        Comparative transcriptome analysis of ATP-binding cassette (ABC) transporter genes in eri-silkworm, Samia cynthia ricini in response to 1- deoxynojirimycin

        1-Deoxynojirimycin (DNJ) as a kind of alkaloid has been confirmed that could modulate glycometabolism andhas toxicity for the eri-silkworm in our previous research. On the contrary, what is the potentially defensivemechanism when the DNJ enters the eri-silkworm. Based on comparative transcriptome sequencing, we foundthat ATP-binding cassette (ABC) transporter genes could be induced significantly by DNJ. In this study, a total of16 putative ABC transporter genes were identified, which can be classified into seven subfamilies, namely oneABCA, four ABCBs, three ABCCs, two ABCDs, one ABCE, three ABCFs, and two ABCGs. Phylogenetic analysisrevealed that ScABCs had strong conservation with Bombyx mori. Reverse transcription quantitative PCR (RTqPCR)suggested that 6 ABC transporters had a strong positive correlation between RT-qPCR and transcriptomedata. Additionally, S. c. ricini ABC transporter C-subfamily 4 (ScABCC4), S. c. ricini ABC transporter G-subfamily4 (ScABCG4), S. c. ricini ABC transporter A-subfamily 3 (ScABCA3) and S. c. ricini ABC transporter C-subfamily10 (ScABCC10) showed different expression pattern in two feed dose (1% and 2% DNJ) and three time points(6h, 12 h, 48 h). This study provides the first study on identification, characterization and expression patterns ofABC transporter genes in S. c. ricini response to DNJ, and lays a foundation for further understanding of theirphysiological roles response to the alkaloid.

        Finite element simulation of unconstraint vibration treatment for fatigued copper film

        In this paper, in order to investigate the residual strain relief in the fatigued copper film with a thickness of 33 μm, an unconstraint vibration treatment process was simulated by finite element (FE) method. The experimental processes, including fatigue and unconstraint vibration processes, were simulated. The fatigue process for the original specimen and the vibration process for the damaged specimen were analyzed by the static and transient dynamic analysis, respectively. The simulation results showed that the unconstraint vibration treatment can lead to the residual strain of the fatigued copper film to be reduced and redistributed. The effect of the vibration parameters on the treatment effectiveness was analyzed by the different fatigue experiments and unconstraint vibration treatments, and the results showed that the FE simulation results are in agreement with the experimental observations.

        <P>Superoxide dismutase (SOD, EC 1.15.1.1) plays an important role in antioxidant defense in organisms exposed to oxygen. However, there is a lack of research into the regulation of SOD activity and structural changes during folding, especially for SOD originating from extremophiles. We studied the inhibitory effects of trifluoroethanol (TFE) on the activity and conformation of manganese-containing SOD (Mn-SOD) from Thermus thermophilus. TFE decreased the degree of secondary structure of Mn-SOD, which directly resulted in enzyme inactivation and disrupted the tertiary structure of Mn-SOD. The kinetic studies showed that TFE-induced inactivation of Mn-SOD is a first-order reaction and that the regional Mn-contained active site is very stable compared to the overall structure. We further simulated the docking between Mn-SOD and TFE (binding energy for Dock 6.3, -9.68 kcal/mol) and predicted that the LEU9, TYR13, and HIS29 residues outside of the active site interact with TFE. Our results provide insight into the inactivation of Mn-SOD during unfolding in the presence of TFE and allow us to describe ligand binding via inhibition kinetics combined with computational predictions.</P>

        The Effect of Postural Correction and Subsequent Balloon Inflation in Deformity Correction of Acute Osteoporotic Vertebral Fractures

        Objective : To determine deformity correction by postural correction and subsequent balloon inflation in acute vertebral compression fractures (OVCFs) and to examine the effect of bone mineral density on deformity correction. Methods : A totol of 50 acute OVCFs received balloon kyphoplasty. Lateral radiographs were taken and analyzed at five different time points : 1) preoperative, 2) after placing the patient in prone hyperextended position, 3) after balloon inflation, 4) after deposition of the cement, and 5) postoperative. All fractures were analyzed for height restoration of anterior (Ha), middle (Hm) and posterior (Hp) vertebra as well as Cobb angle and Kyphotic angle. The bone mineral density (BMD) of lumbar spine was measured by dual-energy X-ray absorptiometry. According to the T-score, the patients were divided into two groups which were osteoporosis group and osteopenia group. Results : Postoperative measurements of Ha, Hm and the Cobb angle demonstrated significant reduction of 4.62 mm, 3.66 mm and $5.34^{\circ}$ compared with the preoperative measurements, respectively (each p<0.05). Postural correction significantly increased Ha by 5.51 mm, Hm by 4.35 mm and improved the Cobb angle by $8.32^{\circ}$ (each p<0.05). Balloon inflation did not demonstrate a significant improvement of Ha, Hm or the Cobb angle compared with baseline prone hyperextended. Postural correction led to greater improvements of Ha, Hm and Cobb angle in osteoporosis group than osteopenia group (each p<0.05). Conclusion : In acute OVCFs, the height restoration was mainly attributed to postural correction rather than deformity correction by balloon inflation. BMD affected deformity correction in the process of postural correction.

