자궁경부 종양에서 Nested PCR을 이용한 인유두종 바이러스 16형 및 18형의 검출과 임상적 의의

The aim of this study is to estimate the prevalence of HPV types(16, 18) in cervix of gynecologic patients and to elucidate the relationship of HPV types(16, 18) and clinical characteristics. Human papilloma virus(HPV) has attracted attention as one of the viruses linked to human cancers. Numerous clinical, epidemiological, and molecular studies indicate that certain genital HPV types are associated with cervical carcinoma. Of 100 types of HPV identified today, at least 40 have been found in the anogenital mucosa. A classification of these types into three groups with differing pathogenicity has been proposed: HPV type 6, 11, 42, 43, and 44 are detected more frequently in benign lesions such as condylomata; HPV type 31, 33, 35, 51, 52, and 58 are found more often in low grade squamous intraepithelial lesions than invasive lesions; and HPV 16, 18, 45, and 56 are predominant in high grade squamous intraepithelial lesions and invasive carcinoma. To detect HPV types prevalent in carcinoma of the uterine cervix, a type-specific, sensitive polymerase chain reaction(PCR)-based assay for HPV types 16, 18 was applied to 18 cervical carcinomas(15 squamous cell carcinomas and 3 adenocarcinomas), 21 cases of cervical intraepithelial neoplasia(CIN), and 42 samples of cervicitis and histologically normal. DNA extraction from cervical swab was done using DNAzol™(MBI, USA) and Nested PCR was done using HPV detection kit(Bioneer, Korea). The amplified product was analyzed directly on the basis of the site of ethidium bromide-stained band visible after agarose gel electrophoresis. The results were as follows 1. The positivity rates of HPV type 16 and HPV type 18 were 12.5%(90/718), 2,5%(18/718). so, HPV 16 positive cases(12.5%) was five folds as HPV 18 positive cases(2.5%) 2. Forty eight percents(39/81) of HPV 16 or HPV 18 positive cases were CIN, microinvasive cancer. 3. Age, Gestation number, Clinical stage, Histologic cell type in relation to HPV types were not statistically significant.

<P>Although the pathogenic effects of alcohol abuse on brain are well established, its specific effects on the intracellular signal transduction pathways of glial cells in the central nervous system (CNS) are poorly understood. In this study, we evaluated how ethanol affects the glial signal transduction associated with inflammatory activation. Lipopolysaccharide (LPS), gangliosides, and interferon (IFN)-γ induced the inflammatory activation of glia, which was differentially influenced by ethanol: 1) ethanol inhibited LPS- or gangliosides-induced, but not IFNγ-induced, glial activation as demonstrated by the production of nitric oxide and the expression of inflammatory genes such as interleukin-1β, tumor necrosis factor-α, IP-10, and CD86; 2) nuclear factor (NF)-κB or JAK/STAT1 pathway was necessary for LPS- or IFNγ-induced glial activation, respectively; 3) ethanol inhibited LPS-induced NF-κB activation; and 4) ethanol did not significantly affect IFNγ-induced STAT1/IRF-1 activation. Based on these results, ethanol seems to inhibit selectively some parts of the glial signal transduction pathways that are associated with inflammatory activation, which may lead to the deregulation of CNS inflammatory responses. © 2005 Wiley-Liss, Inc.</P>

<P>Four novel secondary metabolites possessing a unique 6/5/5 tricyclic ring system, streptoglycerides A-D (<B>1</B>-<B>4</B>), were isolated from a marine actinomycete <I>Streptomyces</I> sp. derived from a mangrove sample collected on Kosrae Island. Their structures and absolute configurations were elucidated by spectroscopic data and electronic circular dichroism data. Streptoglyceride C (<B>3</B>) showed a weak inhibitory effect on the production of nitric oxide in BV-2 microglia cells.</P> [FIG OMISSION]</BR>

