제주지역 모기의 계절적 발생소장 및 Real Time RT-PCR을 이용한 Flavivirus 감염조사(2017)

기후온난화 현상이 지속되고 있는 제주지역에서 환경적으로 다른 지역 모기의 계절적 발생밀도를 조사하기 위해 제주시의 국제공항, 항만구역과 축사 그리고 서귀포 도심지의 11지점을 선정하여 3월부터 11월까지 매달 2회씩 Black light trap과 BG sentinel trap을 이용하여 모기를 채집하였다. 채집된 모기는 5속 7종. 6,042마리였으며, 이 중 빨간집모기(Culex pipiens)가 4,159마리(68.8%)로 우점종이었으며 흰줄숲모기(Aedes albopictus)는 1,348마리(24.4%)였다. Black light trap를 이용한 채집에서 중앙동주민센터는 트랩당 72.8마리를 채집하여 모기 밀도가 가장 높게 나타났으며 제주국제공항은 트랩당 1.4마리로 가장 낮게 나타났다. BG sentinel trap을 이용한 채집에서는 항만에서 트랩당 71.7마리로 가장 많았고 도심지의 걸매생태공원에서 28.3마리로 가장 낮았다. 시기별로 모기의 밀도는 5월부터 증가하기 시작하여 8월에 1,156마리(19.1%)로 가장 높은 밀도를 나타내었다. 채집된 암컷모기를 종별, 시기별, 지점별로 나누어 pool당 50마리 이하로 설정하여 총 364 pools에서 flavivirus 존재여부를 real time RT-PCR로 검사하였으나, 검출되지 않았다. This study examined the seasonal, regional distribution of mosquito vectors related with disease vectors in the Jeju. From March to November, sample were collected from 11 points in four environmentally different sites in Jeju Island. Samples were collected twice a month using a black-light trap and a BG sentinel trap. Overall, five genera and seven species types of 6,042 female mosquitos were collected. Among the collected mosquitos, 4,159 (68.8%) and 1,348 (24.4%) were Culex pipiens and Aedes albopictus, respectively, making them the dominant species. Additionally, collection using the black light trap produced 72.8 entities per trap in Jungang-dong service center in the center of the city, which was the highest value, while the lowest amount of 1.4 per trap was recovered from the airport. When the BG sentinel trap was used, the largest recovery was observed in the port, where there were 71.7 entities per trap, while the lowest amount of 28.3 entities per trap was recovered at Gealmae Eco Park. The overall number of mosquitoes collected started to increase from May, and reached the largest value of 1,156 (19.1%) in August. Trapped mosquitoes are created 364 pools of up to 50 grains per pool, by season, by environmental, and by species. When the pools were used, no flaviviral infection was observed upon real time RT-PCR.

A 3-day-old Appaloosa colt was examined because of acute onset of ataxia, left-sided head tilt, fever and heart murmur. The foal continued to nurse but became increasingly depressed and mild colic. On plain radiological examination, the foal was normal. Uroperitoneum was diagnosed following abdominocentesis. Postmortem examination revealed bladder rupture. The lesion was in the dorsomedial part of the bladder wall. We suggested that the cause of bladder rupture was parturient trauma. Even though the rupture was associated with trauma, the margin of rupture of bladder was clearly smooth and regular.

A cell line of bovine origin was immortalized to isolate foot-and-mouth disease virus (FMDV). The immortalization was performed by infection of bovine primary epithelial cells with a recombinant retrovirus that over expressed the human telomerase (hTERT), after primary culture of fetal bovine kidney tissue and removal of fibroblasts. After cloning the immortalized cell line into single cells, the cloned cell lines were named JNUBK-1, JNUBK-2, JNUBK-3, and JNUBK-4, according to their characteristics. To confirm the epithelial phenotype of the cell lines JNUBK-3 and JNUBK-4, which showed stable proliferation capability over 35 generations after immortalization, the expression of cytokeratin and fibronectin was measured. Finally, the FMDV titer in the JNUBK-3 and JNUBK-4 cell lines was measured and was 800∼2,000 times higher than that of the currently used cell line IRBS-2. In conclusion, more sensitive isolation and production of FMDV became possible through the use of the immortalized JNUBK-3 and JNUBK-4 cell lines.

