Cloning of Human Interleukin-2 cDNA in E. coli by Using Oligonucleotide Primers

사람 인터루킨-2(IL-2)의 cDNA 클론을 oligo-누클레오티드를 primer로 사용하여 분리하였다. 사람 leukaemic T-cell line인 Jurkat 세포로부터 mRNA를 분리 하였다. ss-cDNA를 합성하기 위하여 인터루킨-2를 코딩하는 mRNA의 3' 끝쪽에 상보적인 30 mer oligo-누클레오티드를 역전사 반응시 primer로 사용하였으며, ds-cDNA를 합성하기 위해서는 만들어진 ss-cDNA의 3' 끝쪽에 상보적인 oligo-누클레오티드를 primer로 사용하였다. 이 ds-cDNA를 사용하여 partial cDNA library를 만든 뒤 cDNA 합성에 사용한 oligo-누클레오티드를 probe로 사용하여 콜로니 hybridization을 하여 인터루킨-2 cDNA를 찾기위하여 screen하였다. 약 200개의 transformants 중에서 세 클론이 positive signal을 나타냈다. 제한효소지도를 작성하고 누클레오티드 염기서열을 결정함으로써 이들 세 클론이 모두 인터루킨-2 cDNA를 포함하고 있음을 밝혔다. 이 결과는 우리가 만든 partial cDNA library에 인터루킨-2 cDNA가 Taniguchi 등 (1983)이 만든 total cDNA library에 들어있는 것보다 약 300배 가량 증가되어 있음을 시사한다. The sequence of the oligonucleotide was complementary to the 3' end of the coding sequence of IL-2. ds-cDNA was synthesized by DNA polymerase reaction using another oligonucleotide as a primer. A partial cDNA library was prepared using the ds-cDNA and screened for the presence of IL-2 cDNA by colony hybridization using the same oligonucleotides that were used in the cDNA synthesis reactions as probes. Three out of 200 transformants showed positive signals. Analysis of these three clones by restriction enzyme mapping and nucleotide sequencing showed that all of them contained IL-2 cDNA. Our results indicated that the IL-2 cDNA was enriched in the partial cDNA library about 300 fold over the population of IL-2 cDNA in the total cDNA library reported by Taniguchi et al. (1983).

간경변증 환자들은 소화기 증상을 호소하는 경우가 많다. 위내시경 검사상 울혈성 위염이 자주 관찰되어 왔으며 이러한 위장관 증상이 간문맥압 상승으로 인한 위장관 운동기능장애와 연관되어 있을 가능성이 제시되었다. 본 연구에서는 울혈성 위염을 동반한 간경변증 환자에서의 위배출시간을 측정하고 간경변증간자의 위배출시간에 영향을 주는 요인들을 알아보고자 하였다. 2003년 10월부터 2004년 5월까지 울혈성 위염을 동반한 간경변증 환자 6명, 울혈성 위염을 동반하지 않은 간경변증 환자 4명, 20명의 정상인을 대상으로 위배출시간을 검사하였다. 울혈성 위염이 있는 간경변증 환자군의 위배출 시간이 정상 대조군의 위배출 시간보다 길었으나(331.67±317.39분, 66.55±31.39분), 통계적으로 유의한 차이는 없었다(p=0.072). 비록 통계적으로 유의한 차이를 보이지는 않았지만, 울혈성 위염을 가진 간경변증 환자에서의 위배출시간은 정상 대조군과 비교하였을 때 지연되는 양상을 보였다. 이들 자료는 간경변증 환자에서 나타나는 위배출시간 지연의 병태생리에 울현성 위염이 연관되어 있을 가능성에 대해 제시하고 있다. Patients with liver cirrhosis frequently complain gastrointestinal symptoms. Endoscopic feature of patients with cirrhosis often included congestive gastropathy. Little attention has been directed to the possible effect of portal hypertension on the motor function of the gastrointestinal tract. In this study, we evaluated the gastric emptying time in cirrhotic patients with congestive gastropathy and the factors causing the delay of gastric emptying time in patients with cirrhosis. Between October 2003 and May 2004, 6 Cirrhotic patients with congestive gastropathy, 4 Cirrhotic patients without congestive gastropathy, and 20 controls were participated in this study. The gastric emptying time was longer in cirrhotic patients with congestive gastropathy than in controls (331.67±317.39 vs 66.55±31.39 minutes), but there was no statistically significant difference (p=0.072). Although there was no statically significant difference, the gastric emptying time was more delayed in cirrhotic patients who had congestive gastropathy than normal controls. These data suggest a possibility that congestive gastropathy may contribute to pathogenesis of delayed gastric emptying in cirrhosis.

