Mesophyll and callus tissues of Hibiscus syriacus and H. rosa-sinensis obtained through in vitro culture of immature embryos were used for protoplast isolation, and several factors influencing the yield were investigated in this experiment. Protoplast isolation from mesophyll tissues required preplasmolysis in CPW 13M medium for 3 or 5 hours under dark conditions. Treatment of mesophyll tissues to 1.0mg/ℓ 2,4-D solution for 12 hours prior to preplasmolysis increased yield of protoplast in H. syriacus and 3.0mg/ℓ 2,4-D for 24 hrs in H. rosa-sinensis. 0.7M mannitol in CPW medium was the most effective as osmoticum and protoplast yield was increased in the enzyme mixture of 3-5% Cellulase, 0.5% Macerozyme and 0.2% Driselase. Physiological age of mesophyll tissue in vitro influenced significantly on the protoplast yield:young tissue grown for less than 5-week was more productive than 8-week old ones for high protoplast yield. Optimum amount of mesophyll and callus tissues in a definite 5 ml of enzyme mixture was 400 and 500 mg, respectively and optimum digestion time for healthy protoplast yield ranged from 5 to 7 hours.

Oat roots(Avena sativa) grown for 7 and 9 days were extracted for the total microsomal fraction. It was found that both day 7 and day 9 had similar trends in their response to various stimulators and inhibitors. 50mM KCI was found to slightly stimulate ATPase activity. Sodium vanadate severely inhibited ATPase activity at both 100 and 500μ mole concentrations and KCI was not able to reverse the inhibition of vanadate. KNO3 inhibited ATPase activity in both day 7 and 9 but not to as large an extent as vanadate.

Hypocotyl-derived callus cultures of Brassica pekinensis cv. Chunseng were grown on Murashige-Skoog medium containing NaCl, Na_2SO_4 and mannitol to clarify the effect of salts on callus growth, proline accumulation and water relations. Na_2SO_4 was more inhibitory than NaCl when survival rate, growth and fresh:dry weight ratios of established callus were measured. Fresh weight in 0.25% NaCl treatment was more than twice as increasable in comparison to the same concentration of Na_2SO_4 or control. Proline concentration was increased on either salt, attained at highest levels at the 5th subculture, and fluctuated as affected by both salts or mannitol. Concentrations of redeucing sugars were sharply increased after 2 or 3 times of subculture and decreased by increasing subculture. The water and osmotic potential in callus reached a maximum negative value after two weeks in culture, regardless of salt type and mannitol and tugor remained relatively constant on both salt and mannitol treatments.
