      • Cytotoxicity of Trichoderma spp. Cultural Filtrate Against Human Cervical and Breast Cancer Cell Lines

        Trichoderma spp. are known as a rich source of secondary metabolites with biological activity belonging to a variety of classes of chemical compounds. These fungi also are well known for their ability to produce a wide range of antibiotic substances and to parasitize other fungi. In search for new substances, which might act as anticancer agents, the overall objective of this study was to investigate the cytotoxic effects of Trichoderma harzianum and Trichoderma asperellum cultural filtrates against human cervical and breast cancer cell lines (HeLa and MCF-7 cells respectively). To achieve this objective, cells were exposed to 20, 40, 60, 80 and 100 mg/ml of both T. harzianum cultural filtrate (ThCF) and T. asperellum cultural filtrate (TaCF) for 24h, then the cell viability and the cytotoxic responses were assessed by using trypan blue and 3-(4,5-dimethylthiazol-2yl)-2,5-biphenyl tetrazolium bromide (MTT) assays. Morphological changes in cells were investigated by phase contrast inverted microscopy. The results showed that ThCF and TaCF significantly reduce the cell viability, have cytotoxic effects and alter the cellular morphology of HeLa and MCF-7 cells in a concentration dependent manner. A concentration of 80 and 100mg/ml of ThCF resulted in a sharp decline in the cell viability percent of HeLa and MCF-7 respectively (25.2%, 26.5%) which was recorded by trypan blue assay. The half-maximal inhibitory concentrations ($IC_{50}$) of ThCF and TaCF in HeLa and MCF-7 were recorded as 16.6, 12.0, 19.6 and 0.70mg/ml respectively by MTT assay. These results revealed that ThCF and TaCF have a substantial ability to reduce the viability and proliferation of human cervical and breast cancer cells.

        Minimizing Sensing Decision Error in Cognitive Radio Networks using Evolutionary Algorithms

        Cognitive radio (CR) is envisioned as a promising paradigm of exploiting intelligence for enhancing efficiency of underutilized spectrum bands. In CR, the main concern is to reliably sense the presence of primary users (PUs) to attain protection against harmful interference caused by potential spectrum access of secondary users (SUs). In this paper, evolutionary algorithms, namely, particle swarm optimization (PSO) and genetic algorithm (GA) are proposed to minimize the total sensing decision error at the common soft data fusion (SDF) centre of a structurally-centralized cognitive radio network (CRN). Using these techniques, evolutionary operations are invoked to optimize the weighting coefficients applied on the sensing measurement components received from multiple cooperative SUs. The proposed methods are compared with each other as well as with other conventional deterministic algorithms such as maximal ratio combining (MRC) and equal gain combining (EGC). Computer simulations confirm the superiority of the PSO-based scheme over the GA-based and other conventional MRC and EGC schemes in terms of detection performance. In addition, the PSO-based scheme also shows promising convergence performance as compared to the GA-based scheme. This makes PSO an adequate solution to meet real-time requirements.

        Influence of Ti Additions on Martensitic Transformation and Magnetic Properties of Cast Ni51Fe22-xGa27Tix Shape Memory Alloys

        The effect of Ti addition on the microstructure, martensitic transformation, magnetic and mechanical properties of polycrystalline Ni51Fe22-xGa27Tix (x=0, 2 and 4) ferromagnetic shape memory alloy was investigated by scanning electron microscope, differential scanning calorimetry and X-ray diffraction. The results showed that the martensitic transformation temperature increases monotonously with the increase of fraction of Ti substitution for Fe. The increase in the martensite transformation temperatures should be related to the change of the electron concentration after the addition of Ti to Ni51Fe22-xGa27Tix alloys. According to the results of X-ray diffraction and magnetic properties, Ti has significant effect the structure of Ni51Fe22-x Ga27Tix. Adding of 4 at% Ti altered the structure of the matrix from five-layered tetragonal martensite of Ni51Fe22Ga27 and Ni51Fe20Ga27Ti2 alloys to non-modulated tetragonal martensite. Magnetic properties proved that the alloy transits from ferromagnetic, five-layered tetragonal martensite, to paramagnetic, non-modulated martensite structure,with increasing Ti content to 4 at.%. Saturation magnetization, remnant magnetization and coercivity of the alloy were significantly influenced by Ti additions. Hardness values of Ni51Fe22Ga27 increased by the addition of Ti.

