        Researching Translation Networks in Republican China: A Case Study of Dong Zhongchi’s Pride and Prejudice

        The ‘sociological turn’ in translation studies opens up a new research branch called TranslaTOR Studies (Chesterman, 2009). One of the intentions of this newfound branch is to excavate translators and their literary contributions. However, this translator-centered approach may not be able to unveil lesser -known translators whose existence can only be noted in bibliographic catalogues. In order to tackle this shortcoming, I prose to use Latour’s Actor Network Theory (1987, 1996) to establish the literary ties that connect actors. This paper examines the Chinese translation of Pride and Prejudice by Dong Zhongchi published in 1935. The translated text as well as its paratextual materials will be examined in context with the sociopolitical and sociocultural backdrop of Republican China in order to identify the ideological influence from the networked actors.

      • Prostate Cancer, High Cortisol Levels and Complex Hormonal Interaction

        Prostate cancer (PCa) is one of the most common diseases in men. It is important to assess prognostic factors and whether high cortisol levels and complex hormonal interactions could be responsible for PCa development. We evaluated the relationship between cortisol, leptin and estrogens in 141 men, 71 with PCa and the remaining 70 constituting a low risk group (LRG). They were recruited for this study from a total of 2906 middle-aged men (ages 45-70 years) who completed an evaluation for prostatic diseases at the Urology Division, Hospital de Clínicas "$Jos{\acute{e}}$ de San $Mart{\acute{i}}n$", University of Buenos Aires, in May 2009. In this cross sectional study, cortisol, PSA, total-testosterone, free-testosterone, bioavailable testosterone, LH and estradiol were measured in serum. We observed increased cortisol levels in PCa patients as compared to LRG cases (p=0.004,). Leptin and estradiol levels were also higher in PCa patients (p=0.048; p<0.0001, respectively). Logistic regression analysis indicated that serum cortisol (OR: 1.110 (95% CI 1.016-1.213), p=0.022), estradiol (OR: 1.044 (95% CI 1.008-1.081), p=0.016) and leptin (OR: 1.248 (95% CI 1.048-1.487), p=0.013) explained 27% of the variance of dependent variables, even after adjusting for age, smoking, BMI and waist circumference. We found increased cortisol levels in PCa patients as compared to LRG, as well as an altered circulating hormonal profile.

        Reinforced concrete wall as protection against accidental explosions in the petrochemical industry

        In this paper the study of a reinforced concrete wall used as protection against accidental explosions in the petrochemical industry is presented. Many alternatives of accidental scenarios and sizes of the wall are analyzed and discussed. Two main types of events are considered, both related to vessel bursts: Pressure vessel bursts and BLEVE. The liberated energy from the explosion was calculated following procedures firmly established in the practice and the effects over the structures and the reinforced concrete wall were calculated by using a CFD tool. The results obtained show that the designed wall reduces the values of the peak overpressure and impulse and, as a result, the damage levels to be expected. It was also proved that a reinforced concrete wall can withstand the blast load for the considered events and levels of pressure and impulse, with minor damage and protect the buildings.

      • Energy dissipation by friction for sliding blocks subjected to near-fault seismic base motion

        The objective of this study was to determine friction ratios that maximize energy dissipation on a seismic damper. The aforementioned friction damper basically consists of mass blocks that are able to slide on a flat surface. To carry out this analysis, a numerical-experimental approach was used. Firstly, the theoretical background and equations of motion for a SDOF system consisting of a mass supported on a flat surface with friction are introduced. Special emphasis is made on the fundamentals of stick-slip motion as well as energy considerations. Secondly, experimental studies carried out on a shaking table with harmonic and seismic records are described. These tests consisted of lead blocks contained on a U-shaped channel type aluminum section with its open end facing upwards. This configuration allowed blocks to slide solely in the direction of the base motion. Five different types of contact interfaces were considered to determine potential friction coefficients for the damper's design. Additionally, computational models based on rigid-body dynamics are presented. Numerical results were satisfactory particularly when comparing model's dissipated energy with empirical results. An analysis was carried out by calculating dissipated energy for the experimentally-calibrated models with varying friction ratios. For this purpose, eight near-fault seismic records were selected. Intervals with friction coefficients that maximize energy dissipation are proposed for each record. Finally, relationships between the computed friction ratios and register's peak ground acceleration (PGA) and root mean square acceleration (RMS) are discussed.

