        In Vitro Blood Flow and Cell-Free Layer in Hyperbolic Microchannels: Visualizations and Measurements

        Red blood cells (RBCs) in microchannels has tendency to undergo axial migration due to the parabolic velocity profile, which results in a high shear stress around wall that forces the RBC to move towards the centre induced by the tank treading motion of the RBC membrane. As a result there is a formation of a cell free layer (CFL) with extremely low concentration of cells. Based on this phenomenon, several works have proposed microfluidic designs to separate the suspending physiological fluid from whole in vitro blood. This study aims to characterize the CFL in hyperbolic-shaped microchannels to separate RBCs from plasma. For this purpose, we have investigated the effect of hyperbolic contractions on the CFL by using not only different Hencky strains but also varying the series of contractions. The results show that the hyperbolic contractions with a Hencky strain of 3 and higher, substantially increase the CFL downstream of the contraction region in contrast with the microchannels with a Hencky strain of 2, where the effect is insignificant. Although, the highest CFL thickness occur at microchannels with a Hencky strain of 3.6 and 4.2 the experiments have also shown that cells blockage are more likely to occur at this kind of microchannels. Hence, the most appropriate hyperbolic-shaped microchannels to separate RBCs from plasma is the one with a Hencky strain of 3.         A voice-based annotation system for collaborative computer-aided design

        In this paper, we describe a voice-based interaction mechanism to annotate 3D models directly from a computer-aided design (CAD) modeling environment. The audio signal is captured and automatically transcribed to a textual 3D note, which is attached to the geometry and made available to other product information and business processes across the enterprise via a product data management system. Our approach provides a more natural and intuitive method to capture design and engineering knowledge that is particularly effective when large amounts of information need to be communicated. We discuss the rationale of the software architecture and the value of this modality for capturing knowledge in a collaborative engineering context. Finally, we examine the results of an experiment to validate our proposal. Our results show that 3D annotations are an effective mechanism to communicate design knowledge, which suggests the need for further developments in the areas of multimodal interaction methods and interfaces for CAD and collaborative tools.

        Background: Sorafenib is the fi rst approved molecular-targeted anti-angiogenic drug for advanced hepatocellular carcinoma (HCC). The drug demonstrated signifi cant survival benefi t; however, the benefi t was small and the drug often caused severe adverse effects. In addition, the drug costs too much. Results from previous studies and clinical experiences suggested that both the COX-2 inhibitor meloxicam and the ACE inhibitor perindopril had clear anti-cancer potential on HCC mainly through anti-angiogenic action. These alternatives seem much affordable and feasible to elder HCC patients with liver cirrhosis. Methods: Two Japanese male patients with HCC and hypertension were treated with meloxicam and perindopril. Informed consent was obtained prior to the treatment. Results: For Patient 1 (71 years old), perindopril and vitamin K2 were started after the 2nd surgery for recurrent HCC, which developed 5 years after the 1st surgery. TACE was performed twice to control recurrent HCC. Meloxicam was added to perindopril plus vitamin K2, and this triple treatment was maintained for 5 years, while no additional interventional therapy was done. He died of the cancer 11.5 years after initial diagnosis of HCC. There was no sign of portal tumor thrombosis or distant metastasis. Patient 2 (82 years old): HCC was detected in cirrhotic liver with hepatitis C virus infection; however, he and his family did not choose curative treatment. Perindopril, vitamin K2, and meloxicam were concurrently administered over 6 years, while the tumor grew slowly, not showing portal tumor thrombosis or distant metastasis. He died of pneumonia 6.5 years after initial diagnosis of HCC. Conclusions: The reported cases may suggest potential growth-inhibitory effect of the COX-2 inhibitor and ACE inhibitor-based treatment on HCC for those who do not desire to receive radical locoregional or systemic therapies against the tumor.

