        In this work, we present the comparison between d<SUB>31</SUB> and d<SUB>33</SUB> mode characterization using the PZT micro-actuator for large displacement. The PZT micro-actuator consisted of Si, PZT, and Pt layer on SOI wafer. The electrode shapes were laminated and interdigitated for d<SUB>31</SUB> and d<SUB>33</SUB> mode, respectively. In order to characterize the actuation mode, we measured the displacement using laser interferometer. The maximum displacement of d<SUB>31</SUB> mode was 12.2㎛ at 10V, the actuation characterization of d<SUB>31</SUB> was better than that of d<SUB>33</SUB> mode. We estimated that displacement of d<SUB>33</SUB> mode would be larger than that of d<SUB>31</SUB> above 30V.

        Potential roles of D-serine and serine racemase in experimental temporal lobe epilepsy

        <P>To confirm the roles of D-serinergic gliotransmission in epilepsy, we investigated the relationship between spatiotemporally specific glial responses and the D-serine/serine racemase system in mesial temporal structures following status epilepticus (SE). In control animals, D-serine and serine racemase immunoreactivities were detected mainly in astrocytes. After SE, D-serine and serine racemase immunoreactivities were increased in astrocytes. Double-immunofluorescence study revealed that up-regulation of serine racemase immunoreactivity was relevant not to D-serine immunoreactivity but to nestin or vimentin immunoreactivity. Neither D-serine nor serine racemase was found in naïve or reactive microglia. In addition, phosphorylated N-methyl-D-aspartate (NMDA) receptor subunit 1 (pNR1-Ser896) immunoreactivity in the hippocampus was increased compared with controls. Increased D-serine immunoreactivity showed direct correlation with the phosphorylation of Ser896 of NR1. Given the findings of our previous study, these findings suggest that D-serine and serine racemase in astrocytes may play roles in neuronal hyperexcitability via a cooperative activation of NMDA receptors. Furthermore, serine racemase may be involved in migration and differentiation of immature astrocytes, which is relevant to reactive astrogliosis. © 2010 Wiley-Liss, Inc.</P>

        Implication of intracellular ROS formation, caspase-3 activation and Egr-1 induction in platycodon D-induced apoptosis of U937 human leukemia cells

        Platycodon D is a major constituent of triterpene saponins found in the root of Platycodon grandiflorum, Platycodi Radix, which is widely used in traditional Oriental medicine for the treatment of many chronic inflammatory diseases. The results of previous studies have shown that this compound has in vitro growth-inhibitory activity in human cancer cells, however, the mechanism by which this action occurs is poorly understood. In this study, we examined the effects of platycodon D on the production of reactive oxygen species (ROS) and evaluated the association of these effects with apoptotic tumor cell death using a human leukemic U937 cell line. The results of this study demonstrate that platycodon D mediates ROS production, and that this mediation is followed by a decrease in mitochondrial membrane potential (MMP, ΔΨ<SUB>m</SUB>), activation of caspase-3, and cleavage of poly (ADP-ribose) polymerase (PARP). Both the cytotoxic effects and apoptotic characteristics induced by platycodon D treatment were significantly inhibited by z-DEVD-fmk, a caspase-3 inhibitor, which demonstrates the important role that caspase-3 plays in the observed cytotoxic effect. Additionally, the transcription factor early growth response-1 (Egr-1) gene was transcriptionally activated and the levels of non-steroidal anti-inflammatory drug (NSAID)-activated gene-1 (NAG-1) protein were elevated in platycodon D-treated U937 cells. However, the quenching of ROS generation in response to treatment with a ROS scavenger, N-acetyl-l-cysteine, reversed the platycodon D-induced apoptosis effects via inhibition of Egr-1 activation, ROS production, MMP collapse, and the subsequent activation of caspase-3. Although further studies are needed to demonstrate that increased expression of Egr-1 by platycodon D leads directly to NAG-1 induction and subsequent apoptosis, our observations clearly indicate that ROS induced through Egr-1 activation are involved in the early molecular events involved in the platycodon D-induced apoptotic pathway.

