        The present study was performed to investigate the effect of in-vitro maturation and in-vitro fertilization time in Korean Native Cow(KNC) oocytes on in-vitro development and to examine whether the duration of fertilization time of matured oocytes affects the blastocyst(BL) formation. There was no significant difference in cleavage and 8-cell stage rates among the treatment groups, but BL and BL/8-cell rates were significantly higher(P<0.005) in 18hr maturation(IVM18) group(31.0±5.7 and 82.0±5.1%) than 22 and 24hr maturation(IVM24) group. As the time of in-vitro fertilization time increased, the fertility rates was increased but the BL and BL/8cell rates were decreased. The proportion of BL formed on day 7 and 8 was significantly higher(P<0.05) in IVM18:IVF16 group(100%) than in IVM24:IVF24 group(55%). These results suggest that effective in-vitro production of KNC oocytes need to proper IVM and IVF times, and two conditions must be considered simultaneously.

      • 식물 추출물을 이용한 초파리 기피제 개발

        To study repellent of Drosophila melanogaster, we extracted plant materials from wild sesame leaf, cinnamon, fig leaf, ginko leaf, quince with 100% ethanol or water as an extract solvent and described an insecticidal effect on the plant extract respectively. In the result, the 100% ethanol extract had higher insecticidal effect than the water extract. The ethanol extract of wild sesame leaf showed 58% extinction effect on Drosophila and the extracts of cinnamon and fig leaves had the effect of repellent. Each ethanil extract and the extract of cinnamon water were mixed with the ethanol extract of wild sesame leaf and a secondary test on the repellent effect has been conducted. In case of cinnamon extract mixed with wild sesame leaf, it showed no larva development and low Drosophila influx and also confirmed the same effect in the ethanol extract of fig leaf. The ethanol extract of wild sesame leaf has aromatic compound and high level of flavonoid concentration that affect like lure materials and accelerate a repellent effect of Drosophila. The cinnamon extract has high concentration of phenol compound that reasons material on repellent effect. However, the effect of fig leaf can not describe because it is low concentration of phenol compound and flavonoid compared with wild sesame leaf and cinnamon. Therefore, in case of mixed plant extracts with the ethanol extract of wild sesame leaves it can make repellent product to induce larva inhibition and insecticidal effect.

        New & renewable energy has been brought to the attention of the world as eco-friendly energy source to cope with the fossil fuel depletion and rate of environment load growth. Thus, the government is actively encouraging application of New & renewable energy system. Nevertheless outcome of them has been slumped. This case is caused by shortage of the preliminary inspection and maintenance before installing systems, because scientific planning system is currently required in the pre-planning phase. Before starting to develop the planning tool of new & renewable energy system, to search available factors of development strategy, this study carry out a survey targeting the hands-on workers concerning it. As a result, when new & renewable energy system have been applied, it is evaluated available factors of them such as economics, efficiency, reliability, and so on. Also, a method in planning and design of them has frequently depended on collaborating with specialized agency or experience of working. In addition, a majority of respondents emphasized that exiting building mechanical system and new & renewable energy need to integrate or compose the hybrid system. Finally, it is analysed economical criteria such as minimum of LCC, initial investment have to optimize in advance.

