        사마귀 치료를 위한 Bleomycin 국소주사법

        Intralesional belomycin injection has been an effective treatment of recalcitrant warts since 1970. A 1 U/ml solution of bleomycin sulfate in normal saline or lidocaine solution was injected intralesionally in 72 warts of 33 patients on the hands, forearm, feet, and face. Dosage of bleomycin was dependent on the size of warts, usually 0.1 to 0.15 cc of solution. All patients was permitted to wash the treated area without care. 59 (81.9%) of 72 warts were cured after one injection, 12 (16.7%) warts after two or three bleomycin injections. One large wart was not cured. Most patients experienced minimal painful discomfort for two to three days after one to two days of injection, but did not take the analgesics. There was no evidence of systemic and local toxicities. The most responsive warts shoed hemorrhagic black eschars that separated after 2 to 3 weeks and healed with very little or no scar tissue. Intralesional bleomycin injection is a reliable and useful alternative for the treatment of warts.

      • Bridgehead 유도체들의 반응성 연구

        1-Adamantyl 유도체들에 대한 가용매 분해반응의 속도상수를 여러 압력 하에서 전도도 방법에 의하여 측정하였다. 얻어진 속도상수를 친행성도와 이온화력들을 이용하여 simple Grunwald-Winstein식과 extended Grunwald-Winstein식에 적용하였다. 이러한 실험 결과로부터, 본 물질들에 대한 반응성들을 용매 친행성도의 민감도, l과 용매의 이온화력의 민감도, m을 사용하여 논의하였다. Kinetics for the solvolysis of 1-adamanty1 derivatives in binary solvent mixtures have been measured by using the conductometric method under various pressures. Rate constants obtained by this method were applied to the simple Grunwald-Winstein and extended Grunwald-Winstein equations using the values for solvent nucleophilicity(N_(T)) and solvent ionozing powers(Y_(a)). From these results, Reactivities of 1-adamantyl derivatives were discussed using the the sensitivity of solvent nucleophilicity(l) and solvent ionizing power(m).

        자가 Fibrin Glue와 연골세포를 이용한 연골조직 공학 기법에 관한 연구

        Neomorphogenesis of cartilage using chondrocyte-polymer constructs is a potential source for development of cartilage reconstruction. Current tissue engineering techniques of neocartilage rely on in vivo implantation of polymer-chondrocyte constructs. The purpose of this study was to find a way to bioengineer cartilage in vitro by entrapping chondrocytes in a molded autologous fibrin glue. Chondrocytes isolated from the cartilage of rabbit joints were combined with fibrinogen extracted by a single cryoprecipitation of autologous plasma, and they were then polymerized with thrombin to create a fibrin glue with a final cell density of 2.5x10?? cells/ml. The collagen for a control study was used as a polymer. The polymer-chondrocyte constructs were cultured for 4 weeks and the fibrin-chondrocyte constructs molded in the shape of a human ear were cultured for 6 weeks in vitro. Morphometric, histochemical, and histomorphometric analysis including glycosaminoglycan quantitation confirmed the following results: 1) Highly-concentrated autologous fibrinogen was easily extracted by a single cryoprecipition of autologous olasma. 2) The fibrin-chondrocyte constructs demonstrated the presence of actively proliferating chondrocytes with the production of cartilaginous matrix(collagen and glycosaminoglycan) at 1 week after culture, as well as gross and histologic evidence similar to those of normal cartilage at 3∼4 weeks after culture. 3) The collagen-chondrocyte constructs demonstrated lower degrees of hardness and transparency, as well as a lower density of cells and glycosaminoglycan during the culture period. 4) Neocartilage generated from fibrin-chondrocyte constructs in the shape of a human ear nearly retained their original configuration and size without degeneration for 6 weeks of culture in vitro. This study demonstrated a novel method for bioengineering the molded cartilage in vitro using autologous fibrin glue as a matrix scaffold. The generated cartilage showed gross and histologic evidence similar to those of normal cartilage, retaining the original gross dimension. With further refinement, this may be a new application of tissue engineering for the reconstruction of cartilage.

      • 신장장해 가토에서 Theophylline의 약물동태

        This study was attempted to investigate the pharmacokinetics of theophylline in rabbits with folate induced renal failure. Plasma concentration and AUC of theopylline were increased in rabbits with folate induced renal failure compared with those of normal rabbits. Renal clearance and total body clearance were decreased in rabbits with folate induced renal failure compared with those of normal rabbits. Correlation of serum creatinine concentration and AUC, and correlation of serum creatinine clearance and renal clearance have linear relationship respectively. From the results of this experiment, it is desirable that dosage regimen of theophylline should be adjusted for effective and safe therapy in patient with renal failure in clinical pharmacy practice.

