 증상이 발현되었다. 황달을 동반한 간기능 부전의 소견을 보였으며 피부는 박탈되었다. 부신피질호르몬, 항히스타민제 및 간기능 보조제를 이용한 치료로 간기능 및 피부 병변은 호전되었다. 검사소견상 Ig E가 상승되었고 trichloroethylene에 의한 과민반응으로 추정되었다. Trichloroethylene을 사용하는 소규모 사업장에서의 노출억제 방안을 포함한 보건관리가 요구되며 특수건강진단 주기에 대한 변경이 필요하리라 사료된다. A 28-year-old male began working as a degreaser. The solvent used in the degreasing operation was trichloroethylene. Over the next month the man experienced fever, chills, and an erythematous skin rash and itching. At that time he had a marked elevation in his liver enzyme, with cholestasis. Over the next few days the rash persisted then peeled. There was an elevation of Ig E, and a positive patch test reaction to trichloroethylene. His dermatitis and hepatitis were considered to be mediated by a hypersensitivity mechanism.

        The Inhibitory Mechanism of Aloe Glycoprotein (NY945) on the Mediator Release in the Guinea Pig Lung Mast Cell Activated with Antigen-Antibody Complexes

        It has been reported that the g1ycoprotein extracted from A10e has strong anti-inflammatory response,However, there has been no research report yet about the effect of A10e on allergic hypersensitivity reactivity, By using guinea pig 1ung mast cells, this study aimed to examine the effects of A10e g1ycoprotein (NY945) on the mediator re1eases caused by mast cell activation, and a1so aimed to assess the effects of NY945 on the mechanism of mediator re1eases in the mast cell activation. We partially purified mast cell from guinea pig 1ung tissues by using the enzyme digestion, the rough and the discontinuous density percoll gradient method. Mast cells were sensitized with IgG1 (anti-OA) and challenged with ova1bumin. Histamine was assayed by fluorometric ana1yzer, 1eukotrienes by radioimmunoassay. The phospholipase D activity was assessed by the production of 1abe1ed phosphatidy1alcoho1. The amount of mass 1,2-diacy1g1ycero1 (DAG) was measured by the eH]DAG produced when pre1abe1ed with eH]myristic acid. The phosph01ipid methy1ation was assessed by measuring the incorporation of the eH]methy1 moiety into phosph01ipids of cellu1ar membranes. Pretreatment of NY945 (10 μg) significant1y decreased histamine and 1eukotrienes re1eases during mast cell activation. The decrease of histamine re1ease was stronger than that of 1eukotriene during mast cell activation. The phospholipase D activity increased by the mast cell activation was decreased by the dose-dependent manner in the pretreatment of NY945. The amount of DAG produced by PLC activity was decreased by NY945 pretreatment. The amount of mass 1, 2-diacy1g1ycero1 produced by activation of mast cells was decreased in the pretreatment of NY945. NY945 pretreatment strong1y inhibited the incorporation of the eH]methy1 moiety into phosph01ipids. The data suggest that NY945 purified from A10e inhibi엄 in part an increase of 1, 2-diacy1g1ycero1 which is produced by activating mast cells with antigen-antibody reactions, which is mediated via phosphatidylcholinephospholipase D and phosphatidylinosit01-phospholipase C systems, and then followed by the inhibition of histamine re1ease. Furthermore, NY945 reduces the production of phosphatidylch01ine by inhibiting the methy1transferase 1 and II, which decreases the conversion of phosphatidylcho1ine into arachidonic acid and inhibits the production of 1eukotrienes.

      • Characteriztion of Potato Spindle Tuber Viroid Isolates with Different Symptoms by Polyacrylamide Gel Electrophoresis

        지표식물인 토마토(cv. Rutgers)에 mild, intermediate 및 severe 분리형(isolate)의 Potato spindle tuber viroid(PSTV)를 접종시킨 다음 같은 조건 아래에서 생장시킨 결과, 접종된 자표 식물은 각기 다른 증상을 나타내었다. Mild 분리형에 접종된 식물은 건전주와 비슷한 생장을 보였으나, 전기영동을 실시한 결과에서는 intermediate 및 severe 분리형에서 나타난 것과 마찬가지로 뚜렷한 viroid-band를 볼 수 있었다. 각기 다른 PSTV 분리형들의 고리 및 막대모양 RNA의 전기영동적 이동성의 차이점을 조사하고자 변성 전기영동을 실시한 결과, 각 분리형들사이에 전기영동적 이동성의 차이점은 나타나지 않았다.