        Augmenting Effect of DA-9601 on Ghrelin in an Acute Gastric Injury Model

        Background/Aims: Acute gastric injury by alcohol or indomethacin has been reported to be prevented by DA-9601, an extract of the herb Artemisia asiatica. Ghrelin, an endogenously produced gastrointestinal peptide hormone, has also been demonstrated to play a role in gastric mucosal defense. The aim of this study was to investigate the effects of DA- 9601 on ghrelin in an acute gastric injury model induced by alcohol or indomethacin. Methods: A total of 140 Sprague- Dawley rats were divided into two groups, a placebo group and a DA-9601-pretreated group. Thirty minutes later, half of the rats in each group received ethanol injury and the other half received indomethacin injury. Levels of serum ghrelin and gastric mucosal ghrelin mRNA were measured by ELISA and RT-PCR, respectively. Results: Immediately after ethanol administration, ghrelin increased in both groups pretreated with DA-9601 and placebo. However, the increase occurred more rapidly and was higher in the DA-9601-pretreated rats than in the controls that did not receive DA-9601-pretreatment. Similarly, from 30 minutes to 2 hours after indomethacin administration, the DA-9601-pretreated rats showed a significant increase in serum and gastric mucosal ghrelin concentrations, whereas placebo-pretreated rats showed only a mild increase. Conclusions: DA-9601 potentiates the endogenous production and secretion of ghrelin in acute gastric injury models induced by ethanol or indomethacin. (Gut Liver 2011;5:52-56)

        Streptozotocin - 유도 당뇨모델을 이용한 쥐눈이콩 분말의 혈당강하 및 인슐린 감수성의 효과

        본 연구에서는 임실특산물인 쥐눈이콩및 쥐눈이식초콩의 혈당강하 및 인슐린 감수성에 대한 효과를 검정콩, 메주콩과 비교하여 조사하였다. 식이물질의 성분검사 결과 쥐눈이콩과 쥐눈이식초콩이 검정콩과 메주콩에 비하여 당뇨개선의 효과가 있는 여러 아미노산 및 식이섬유 함량이 유의적으로 높게 측정되었다. 실험을 위해 평균체중 195.5±0.98 g의 숫컷쥐를 정상군(NC), 당뇨대조군(DI, DC), 검정콩식이군(DB), 메주콩식이군(DY), 쥐눈이콩식이군(DS), 쥐눈이식초콩식이군(DSS)으로 총 7개군으로 나누어 해당식이첨가 시료로 4주간 사육하였다. 당뇨유발을 위해 STZ를 1회(70 mg/kg/rat) 투여하였으며 그 결과 체중변화는 당뇨대조군(DC)에서 21.12 g 감소함에 반하여 각각의 시료를 첨가식이한 당뇨실험군에서 특히 쥐눈이콩(DS)과 쥐눈이식초콩(DSS) 식이군은 높은 식이이용 효율(FER)을 보여 각각 14.73±3.65 g 및 16.71±5.54 g의 몸무게 증가를 보였다. 음용수 및 뇨량의 측정결과 역시 당뇨실험군에서 유의적인 감소(p<0.05)를 보였다. 혈당량은 당뇨대조군(DC)에 비하여 검정콩(DB), 메주콩(DY), 쥐눈이콩(DS), 쥐눈이식초콩(DSS) 식이군이 각각 17.9%, 16.9%, 10.35%, 19.54%씩 유의하게 감소하였으나 정상대조군(NC)보다는 높은 수치를 나타내었다. 또한 당뇨쥐의 인슐린 감수성에 대한 관찰결과 당뇨대조군 중 인슐린 무처지군(DC)에 비하여 모든 당뇨실험군에서 높은 값을 보였다. 결론적으로 임실산 쥐눈이콩과 쥐눈이식초콩은 당뇨쥐의 혈당강하 및 인슐린감수성에 대한 개선작용의 가능성이 있으므로 당뇨합병증 예방의 기능성 식품으로 기대된다. Beans are acknowledged to be food resources, which have more abundant proteins and fats. The constituent parts of beans (i.e. aspartic aid, glycine, arginine) are effective against diabetes, and dietary fiber contained in the beans has an important property to maintain insulin sensitivity. Based on these, using streptozotocin (STZ)-induced diabetic rats, this study examined how the rat-eye soybean, which is principal products of the Imsil province, is effective to attenuate and/or prevent the development of diabetes mellitus. We divided rats into the non-diabetic and diabetic group, and diabetic group was further subdivided into six experimental groups [DC, diabetic control; DI, diabetes with insulin treatment (4~6 IU/rat); DB, diabetes with black bean;DY, diabetes with yellow soybean; DS, diabetes with rat-eye soybean; DSS, diabetes with vinegar-fermented rat-eye soybean. All bean treatment (1.5 ㎎/1 g body weight).]. Food efficiency ratio (FER), body weight and insulin sensitivity in diabetic rats were significantly reduced compared to those in normal control animals. These reductions were obviously attenuated by administration of a variety of beans used in this study (20~30%), and the recovery effects were comparable to the results obtained by insulin treatment. Taken together, this study suggests that all beans used may have an essential property to improve and/or attenuate the development of diabetes mellitus in rats.