Inhibitory effect of Scutellaria baicalensis ethanol extracts (SR) on chemical mediator release and immunoglobulin (Ig) production from Sprague-Dawley rats originated cells as type I allergic reaction was examined. SR showed concentration-dependent inhibition on basal and concanavalin A (ConA)-stimulated Ig production. In the mesenteric lymph node lymphocytes, the inhibitory effect of SR on the IgE production in the presence of Con A was stronger than these on IgA and IgG production. Moreover, tumor necrosis factor-alpha $(TNF-{\alpha})$ production-inhibiting effect of SR in the presence ConA was observed. However, SR did not affect the production of $interferon-{\gamma}$. SR also inhibited histamine release from the peritoneal exudate cells stimulated with a calcium ionophore A23187. In the case of leukotriene B4, SR markedly inhibited it at the concentration of 100 mg/ml. From these results, ethanol extracts obtained from Scutellaria baicalensis may have an anti-allergic effect on the intestinal system of rats.
Signaling Pathway Analysis of $MPP^+$-treated Human Neuroblastoma SH-SY5Y Cells

Parkinson's disease (PD) is a common chronic neurodegenerative disorder primarily caused by death of dopaminergic neurons in the substantia nigra pars compacta (SN). Human neuroblastoma SH-SY5Y cells have been broadly utilized in studies of mechanisms of the pathogenesis underlying $MPP^+$-induced PD mimics. However, the pathways impacted in $MPP^+$-treated SH-SY5Y cells have not been examined at genome-wide level. Here, we examined genome-wide response of $MPP^+$-treated SH-SY5Y cells with whole genome expression array including 47,231 probes and identified 169 genes that were significantly differentially expressed in a significance analysis of microarrays. Gene ontology (GO) terms such as GO:0051900 (regulation of mitochondrial depolarization), GO:0008637 (apoptotic mitochondrial changes), GO:0090090 (negative regulation of canonical Wnt receptor signaling pathway), and GO:0009968 (negative regulation of signal transduction) were significantly overrepresented in a differentially expressed genes (DEGs) analysis. The DEG information and their fold changes in expression were used to identify the pathways impacted. Five pathways, including systemic lupus erythematosus, alcoholism, prion diseases, the Wnt signaling pathway, and axon guidance had a significant impact at a threshold of $P_G=0.05$. Our results suggest that $MPP^+$ toxicity of SH-SY5Y cells might be related to transcriptional dysregulation by a nucleosome structural defect, endoplasmic reticulum stress, and inhibited canonical Wnt signaling.

<P><B><I>Importance of the field:</I></B> Increasing evidence indicates that glial cells play a pivotal role in a wide range of brain diseases. As glial cells orchestrate inflammatory responses in the CNS, recent studies have focused on glial cells and neuroinflammation as drug targets for the treatment of neuroinflammatory and neurodegenerative diseases.</P><P><B><I>Areas covered in this review:</I></B> In this review, we aim to give an overview of the current literature and patents for inhibitors of inflammatory oxidative enzymes in glia such as NADPH oxidase, myeloperoxidase, COX-2 and 5-lipooxygenase.</P><P><B><I>What the reader will gain:</I></B> Recent literature and patents on natural products or small molecule-based inhibitors of glial oxidative enzymes are reviewed.</P><P><B><I>Take home message:</I></B> Extensive studies and patents recently reported in this field suggest that glial inhibitors may soon proceed to clinical trials. However, before glial inhibitors can serve as novel drugs for the treatment of neuroinflammatory disorders, the neurotoxic and neuroprotective effects of glial neuroinflammatory responses need to be better dissected.</P>
대학의 외식창업교육 서비스품질이 학생만족도와 창업의지에 미치는 영향: 학교기업 프로그램 참여자들을 중심으로

This study analyzes the correlation between college food service entrepreneurship education, student satisfaction, self-efficacy, and entrepreneurial intention. Major results of the study are as follows. First, Out of five sub-factors of service quality (SERVQUAL) of foodservice start-up education, the factors of materiality, reliability, and empathy were verified to have a significant effect on student satisfaction, while responsiveness and certainty did not make significant effects on student satisfaction. Second, students" satisfaction with foodservice entrepreneurship education did not directly affect the will of entrepreneurial intention of the students. It rather indirectly affects through self-efficacy. Third, student satisfaction was found to have a positive effect on self-efficacy. Fourth, self-efficacy was found to have a positive effect on entrepreneurial intention. Fifth, self-efficacy was found to play a full mediating role in the relationship between student satisfaction and entrepreneurship. This study is meaningful in that it presented basic data for future start-up research as a concrete empirical analysis on the effect of the service quality of college foodservice entrepreneurship education on student satisfaction and entrepreneurial intention. In particular, this study can be differentiated from other existing studies in that it found that the positive effect of self-efficacy is a variable that enhances the entrepreneurial intention through educational satisfaction.