        Occurrence of Apple scar viroid-Korean strain (ASSVd-K) in Apples Cultivated in Korea

        Apple is the most economically important fruit in Korea. The suspected viroid disease of dapple apple was found in apple fruits cultivated in Kyungpook province. Symptoms begin in mid-July as small circular spots, which stand out against the background color on the young fruit. Dappling of the fruit becomes more intense and easier to detect as the fruit approaches maturity; the affected spots remain yellowish as the fruit matures. no leaf or bark syndromes have been associated with this disease. The infected fruits are downgraded considerably during quality grading. The low molecular weight RNA containing viroid RNA molecules were extracted from the peels of the apples with dapple symptoms. The RNA molecules were extracted from the apples using Qiagen column chromatography. The purified RNAs were used for the synthesis of cDNA with RT-PCR. The PCR products were then ligated into a pGEM-T Easy vector, cloned and sequenced. The sequence of the viroid RNA molecule shows 331 nucleotides with one base difference ("G" insertion between the position of 133 and 134) compared with that of the Apple scar skin viroid (ASSVd) reported by Hashimoto and Koganezawa in Japan. This is the first report on the occurrence of the ASSVd in apple trees cultivated in Korea, as well as the identification of a new Korean strain of the ASSVd.the ASSVd.

      • Overexpression, Purification and Truncation Analysis of RmlC Protein of Mycobacterium tuberculosis

        dTDP-rhamnose provides L-rhamnose to the bridge-like structure between mycolyl arabinogalactan and peptidoglycan of the mycobacterial cell wall. dTDP-rhamnose is composed of glucose-l-phosphate and dTTP by four enzymes encoded by rmlA-D. To determine the region(s) of RmlC protein essential for its dTDP-4-keto-6-deoxyglucose epimerase activity, we overexpressed both whole (202 amino acids) and three different truncated (N-terminal 106 or 150 or C-terminal 97 amino acids) RmlC proteins of Mycobacterium tuberculosis. The RmlC enzyme activity in the soluble lysates of ${\Delta}rmlC$ E. coli strain $S{\Phi}874$ (DE3 PlysS) expressing the wild type or truncated rmlC genes was initially analyzed by three sequential reactions from dTDP-glucose to dTDP-rhamnose in the presence of purified RmlB and RmlD. All three soluble lysates containing the truncated RmlC proteins showed no enzyme activity, while that containing the wild type RmlC was active. This wild type RmlC was then overexpressed and purified. The incubation of the purified RmlC enzyme so obtained with dTDP-4-keto-6-deoxyglucose resulted in the conversion of dTDP-4-keto-rhamnose. The results show that the truncated regions of the RmlC protein are important for the RmlC enzyme activity in M. tuberculosis.

        Detection and Distribution of Apple scar skin viroid-Korean Strain (ASSVd-K) from Apples Cultivated in Korea

        Apple scar skin viroid (ASSVd) has been one of the most destructive diseases in Korean apple orchards. Symptoms of the scar skin viroid disease were detected in various apple cultivars, namely, Sansa, Fuji, Chukwang, Miki-Life, Hongro, and Songbongeum cultivated in the southern part of Korea. The RNA molecules were extracted from the apples bearing dapple apple symptoms with the application of CF-11 RNA extraction method. The purified RNAs were used for the synthesis of cDNA with RT-PCR. The PCR products were cloned and sequenced. The viroid RNA molecules from the six different cultivars bearing the dapple symptos showed the same nucleotide sequences as that of the Korean strain of ASSVd(ASSVd-K). ASSVd-K was detected from apple orchards in Kunwi, Sangju, Uiseong, Yeong-yang, Andong, and Youngduk in Gyeongbuk Province in 2001, and in Muju in Jeonbuk Province in 2002. As the viroid disease could be propagated vegetatively, it can be widely transmitted gradually in Korea.

      • 금속재와 적층복합재 면재를 갖는 샌드위치 패널의 저속충격 특성 연구

        In this paper, the low velocity response of four different sandwich panels with metal and laminate composite facesheets has been investigated by conducting drop- weight impact tests using an instrumented falling-weight impact tower. Square samples of 100m m sides were subjected low-velocity impact loading using an instrumented testing machine at six energy levels. Impact para meters like maximum force, time to maximum force, deflection at maximum force and absorbed energy were evaluated and compared for four different types of sandwich panels. The impact test results show that sandwich panel with composite laminate facesheet could not observe damage mode of a permanent visible indentation after impact and has a good impact damage resistance in comparison with sandwich panel with metal aluminum facesheet.