        Chinese prescription Kangen-karyu and Salviae Miltiorrhizae Radix improve age-related oxidative stress and inflammatory response through the PI3K/Akt or MAPK pathways.

        <P>This study examined whether Kangen-karyu and its crude drug, Salviae Miltiorrhizae Radix, have a reno-protective effect on the age-related oxidative stress and inflammatory response through the phosphoinositide 3-kinase (PI3K)/Akt or mitogen-activated protein kinase (MAPK) pathways in aged rats. Kangen-karyu or Salviae Miltiorrhizae Radix (20 mg/kg body weight/day) was administered orally to old groups of rats for 16 days, and their effects were compared with the vehicle-treated old and young rats. The administration of Kangen-karyu caused a slight decrease in the serum glucose level and a significant decrease in the serum insulin level in the old rats. The increased levels of serum renal functional parameter (urea-nitrogen) and oxidative parameter were significantly reduced by both Kangen-karyu and Salviae Miltiorrhizae Radix. The old rats exhibited a dysregulation of the protein expression related to insulin resistance, oxidative stress, and inflammation in the kidneys, but Kangen-karyu administration significantly reduced the expression of the inflammatory proteins through the PI3K/Akt pathway. On the other hand, the Salviae Miltiorrhizae Radix-treated old rats showed a decrease in the inflammatory cytokines through the MAPK pathway. These results provide important evidence that Kangen-karyu and Salviae Miltiorrhizae Radix have a pleiotropic effect on the PI3K/Akt and MAPK pathways, showing renoprotective effects against the development of inflammation in old rats. This study provides scientific evidence that Kangen-karyu and Salviae Miltiorrhizae Radix improve the inflammatory responses via the PI3K/Akt or MAPK pathways in the kidney of old rats.</P>

        국내 강직성척추염 환자를 대상으로 한 Infliximab 치료

        Objective: Infliximab, a monoclonal antibody to tumor necrosis factor-α, is effective in patients with ankylosing spondylitis (AS), who have not responded to conventional therapy. There were no data on the efficacy and side effect of infliximab in patients with AS in Korea. The objective of this study is to observe the efficacy and adverse effect of infliximab retrospectively in Korean patients with AS. Methods: We reviewed the medical records of thirty-three AS patients. The patients were enrolled to fulfill the modified New York criteria of AS and be in active disease state and resist to conventional therapy. Patients were given 3~5 mg/kg of infliximab infusions at weeks 0, 2, 8 and 16. Information on C-reactive protein (CRP), erythrocyte sedimentation rate (ESR) and antinuclear antibody (ANA) test was collected at each infusion. The paired t-test was used for comparison between the visits. Results: There were 29 male and 4 female patients. The mean age at first infliximab treatment was 34.6±9.8 years. All patients were HLA-B27 positive. ESR and CRP decreased significantly from baseline to 16 weeks after treatment (p<0.001, respectively). The mean ESR was 76.1±36.5 mm/h at baseline and 21.3±31.6 mm/h at 16 weeks. The mean CRP was 6.4±4.8mg/dL at baseline and 1.3±2.1 mg/dL at 16 weeks. Only 1 out of 33 patients got worse. All patients were tested negative for ANA at baseline. After 16 weeks of therapy, the induction of ANA was observed in 8 patients, but no patients have lupus-like symptoms. Conclusion: Infliximab is an effective therapy with non-specific adverse effect in AS non-responsive to conventional therapy in Korea.