      • p-Nitrophenyl Chloroformate의 가에탄올 분해반응에 대한 속도론적 연구

        Acetone 용매 내에서 p-nitrophenyl chloroformate의 가에탄올 분해반응의 속도를 25.0℃에서 적정법에 의해 측정하였다. 측정된 반응속도상수로부터 초기에탄올 농도에 대한 실험적 2차 속도상수, k_(2)를 플로트한 결과 직선성이 좋음을 알 수 있었다. 이렇게 얻어진 속도론적 데이터로부터 p-nitrophenyl chloroformate의 속도식은 d[HCI]/ dt=k_(2)[p-NPC][EtOH]+ k_(3)[p-NPC][EtOH]^(2)임을 알았다. 그리고 활성화 엔탈피,ΔH^(≠)와 활성화 엔트로피, ΔS^(≠)를 구하였다. 이상의 결과로 acetone 용매 내에서 p-nitrophenyl chloroformate와 ethanol의 반응은 전체 반응에 대해 2차 반응과 3차 반응의 혼합 경쟁 반응인 이분자 첨가-제거 반응 메카니즘으로 진행됨을 알 수 있었다. The rates of ethanolysis of p-nitrophenyl chlorofonnate in acetone were measured using the titration method at 25.0℃ From these rate constants, plots of the experimental second-order rate constant, k_(2)^(≠) against the initial ethanol concentration were linear. From the kinetics data obtained, p-nitrophenyl chlorofonnate was found to be represented by d[HCI]/dt = k_(2)[p-NPC][EtOH] + k_(3) [p-NPC][EtOH]^(2). The values of activation enthalpy and entropy for the ethanolyses of p-nitrophenyl chlorofonnate was determined from the rate constants measured at various temperatures.? From these results, the reaction of these substrates with ethanol in acetone was mixed second-and third-order reaction for over-all reaction. and it could be estimated that this reaction has bimolecular addition-elimination mechanism.

      • a-Bromo-para Toluic Acid의 친핵치환반응에 대한 연구

        The specific rates for the solvolyses of α-bromo-r-toluic acid in a wide range of solvents have been measured by using the titrimetric method at 45.0 f. From the obtained kinetic data, the specific rates were applied to the extended Grunwald-Winstein equation usingthe values for N_(T) and Y_(Br). In addition to the kinetic study, gas chromatography was used to identify the products as well as to determine their relative percentages. Also, the values of activation enthalpy and activation entropy for the solvolyses of a-bromo-p-toluic acid were calculated from the rate constants measured at various temperatures. From those results, it could be estimated that the solvolysis of o -bromo-p-toluic acid proceeded through a bimolecular pathway which prefers a bond formation(associative ,S_(N)2) in all the solvents. 여러 이성분 혼합 용매 내에서 a-bromo-p-toluic acid의 가용매 분해 반응에 대한 반응 속도상수를 45.0℃에서 적정법을 이용하여 측정하였다. 얻어진 반응 속도 데이터를 이용하여 extended Grunwald-Winstein식에 적용하였다. GC 분석법을 이용하여 a-bromo-p-toluic acid 반응 생성물들을 확인하고, 그들의 상대적인 퍼센트를 결정하였따. 또한 a-bromo-p-toluic acid의 가용매 분해 반응에 대한 활성화 엔탈피와 활성화 엔트로피를 반응 속도 상수로부터 계산하였다. 이러한 실험 결과로부터 a-bromo-p-toluic acid는 모든 용매 내에서 결합 형성이 우선하는 이분자 반응으로 추정됨을 알 수 있었다.

      • 混合溶媒內에서 p-Nitrobenzyl Chloroformate의 아실화 反應에 관한 硏究

        The Kinetics of the solvolysis of p-nitrobenzyl chloroformate in hydroxylic alcohol solvents and the methanolysis of this substrate in acetone have been studied by titrimetric method at several temperatures and over a wide range of methanol concentration. The values of the activation parameter were determined from rate constants. The products of this substrate were analyzed by GC in aqueous ethanol. The mechanisms of these reactions were also estimated from the extended Grunwald-Winstein equation and the isotopic effects in methanol. From these results, it could be estimated that the solvolysis of p-nitrobenzyl chloroformate have bimolecular addition-elimination mechanism. And the reaction of this substrate with methanol in acetone is mixed second- and third- order reactions for over-all reaction. The results are consistent with a bimolecular reaction, and are explained by tetrahedral intermediate mechanism.

      • 1-Adamantyl Bromide의 압력과 온도의 영향

        여러 혼합 용매내에서 1-adamantly bromide의 가용매 분해 반응속도를 여러 온도와 압력하에서 전도도 방법에 의하여 측정하였다. 이들 속도 상수로부터 활성화 부피, ?V_(o)^(≠)활성화 엔탈피, ?H^(≠), 활성화 엔트로피 ?S^(≠)을 구하였다.이때 모든 혼합물내에서 ?V_(o)^(≠) 와 ?S^(≠)는 음의 값을 나타내었으며, ?H^(≠)는 양의 값을 얻었다. 또한 활성화 부피와 활성화 엔트로피 값들을 플로트하여 본 반응에 대한 반응 경향성을 조사하였다. Rates of the solvolysis of l-adamantyl bromide in various mixed solvents have been measured using the conductometric method at various temperatures and pressures. Rate constants obtained by this method were applied to the activated parameters. Values of the activation volumes, ?V_(o)^(≠) and the activation entropies are both negative while values of the activation enthalpies, ?V_(o)^(≠) are positive. The reactivities of these reactions were also estimated from the correlation of the activation volumes with the activation entropies.