The disease causing colic is the greatest cause of death in the domestic horse population today and accidental deaths as a result of colic leads to decreased productivity and economic losses in the horse breeding industry. Large colon impaction is the most common cause of colic in horses. Horses may ingest sand either by eating from the ground in sandy locations, or because of its inclusion in hay. In certain areas sand impaction is relatively common. The horse (4 year, female) presented with abdominal pain, which was charactered by willing to lie down, pawing, and looking at flank. A month later, the horse died suddenly at sand paddock. Post-mortem examination revealed the sand impaction as a lot of feces, sand and gravels in the right dorsal colon. In addition, there was necrosis and debilitation in mucosal lining of the large colon and rupture site on the right dorsal colon. The purpose of this article is to review sand impaction and emergencies in Thoroughbred horses.
감마선을 조사한 마우스의 조혈 및 소장줄기세포에 대한 fucoidan의 방호효과

We investigated the potential of fucoidan for its ability to provide protection from gamma rayinduced damage. In our results, the fucoidan significantly improved the counts of endogenous colony forming unit to $9.5 {\pm} 1.5$, from $5.5 {\pm} 2.5$ compared with un-treated irradiated control group at 10 day after 7 Gy whole body irradiation. After 2 Gy irradiation, fucoidan treatment attenuated the percent of tail DNA of splenocytes, parameters of DNA damage, from $30.17 {\pm} 1.7%$ to $13.67 {\pm} 2.81%$ 2.81% by comet assay and also accelerated the proliferation of splenocytes, compared with un-treated irradiated control group by 3Hthymidine incorporation assay. Furthermore, fucoidan decreased the number of apoptotic fragments per intestinal crypt by 31.8% at 1 days after 2 Gy irradiation. These results indicated that the fucoidan significantly improved the hematopoietic recovery, prevented the DNA damage in immune cells and enhanced their proliferation, which had been suppressed by ionizing radiation. in addition, fucoidan rescued intestinal cells from radiation-induced apoptosis. Thus, this study raises the possibility of using fucoidan as adjuvant therapeutic agent after radiotherapy.
Thoroughbred 씨암말에서 자궁파열과 연관된 대장탈출증 및 망아지의 각인 1례

In this report, we described a case of acute colic with uterine rupture in a Thoroughbred broodmare during parturition. A 14-year-old broodmare showed acute colic characterized by rolling just after delivering of a healthy foal. Large intestine was prolapsed through vulva, contaminated with soil and its serosa was heavily damaged. We performed a euthanasia according to the owner's request. Post mortem examination revealed about 15 cm rupture on ventral side of the uterus. In addition, the orphan foal follow his owner after necropsy on his dam.

A variety of enteric deseases may result in the manifestation of abdominal pain in horse. An acute colic in a 5-month-old Thoroughbred filly is described. The filly was presented with acute colic characterized by rolling and pawing. Next day she suffered from severe abdominal pain and subsequently died. Post mortem examination revealed very heavy ascarid, Parascaris equorum (P. equrum) infection in cmall intestine. In addition, it is also noted the presence of P. equorum in the stomach. Infection due to P. equorummanifests itself in foals between 6-8weeks and one year of age. P. equorum is a common and ubiquitous parasite that persists for many years in stables and on pasture in spite of good hygiene and anthelmintic control programs.

Equine herpesvirus type 1 (EHV-1) is an economically important pathogen of horses and exerts its major impact by inducing abortion storms and sporadic abortions in pregnant mares, early neonatal death in foals, and respiratory disease in young horses. Although equine herpesvirus type 4 (EHV-4) rarely causes clinical manifestations of disease in organs other than the respiratory track, isolated cases of myeloencephalopathy and sporadic abortions have been reported in EHV-4 infections. Here, we report an abortion storm in Thoroughbred breeding farms in Jeju island, South Korea. It occurred for 16 days from first abortion to last one. There were no clinical signs prior to abortion and stillbirth in broodmares. Two PCR-primers were made on glycoprotein B gene of EHV-1 and EHV-4 to amplify specific common regions of the viruses. We could detect the virus specific genes in aborted samples by PCR, and concluded that the cause of abortion storm was EHV infection. This report describes the first abortion storm case caused by EHV in Thoroughbred breeding farms in South Korea.