      • BRCA1 Gene Mutations and Influence of Chemotherapy on Autophagy and Apoptotic Mechanisms in Egyptian Breast Cancer Patients

        Background: It is well established that mutations in the BRCA1 gene are a major risk factor for breast cancer. Induction of cancer cell death and inhibition of survival are the main principles of cancer therapy. In this context, autophagy may have dual roles in cancer, acting on the one hand as a tumor suppressor and on the other as a mechanism of cell survival that can promote the growth of established tumors. Therefore, understanding the role of autophagy in cancer treatment is critical. Moreover, defects in apoptosis, programmed cell death, may lead to increased resistance to chemotherapy. Purpose: The aim of the present study was to detect BRCA1 gene mutations in order to throw more light on their roles as risk factors for breast cancer in Egypt. Secondly the role of autophagy and apoptosis in determining response to a fluorouracil, doxorubicin, cyclophosphamide (FAC) regimen was investigated. Materials and Methods: Forty-five female breast cancer cases and thirty apparently healthy females were enrolled in the present study. Serum levels of autophagic biomarkers, Beclin 1 and LC3 as well as the serum levels of apoptosis biomarkers Bcl-2 and Caspase-3 were measured before and after chemotherapy. Results: BRCA1 mutations were found in 5 (16.7%) and 44 (99.8%) of the controls and cancer patients, the most frequent being 5382insC followed by C61G and 185 delAG. The results revealed that chemotherapy caused elevation in serum concentration levels of the autophagic biomarkers (Beclin 1 and LC3). This elevation was associated with a significant decrease in serum concentration levels of Bcl-2 and significant increase in caspase-3 concentration levels (apoptotic markers). Conclusions: The results of the present study indicate a very high level of BRCA mutations in breast cancer cases in Egypt and point to involvement of autophagic and apoptotic machinery activation in response to FAC chemotherapy.

        In present paper, we introduce and study the concepts of T₀<SUP>(i;j)</SUP>-, T₁<SUP>(i;j)</SUP> -, T₂<SUP>(i;j)</SUP>(pairwise Hausdorff)-, TR<SUP>(i;j)</SUP> (pairwise regularity)-, TN<SUP>(i;j)</SUP> (pairwise normality)-, R₀<SUP>(i;j)</SUP>-, R₁<SUP>(i;j)</SUP>-, T₃<SUP>(i;j)</SUP> - and T₄<SUP>(i;j)</SUP> - separation axioms in fuzzifying bitopological spaces and study some relations between them. Also we investigate the image of these kinds of fuzzifying bitopological spaces under some types of fuzzy mappings.

        Effect of Pre-training and Post-training Nordic Exercise on Hamstring Injury Prevention, Recurrence, and Severity in Soccer Players

        Objective To investigate the effect of adding Nordic exercise as post-training in decreasing hamstring initial, recurrent injuries rates, and their severity. Methods In this randomly controlled trial study, 34 professional football players aged 21 to 35 years were randomly assigned into two groups (17 players each) from Sporting clubs at Alexandria, Egypt. For group one, Nordic hamstring exercise (NHE) was performed pre-training and post-training. For group two, NHE was only performed pre-training. The control group was the same team during the previous season. Length of the trial was 12 weeks. The Australian football association injury form was used to collect incidence of injuries for each subject in both groups. Results Pooled results based on total injuries showed that group one had significantly less hamstring initial injuries (92% less) than the previous season, while group two had 80% less initial injuries and 85% less recurrent injuries than previous season. Regarding the severity of injuries in term of mean number of absent days, it was 1 day for group one and 2.7 days for group two while it was 7.95 days for the previous season during total risk time of 116.3±13.2 and 117.6±5.7 exposure hours for group one and group two, respectively. Conclusion The use of NHE as a prevention protocol was effective in reducing all hamstring injuries with the use of NHE during pre-training and post-training having the greatest effect.

        Effect of Bimodal-Sized Hybrid TiC–CNT Reinforcement on the Mechanical Properties and Coefficient of Thermal Expansion of Aluminium Matrix Composites

        Aluminium (Al) matrix reinforced with carbon nanotubes (CNT), micron-sized titanium carbide (TiC) particles, and bimodal(nano + micron) hybrid TiC–CNT is fabricated by solution ball milling, followed by cold compaction and vacuum sinteringto improve the mechanical properties and reduce thermal expansion. The hardness, wear resistance, compressive strength andCTE of pure Al, 0.6 wt% CNT/Al, 10 wt% TiC/Al, and hybrid 10–0.6 wt% TiC–CNT/Al composites have been investigated inthis work. Analysis of strengthening mechanisms based on theoretical models, microstructure, and properties of constituentmaterials is performed. Microstructure analysis reveals an excellent distribution of the reinforcement phase and no new phaseformation in sintered composites. The hardness value of bimodal TiC–CNT reinforced Al composite is significantly higherthan monomodal TiC reinforced composite, reaching 2.3 times the hardness value of pure Al. Similarly, the wear resistanceimproved, and CTE reduced with CNT and TiC addition but is even significantly better in the hybrid reinforced composite. Experimental values of CTE show good agreement with the theoretical model. The strength and ductility of materials aremutually exclusive, but the compressive strength of pure Al has been doubled without significant loss in ductility throughthe use of bimodal-sized hybrid TiC–CNT reinforcement in this work.