      • Molecular Cloning and Bioinformatic Analysis of SPATA4 Gene

        Full-length cDNA sequences of four novel SPATA4 genes in chimpanzee, cow, chicken and ascidian were identified by bioinformatic analysis using mouse or human SPATA4 cDNA fragment as electronic probe. All these genes have 6 exons and have similar protein molecular weight and do not localize in sex chromosome. The mouse SPATA4 sequence is identified as significantly changed in cryptorchidism, which shares no significant homology with any known protein in swissprot databases except for the homologous genes in various vertebrates. Our searching results showed that all SPATA4 proteins have a putative conserved domain DUF1042. The percentages of putative SPATA4 protein sequence identity ranging from 30% to 99%. The high similarity was also found in 1 kb promoter regions of human, mouse and rat SPATA4 gene. The similarities of the sequences upstream of SPATA4 promoter also have a high proportion. The results of searching SymAtlas (http://symatlas.gnf.org/SymAtlas/) showed that human SPATA4 has a high expression in testis, especially in testis interstitial, leydig cell, seminiferous tubule and germ cell. Mouse SPATA4 was observed exclusively in adult mouse testis and almost no signal was detected in other tissues. The pI values of the protein are negative, ranging from 9.44 to 10.15. The subcellular location of the protein is usually in the nucleus. And the signal peptide possibilities for SPATA4 are always zero. Using the SNPs data in NCBI, we found 33 SNPs in human SPATA4 gene genomic DNA region, with the distribution of 29 SNPs in the introns. CpG island searching gives the data about CpG island, which shows that the regions of the CpG island have a high similarity with each other, though the length of the CpG island is different from each other.This research is a fundamental work in the fields of the bioinformational analysis, and also put forward a new way for the bioinformatic analysis of other genes.

        북한의 항일서사 연구시론

        북한문학에 있어서 항일투쟁문학은 김일성의 정통성과 관계되는 부분으로서 중요한 위치를 차지한다. 특히 주체시대에 항일투쟁문학은 문학의 주요 흐름으로 자리 잡았다. 사실 북한의 항일투쟁문학은 해방공간에서부터 시작되어 주체시대에 들어서서 고조를 이루었다. 즉 해방공간에 있어서 "김일성 장군의 노래"는 그 서곡을 이루었고 “백두산”은 ‘영웅서사시’를 형성했다. 주체시대에 있어서는 “불멸의 총서” 해방 전 편 ‘항일투쟁편’을 창작하여 김일성의 항일서사를 전면적이고 체계적으로 진행했다. 본고는 해방공간과 주체시대에 있어서 김일성항일서사를 고찰했다. 즉 해방공간에 있어서 김일성을 건국의 주인공으로서 ‘영웅서사시’와 주체시대에 있어서 유일사상체계를 수립에 있어서 김일성 신격화와 우상화 작업으로서의 ‘영웅서사시’를 고찰했다. 결론적으로 보면 북한에서의 항일서사는 김일성을 둘러싸고 그의 정치적 자본을 얻고 확립하는 문학의 시녀적 성격이 다분하다. 이런 ‘영웅서사시’적 작업을 통하여 김일성은 ‘절세의 애국자’, ‘만고의 빨찌산’, ‘김일성장군’ 및 천출위인, 백전백승의 강철의 영장의 형상이 수립되고 현실적으로 "위대한 수령"으로 군림하고 인민들은 "수령복"을 만끽하는 정치적 역학관계를 형성한다. The anti-Japanese War literature of North Korean literature is consistent with the orthodoxy of Kim Il-sung. Especially in the “Juche Age”, the anti-Japanese War literature was in a dominant position in the whole North Korean literature. The literature began in the liberation war period and reached its peak in the “Juche Age”. Concretely speaking, “Song to General Kim Il-sung” in the liberation war period opened the prologue, “Beakdu Mountain” promoted “heroic narrative poem”, and Immortal Series’ prequel—“anti-Japanese War piece” in “Juche Age” started to elaborate Kim Il-sung’s anti-Japanese struggle history comprehensively and systematically. Given all this, this paper respectively investigates Kim Il-sung’s anti-Japanese struggle history in the liberation war period and “Juche Age”. It refers to the “heroic narrative poem” of the Founding Father Kim Il-sung in the liberation war period and the deific and idolized “heroic narrative poem” of Kim Il-sung in the only established ideological system of “Juche Age”. According to the investigation, the narrative of North Korean anti-Japanese War took Kim Il-sung as the main line and tried to establish his political capital. By means of the “heroic narrative poem”, Kim Il-sung is shaped as “a loyal patriot”, “an immortal guerrilla”, “General Kim Il-sung”, “the great man from above”, “an ever-victorious iron soldier”, and “a great leader”. At the same time, the “heroic narrative poem” delineates the political dynamics diagram that people enjoy “the leader’s blessing”.