Spinal epidural abscess (SEA) is an unusual form of spinal infection. Performing multilevel laminectomies is controversial in cases of extensive SEA considering the long surgical time and mechanical instability. Here, we report the case of an older woman with extensive SEA and poor general condition who was successfully treated with a less invasive treatment, namely skipped laminotomy using a pediatric feeding tube. A 79-year-old woman complained of progressive weakness in both legs, fever, and back pain. An extensive epidural abscess from the T3 to L5 vertebrae was observed on thoracic and lumbar magnetic resonance imaging (MRI). We performed skipped laminotomy at the T8 and T12 levels, and a 5-Fr pediatric feeding tube was advanced from the caudal level toward the rostral area and rostral level toward caudal level into the dorsal epidural space. Subsequently, regurgitation was performed with saline through the pediatric feeding tube at each level. Following this, to further irrigate the unexposed epidural abscess through laminotomy, the epidural space was washed by continuous irrigation, and the irrigation system was maintained for 48 hours. Follow-up MRI performed 3 weeks after the procedure confirmed near complete removal of the abscess in the thoracic spine, with a small residual abscess in the lumbar spine.

4차 산업혁명은 광범위한 사회 분야를 발전시키고 있으며, 특히 드론은 민간분야에서의 활용이 증가함에 따라 민간분야 기술의 군사적 활용도가 높아지고 있다. 각국에서는 최근의 다양한 실전 사례를 바탕으로 드론에 대한 군사적 효욜성을 높게 평가하고 있으며, 각국의 현실에 적합한 드론 전력화 목표를 달성하기 위해 활발한 연구개발이 진행 중이다. 특히 최근의 리비아 내전과 아제르바이잔-아르메니아 전쟁은 비정규전과 정규전에서의 드론활용이 더 이상 선택이 아닌 필수가 되었음을 보이고 있다. 이러한 세계적인 추세에 따라 국내 또한 다양한 드론 연구가 진행 중이지만 주로 민간에서 활용하기 위한 연구로 군사적 활용을 위한 연구는 전통적인 고정익, 회전익 체계로 제한되어있다. 본 논문에서는 국내·외 다양한 드론 및 관련기술 개발동향을 분석하고, 세계 각국이 추진하고 있는 드론의 군사적 발전추세를 분석하여 드론의 효율적인 군사적 활용을 위한 다섯 가지 주요 기술에 대한 기술발전 방안을 제시한다. The Fourth Industrial Revolution has caused development in many aspects of the society. Especially, the increasingly used drone technology for civil purposes is utilized as technology for the military in large numbers. Furthermore, many countries highly acknowledge the military utility of drones based on various recent real warfare cases. Therefore, active research & development is underway to achieve force integration of drones suitable for each country"s reality. The recent Libyan Civil War and the Azerbaijan-Armenian War show that drone use in non-regular and regular wars was no longer an option but a necessity. So, various drone studies are underway in Korea, following this global trend. Still, research on military utilization of drones is limited to drones of the traditional fixed/rotational-wing system. Hence, this paper analyzes the development trends of various drones and related technologies at home and abroad. It further analyzes the military development trends of drones in countries around the world, and presents development plans for five major technologies for the effective military use of drones.
