This study was aimed to identify the effect of recombinant human Granulocyte Macrophge-Colony Stimulating Factor (rhGM-CSF) on the mucous membrane of hamster cheek pouch. After anesthetized with ketamine and xylazine, hamsters were topically administered with rhGM-CSF at the dose of 0.4 ㎍/㎖, 2㎍/㎖, 10 ㎍/㎖, 50 ㎍/㎖, and 250 ㎍/㎖ for 14 days. The thickness of the sratum basale, stratum spinosum, and stratum corneum was determined in each group. And cell proliferation in mucosal layer was also evaluated by immunohistochemistry. The thickness of stratum spinosum increased in the group administered with rhGM-CSF of 0.4 ㎍/㎖ (1.75㎛±0.69), but decreased in the groups administered with that of over 2 ㎍/㎖. In immunohistochemistry, high Ki-67 positive response was also detected in the hamster administered with rhGM-CSF of 0.4㎍/㎖. This study showed that rhGM-CSF induces the epithelial proliferation at concentration of 0.4㎍/㎖.

Objective: Instent restenosis (ISR) remains still an important issue even in drug-eluting stents era. We hypothesized that higher inflammatory reaction, which is known as an important atherosclerotic process, at the culprit lesion may have higher restenosis. We sought to evaluate the baseline coronary plaque composition in patients with ISR. Methods: Study population consisted of 241 patients with coronary artery disease, who underwent percutaneous coronary intervention with virtual histology- intravascular ultrasound (VH-IVUS) and 9 months follow up coronary angiography. We compared coronary plaque composition between patients with ISR and those without ISR. Results: Patients with ISR (n=27, 11.2%) were likely to be older (66.2±9.5 years vs. 58.7±11.7 years, p=0.002), increased high sensitivity C-reactive protein (hs-CRP, 1.60±3.59 mg/dl vs. 0.31±0.76 mg/dl, p<0.001) than those without ISR (n=214, 88.8%). Baseline angiographic, procedural findings and gray scale IVUS findings showed no significant differences between 2 groups. VH-IVUS examination showed necrotic core volume (22.1±19.9 mm3 vs.14.2±12.7 mm3, p=0.045) and percent necrotic core volume (14.3±8.7% vs. 19.5±9.1%, p=0.005) were higher in those without ISR than those with ISR, whereas percent fibrofatty volume was higher (16.6±9.7% vs. 12.4±8.4%, p=0.018) in those with ISR than those without ISR. The independent predictors for ISR were hs-CRP (Odd ratio=3.334, 95% confidence interval (CI); 1.158~9.596, p=0.026), and age (Odd ratio=3.557, 95% CI; 1.242~10.192, p=0.018). Conclusions: This study suggests that ISR was not associated with baseline coronary plaque composition but old age and an increased inflammatory marker such as hs-CRP. Aim:?It is still unclear which layer (intima or media) is mainly involved in increased carotid intima-media thickness (CIMT) by aging and also unclear regarding CIMT value suggesting high cardiovascular risk, although 75th percentile value of CIMT is known as a high risk in asymptomatic adults. We sought to find the changes of carotid intima thickness (CIT) and carotid media thickness (CMT) by aging and the 75th percentile value of CIMT in asymptomatic Korean adults.?Method:?This is an observational cohort study. Carotid ultrasound findings (n=2,204 from 12 hospital) were prospectively collected. The carotid images were sent to Korea Research Institute of Standards and Science for analysis using specialized software which can measure intima and media wall also.?Results:?Mean age was 58.1±13.5 years old (52% of men). Pearson correlation coefficient between age and right CIMT (r=0.489, p<0.001) and right CMT (r=0.482, p<0.001) were higher than those between age and right CIT (r=0.284, p<0.001). Mean right CIMT in male and female was 0.696±0.163 mm and 0.686±0.167 mm (p=0.180) and the 75 percentile value was 0.778mm and 0.771 mm, respectively. Mean right CIT was 0.311±0.069 mm and 0.303±0.064mm (p=0.009) and mean right CMT was 0.391±0.124 mm and 0.388±0.131 mm (p=0.694) in male and female, respectively. Left carotid ultrasound findings showed similar to the right one.?Conclusions:?An increased CIMT by aging was mainly due to increased CMT rather than CIT in asymptomatic adults. The 75th percentile values of right CIMT were 0.778 mm and 0.771 mm in asymptomatic Korean male and female adults, respectively.?