        Synergistic Effects of Leflunomide and Benazepril in Streptozotocin-Induced Diabetic Nephropathy

        <P>Abstract</P><P><B><I>Background:</I></B> Leflunomide (LEF) and benazepril have renoprotective effects on diabetic nephropathy (DN) through their anti-inflammatory and anti-fibrotic activities. This study investigated whether combined treatment using LEF and benazepril affords superior protection compared with the respective monotherapies. <B><I>Methods:</I></B> Diabetes was induced with streptozotocin (STZ, 65 mg/kg) by intraperitoneal injection in male Wistar rats. Two weeks after STZ injection, diabetic rats were treated daily for 12 weeks with LEF (10 mg/kg), benazepril (10 mg/kg), or a combination of both. Basic parameters (body weight, fasting blood glucose level, and 24 h urinary protein excretion), histopathology, inflammatory [inflammatory cell infiltration (ED-1), monocyte chemoattractant protein-1 (MCP-1), and Toll-like receptor-2 (TLR-2)] and glomerulosclerotic factors [transforming growth factor-β<SUB>1</SUB> (TGF-β<SUB>1</SUB>) and connective tissue growth factor (CTGF)], and oxidative stress (8-hydroxy-2'-deoxyguanosine, 8-OHdG) were studied. <B><I>Results:</I></B> Benazepril or LEF treatment significantly prevented body weight loss and 24 h urinary protein excretion induced by diabetes; combined treatment with LEF and benazepril further improved these parameters compared with giving each drug alone (all p < 0.01). Increased expression of inflammatory (MCP-1 and TLR-2) and glomerulosclerotic (TGF-β<SUB>1</SUB> and CTGF) factors in diabetic rat kidney was reduced by treatment with either LEF or benazepril and was further reduced by the combined administration of the two drugs (p < 0.01). These effects were accompanied by suppression of urinary 8-OHdG excretion. There was no significant between-group difference in blood glucose level. <B><I>Conclusions:</I></B> LEF treatment lessens DN, and combined treatment with LEF and benazepril provides synergistic effects in preventing DN.</P><P>© 2014 S.         Experimental and Numerical Investigation on Load-Bearing Performance of Aluminum Alloy Upright Column in Curtain Walls Under Wind Pressure

        In this paper, the wind resistance performance test and numerical simulation analysis of aluminum alloy columns with different structural measures in unit glass curtain walls are carried out. The two-layer experimental model were composed of 12 unit curtain walls, of which there are 2 beams at a distance of 1.2 m from the top and bottom in 9 unit curtain walls respectively, and there is only one beam at a distance of 1.2 m from the top in 3 unit curtain walls. The dimension of each unit curtain wall is 1.2 m × 4.0 m. The section shape of the experimental model is L-shaped. Firstly, the experimental investigation and analysis of the model with arrangement of one pair of hooks under positive and negative wind were carried out. Secondly, the experimental investigation and analysis of the model with no hooks under positive and negative wind were carried out. The out-of-plane deformation, in-plane deformationand the strain of the columns were collected and analyzed. Finally, finite element analysis model was established. The similarities and differences of the analysis results and the experimental results were analyzed. The results show that the beam has a small constraint on the columns whether the wind pressure is positive or negative when the number and position of hooks are reasonable. When the wind pressure is negative, the hook has little effect on the out-of-plane deformation and stability. Under the action of positive wind, the hooks can significantly reduce the in-plane deformation and restrain the lateral torsion deformation of the columns.