        Maitake (D Fraction) Mushroom Extract Induces Apoptosis in Breast Cancer Cells by BAK-1 Gene Activation

        For many years mushrooms have been used empirically in traditional medicine to treat several diseases. Study of the maitake mushroom, with its immunomodulatory and antitumoral properties, has led to the isolation of several bioactive compounds. One of these, D fraction, is known to reduce tumor cell viability. This study examined the effect of isolated D fraction on viability and apoptosis of human breast cancer cells (MCF7). These cells were treated with maitake (D fraction) extract at 18 μg/mL, 36 μg/mL, 91 μg/mL, 183 μg/mL, or 367 μg/mL or were left untreated (control) for 24 hours. MCF7 incubation with the maitake extract resulted in decreased cell viability [3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium assay] in a dose-dependent manner. Apoptosis was statistically significantly increased in a dose-dependent manner at every concentration tested (terminal deoxynucleotidyl transferase-mediated deoxyuridine triphosphate nick-end labeling assay). Upon incubation with D fraction, a microarray assay revealed upregulation of BAK-1 and cytochrome c transcripts, 2 proteins directly involved in the apoptotic pathway. Reverse transcriptase polymerase chain reaction studies confirmed these findings; BAK-1 was one of most overexpressed gene, as observed by microarray assay. These findings confirm the apoptotic effect of maitake D fraction in breast cancer cells and further highlight the involvement of cytochrome c release to the cytoplasm. Cytoplasmic release of cytochrome c, another player in the apoptotic pathway, was also increased after incubation with D fraction in a dose-dependent manner. This finding indicates that the effect of this compound involves mitochondrial dysfunction. The identification of the molecular mechanisms by which D fraction exerts its effects is crucial for the development of preventive and therapeutic strategies for cancer.

        Bee Venom Acupuncture Reduces Interleukin-6, Increases Interleukin-10, and Induces Locomotor Recovery in a Model of Spinal Cord Compression

        Spinal cord injuries (SCIs) initiate a series of molecular and cellular events in which inflammatory responses can lead to major neurological dysfunctions. The present study aims to investigate whether bee venom (BV) acupuncture applied at acupoints ST36 (Zusanli) and GV3 (Yaoyangquan) could minimize locomotor deficits and the magnitude of neural tissue losses, and change the balance between pro- and anti-inflammatory cytokines after an SCI by compression. Wistar rats were subjected to an SCI model by compression in which a 2-French Fogarty embolectomy catheter was inflated in the extradural space. The effects of BV acupuncture, in which 20 μL of BV diluted in saline (0.08 mg/kg) was injected at acupoints GV3 and ST36 [BV(ST36+GV3)-SCI] was compared with BV injected at nonacupoints [BV(NP)-SCI] and with no treatment [group subjected only to SCI (CTL-SCI)]. The BV(ST36+GV3)-SCI group showed a significant improvement in the locomotor performance and a decrease of lesion size compared with the controls. BV acupuncture at the ST36 + GV3 increased the expression of interleukin-10 (anti-inflammatory) at 6 hours and reduced the expression of interleukin-6 (proinflammatory) at 24 hours after SCI compared with the controls. Our results suggest that BV acupuncture can reduce neuroinflammation and induce recovery in the SCI compression model.

        Tradition preservation through fashion consumption: Contemporary clothes on Gaucho traditionalist culture in the South of Brazil

        Fashion inspired by local culture references is our starting point for understanding how fashion, as a contemporary and dynamic dimension, can preserve the capacity of representing and maintaining local traditions. Through an interpretive approach, we look at the relationship between traditional clothing preservation and the creation of a contemporary fashion associated with the gaucho traditionalist culture in southern Brazil. The study counts on 22 indepth interviews with consumers and producers of the traditional and contemporary gaucho fashion movement, as well as document analysis and netnographic data exploration on gaucho traditions. Results indicate that even facing the resistance from conservative groups that aim to keep clothing stable over time, fashion producers and consumers associated the contemporary gaucho clothes as a modern mechanism for tradition preservation. It allows the adjustment of tradition representation in line with changes in social roles, especially gender patterns. Findings reveal that contemporary gaucho fashion challenges the rigid conception of tradition, once it represents a democratic instrument able to supply a sense of belonging adjusted to contemporary time. Finally, we theorize about the ability of symbolic representations of a particular culture to be transferred to emerging fashion elements over time, without losing the bond with the local tradition.