        Protein Profile in Corpus Luteum during Pregnancy in Korean Native Cows

        Steroidogenesis requires coordination of the anabolic and catabolic pathways of lipid metabolism, but the profile of proteins associated with progesterone synthesis in cyclic and pregnant corpus luteum (CL) is not well-known in cattle. In Experiment 1, plasma progesterone level was monitored in cyclic cows (n = 5) and pregnant cows (n = 6; until d-90). A significant decline in the plasma progesterone level occurred at d-19 of cyclic cows. Progesterone level in abbatoir-derived luteal tissues was also determined at d 1 to 5, 6 to 13 and 14 to 20 of cyclic cows, and d-60 and -90 of pregnant cows (n = 5 each). Progesterone level in d-60 CL was not different from those in d 6 to 13 CL and d-90 CL, although the difference between d 6 to 13 and d-90 was significant. In Experiment 2, protein expression pattern in CL at d-90 (n = 4) was compared with that in CL of cyclic cows at d 6 to 13 (n = 5). Significant changes in the level of protein expression were detected in 32 protein spots by two-dimensional polyacrylamide gel electrophoresis (2-DE), and 23 of them were identified by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF-MS). Six proteins were found only in pregnant CL, while the other 17 proteins were found only in cyclic CL. Among the above 6 proteins, vimentin which is involved in the regulation of post-implantation development was included. Thus, the protein expression pattern in CL was disorientated from cyclic luteal phase to mid pregnancy, and alterations in specific CL protein expression may contribute to the maintenance of pregnancy in Korean native cows.

        d-pinitol has been demonstrated to exert insulin-like and anti-inflammatory activities. However, its anti-allergic effect in the Th1/Th2 immune response is poorly understood. Recently, it was shown that T-bet and GATA-3 are master Th1 and Th2 regulatory transcription factors. In this study, we have attempted to determine whether d-pinitol regulates Th1/Th2 cytokine production, T-bet and GATA-3 gene expression in OVA-induced asthma model mice. We also examined to ascertain whether d-pinitol could influence eosinophil peroxidase (EPO) activity. After being sensitized and challenged with ovalbumin (OVA) showed typical asthmatic reactions. These reactions included an increase in the number of eosinophils in bronchoalveolar lavage (BAL) fluid, an increase in inflammatory cell infiltration into the lung tissue around blood vessels and airways, airway luminal narrowing, and the development of airway hyper-responsiveness (AHR). The administration of d-pinitol before the last airway OVA challenge resulted in a significant inhibition of all asthmatic reactions. Accordingly, this study may provide evidence that d-pinitol plays a critical role in the amelioration of the pathogenetic process of asthma in mice. These findings provide new insight into the immunopharmacological role of d-pinitol in terms of its effects in a murine model of asthma, and also broaden current perspectives in our understanding of the immunopharmacological functions of d-pinitol.

      • A 3D human neural cell culture system for modeling Alzheimer's disease

        Stem cell technologies have facilitated the development of human cellular disease models that can be used to study pathogenesis and test therapeutic candidates. These models hold promise for complex neurological diseases such as Alzheimer's disease (AD), because existing animal models have been unable to fully recapitulate all aspects of pathology. We recently reported the characterization of a novel 3D culture system that exhibits key events in AD pathogenesis, including extracellular aggregation of amyloid-β (Aβ) and accumulation of hyperphosphorylated tau. Here we provide instructions for the generation and analysis of 3D human neural cell cultures, including the production of genetically modified human neural progenitor cells (hNPCs) with familial AD mutations, the differentiation of the hNPCs in a 3D matrix and the analysis of AD pathogenesis. The 3D culture generation takes 1–2 d. The aggregation of Aβ is observed after 6 weeks of differentiation, followed by robust tau pathology after 10–14 weeks.