      • Impact of Cellular Genetic Make-up on Colorectal Cancer Cell Lines Response to Ellagic Acid: Implications of small interfering RNA

        Background: $K^-Ras$ activation is an early event in colorectal carcinogenesis and associated mutations have been reported in about 40% of colorectal cancer patients. These mutations have always been responsible for enhancing malignancy and silencing them is associated with attenuation of tumorigenicity. Among downstream effectors are the RAF/MEK/ERK and the PI3K/Akt signaling pathways. PI3K/Akt signaling leads to reduction of apoptosis, stimulated cell growth and enhanced proliferation. Ellagic acid (EA), a naturally occurring antioxidant, has recently emerged as a promising anti-cancer agent. Purpose: To evaluate the impact of cellular genetic makeup of two colon cancer cell lines with different genetic backgrounds, HCT-116 ($K^-Ras^-/p53^+$) and Caco-2 ($K^-Ras^+/p53^-$), on response to potential anti-tumour effects of EA. In addition, the influence of $K^-Ras$ silencing in HCT-116 cells was investigated. Materials and Methods: Cellular proliferation, morphology and cell cycle analysis were carried out in addition to Western blotting for detecting total Akt and p-Akt (at Thr308 and Ser473) in the presence and absence of different concentrations of EA. Cell proliferation was also assessed in cells transfected with different concentrations of $K^-Ras$ siRNA or incubated with ellagic acid following transfection. Results: The results of the present study revealed that EA exerts anti-proliferative and dose-dependent pro-apoptotic effects. Cytostatic and cytotoxic effects were also observed. p-Akt (at Thr308 and Ser473) was downregulated. Moreover, EA treatment was found to (i) reduce $K^-Ras$ protein expression; (ii) in cells transfected with siRNA and co-treated with EA, pronounced anti-proliferative effects as well as depletion of p-Akt (at Thr308) were detected. Conclusions: Cellular genetic makeup ($K^-Ras^-/p53^-$) was not likely to impose limitations on targeting EA in treatment of colon cancer. EA had a multi-disciplinary pro-apoptotic anti-proliferative approach, having inhibited Akt phosphorylation, induced cell cycle arrest and showed an anti-proliferative potential in HCT-116 cells (expressing mutant $K^-Ras$).

        Effects of a Low-Fermentable Oligosaccharides, Disaccharides, Monosaccharides, and Polyol Diet on Symptoms of Functional Abdominal Pain in Pediatric Patients

        Purpose: Recently, great interest has been focused on dietary fermentable oligosaccharides, disaccharides, monosaccharides, and polyols (FODMAPs) for the treatment of functional abdominal pain (FAP). Several meta-analyses, including those on the adult population, have been published, potentiating its role. However, pediatric studies are scarce. We aimed to evaluate the effect of a low-FODMAP diet on the severity of FAP in children. Methods: This clinical trial included 50 patients aged 3–18 years with irritable bowel syndrome and FAP that were not otherwise specified. The patients were instructed to receive a low FODMAP diet guided by a dietitian. The primary outcome was the percentage of responders after 2 months of dietary intervention compared with baseline. Other outcomes included changes in stool consistency and quality of life (QoL) scores using the KIDSCREEN-10 questionnaire, and weight-for-age z-scores. Results: After the dietary intervention, 74% of patients showed more than 30% lower pain intensity, as examined using the Wong-Baker Faces pain rating scale. Their QoL significantly improved, and patients have gained weight. Conclusion: A low FODMAP diet can improve pain intensity and QoL among children with functional abdominal pain, with no detrimental effects on body weight.

        The Effect of Superplasticizers on Eco-friendly Low-Energy One-Part Alkali-Activated Slag

        One-part alkali-activated materials (OP-AAM) have become a promising binder with low carbon and energy requirements associated with superior mechanical and durability characteristics. This study aims to employ commercial superplasticizers (naphthalene-based “Nb-SP” and polycarboxylate-based “PCb-SP”), as well as laboratory-prepared one (phenol–formaldehyde sulfanilate “PFS-SP”) in enhancing the properties of OP-AAM. The main problem of superplasticizers (SPs) in the AAM is their hydrolysis in the alkaline activator (NaOH) used in the activation reactions. Therefore, the thermo-chemical treatment process was utilized to mitigate the high activator alkalinity by impeding the NaOH in the aluminosilicate precursor matrix. The OP-AAM was fabricated from thermo-chemical treatment powder (TCT-P) resulting from sintering blast furnace slag (GGBFS) with 10 wt% NaOH at 300 and 500 °C. The XRD-pattern showed that NaOH was impeded in the GGBFS via sodium aluminum silicate phase formation after sintering at 500 °C. The results showed that the admixed OP-AAM prepared from TCT-P at 500 °C greatly enhanced the workability and mechanical properties. The PFS-SP proved its efficiency in improving the properties of OP-AAM prepared TCT-P at 300 and 500 °C, referring to its high stability in an alkaline medium. While PCb-SP reinforced the properties of OP-AAM prepared from TCT-P at 500 °C only, proving that PCb-SP promotes high capability in TCT-P-500 as well as in Portland cement.