        Analysis of Heme Biosynthetic Pathways in a Recombinant Escherichia coli

        Bacterial heme was produced from a genetic-engineered Escherichia coli via the porphyrin pathway and it was useful as an iron resource for animal feed. The amount of the E. coli-synthesized heme, however, was only few milligrams in a culture broth and it was not enough for industrial applications. To analyze heme biosynthetic pathways, an engineered E. coli artificially overexpressing ALA synthase (hemA from Rhodobacter sphaeroides) and pantothenate kinase (coaA gene from self geneome) was constructed as a bacterial heme-producing strain, and both the transcription levels of pathway genes and the intermediates concentrations were determined from batch and continuous cultures. Transcription levels of the pathway genes were not significantly changed among the tested conditions. Intracellular intermediate concentrations indicated that aminolevulinic acid (ALA) and coenzyme A (CoA) were enhanced by the hemA-coaA co-expression. Intracellular coproporphyrinogen I and protoporphyrin IX accumulation suggested that the bottleneck steps in the heme biosynthetic pathway could be the spontaneous conversion of HMB to coproporphyrinogen I and the limited conversion of protoporphyrin IX to heme, respectively. A strategy to increase the conversion of ALA to heme is discussed based on the results.

        Use of Recombinant Entamoeba histolytica Cysteine Proteinase 1 To Identify a Potent Inhibitor of Amebic Invasion in a Human Colonic Model

        <B>ABSTRACT</B><P>Cysteine proteinases are key virulence factors of the protozoan parasite <I>Entamoeba histolytica</I>. We have shown that cysteine proteinases play a central role in tissue invasion and disruption of host defenses by digesting components of the extracellular matrix, immunoglobulins, complement, and cytokines. Analysis of the <I>E. histolytica</I> genome project has revealed more than 40 genes encoding cysteine proteinases. We have focused on <I>E. histolytica</I> cysteine proteinase 1 (EhCP1) because it is one of two cysteine proteinases unique to invasive <I>E. histolytica</I> and is highly expressed and released. Recombinant EhCP1 was expressed in <I>Escherichia coli</I> and refolded to an active enzyme with a pH optimum of 6.0. We used positional-scanning synthetic tetrapeptide combinatorial libraries to map the specificity of the P1 to P4 subsites of the active site cleft. Arginine was strongly preferred at P2, an unusual specificity among clan CA proteinases. A new vinyl sulfone inhibitor, WRR483, was synthesized based on this specificity to target EhCP1. Recombinant EhCP1 cleaved key components of the host immune system, C3, immunoglobulin G, and pro-interleukin-18, in a time- and dose-dependent manner. EhCP1 localized to large cytoplasmic vesicles, distinct from the sites of other proteinases. To gain insight into the role of secreted cysteine proteinases in amebic invasion, we tested the effect of the vinyl sulfone cysteine proteinase inhibitors K11777 and WRR483 on invasion of human colonic xenografts. The resultant dramatic inhibition of invasion by both inhibitors in this human colonic model of amebiasis strongly suggests a significant role of secreted amebic proteinases, such as EhCP1, in the pathogenesis of amebiasis.</P>

        Effect of Pectin on the Expression of Proteins Associated with Mitochondrial Biogenesis and Cell Senescence in HT29-Human Colorectal Adenocarcinoma Cells

        Mitochondria dynamic is regulated by different proteins, maintaining a balance between fission and fusion. An imbalance towards mitochondrial fission has been associated with tumor cell proliferation. The aim of this study was to analyze whether pectin modifies the viability of human colon cancer cells and the expression of proteins involved in mitochondrial fusion and fission. The human colon carcinoma cell line HT29 cells was growth in 10% fetal bovine serum in the absence and presence of pectin. Pectin reduced HT29 cell viability in a concentration-dependent manner, reaching a plateau at 150∼300 μ㏖/L pectin. The presence of 200 μ㏖/L pectin reduced the expression of dynamin-related protein-1 and increased expression of the mitochondrial fusion-associated proteins mitofusin-1 and 2. Expression of cyclin B1, a protein involved in G2/M transition, was found decreased in pectin-incubated HT29 cells. Moreover, expression of p53 protein, the amount of p53 in the nucleous and β-galactosidase activity, which are all biomarkers for cellular senescence, were significantly higher in pectin-incubated HT29 cells than in HT29 cells incubated without pectin. Expression of the protein B-cell lymphoma 2 (Bcl-2) homologous antagonist/killer was increased in response to incubation with pectin. However, incubation with pectin did not affect expression of Bcl-2-associated X protein or Bcl-2, or the caspase-3 activity. Overall, we concluded that pectin reduces the viability of human HT29 colon cancer cells, which is accompanied with a shift in the expression of proteins associated with mitochondrial dynamics towards mitochondrial fusion. Moreover, incubation with pectin favors cellular senescence over apoptosis in HT29 cells.