        초기 계배에 대한 Actinomycin D와 Puromycin의 영향

        Chick embryos received a single injection of actinomycin D($0.1{\mu}g,\;0.05{\mu}g\;or\;0.1{\mu}g$) or puromycin($10.0{\mu}g,\;30.0{\mu}g\;or\;50.0{\mu}g$) into the yolk sac of Arbor acres chick embryos either prior to incubation or at certain periods of time (48, 96 and 144 hours) after incubation. After 10days of incubation, surviving embryos were investigated morphologically and biochemically. Embryos treated with actinomycin D or puromycin showed a high mortality when they were exposed prior to incubation and at 48 hours after incubation. Electron micrographs of chondrocytes in tarso-metatarsal of antibiotics (actinomycin D or puromycin) treated embryos showed the destruction of cytoplasm and nuclei when they were exposed prior to incubation. Endoplasmic reticulum was expanded and mitochondria were damaged in chondrocytes of surving embryos treated with low doses at 48 hours, 96 hours or 144 hours after incubation. The activities of enzymes such as lactate dehydrogenase, malate dehydrogenase and succinate dehydrogenase in embryos treated with actinomycin D or puromycin were much less than those of the saline treated group. Also, the amounts of DNA, RNA and protein were greatly decreased.

      • Valeriana officinalis extract and its main component, valerenic acid, ameliorate d-galactose-induced reductions in memory, cell proliferation, and neuroblast differentiation by reducing corticosterone levels and lipid peroxidation

        Valeriana officinalis is used in herbal medicine of many cultures as mild sedatives and tranquilizers. In this study, we investigated the effects of extract from valerian root extracts and its major component, valerenic acid on memory function, cell proliferation, neuroblast differentiation, serum corticosterone, and lipid peroxidation in adult and aged mice. For the aging model, d-galactose (100mg/kg) was administered subcutaneously to 6-week-old male mice for 10weeks. At 13weeks of age, valerian root extracts (100mg/kg) or valerenic acid (340μg/kg) was administered orally to control and d-galactose-treated mice for 3weeks. The dosage of valerenic acid (340μg/kg), which is the active ingredient of valerian root extract, was determined by the content of valerenic acid in valerian root extract (3.401+/-0.066mg/g) measured by HPLC. The administration of valerian root extract and valerenic acid significantly improved the preferential exploration of new objects in novel object recognition test and the escape latency, swimming speeds, platform crossings, and spatial preference for the target quadrant in Morris water maze test compared to the d-galactose-treated mice. Cell proliferation and neuroblast differentiation were significantly decreased, while serum corticosterone level and lipid peroxidation in hippocampus were significantly increased in the d-galactose-treated group compared to that in the control group. The administration of valerian root extract significantly ameliorated these changes in the dentate gyrus of both control and d-galactose-treated groups. In addition, valerenic acid also mitigated the d-galactose-induced reduction of these changes. These results indicate that valerian root extract and valerenic acid enhance cognitive function, promote cell proliferation and neuroblast differentiation, and reduce serum corticosterone and lipid peroxidation in aged mice.

        In this paper, for moving heavy object a new type of human-sized Biped Walking Robot, KUBIR2, driven by the closed-chain type of joint actuator was developed Kubir2 is totally composed of29 degree-of-freedom as head is 4 d.o.f, arms are 12 d.o.f, legs are 12 d.o.fand waist is 1 d.o.f. The weight of the robot is 75(Kg) with 160㎝ tall, but the robot is designed to be capable of supporting 120Kg. The weight of one arm is light enough to be 9kg but capable of delivering 10㎏ load. Linear movement of joint actuators is composed of the four-bar-link mechanism driven by the ball screw which has high strength and high gear ratio. The control system of the robot is developed by using embedded computer and TMS320LF2407 DSP microprocessors. Each joint actuator is controlled by using DSP microprocessors. The communication system between the main controller and joint controllers is developed using the CAN communication system. The performance of the developed robot was validated through walking experiments with supporting 15(㎏) load.
