Heterophyopsis continua에 대한 praziquantel 시험관내 효과의 주사현미경적 관찰

This study was carried out to observe in vitro effect of praziquantel on the tegumental changes of Heterophyopsis continua with scanning electron microscope. Metacercariae were collected from the perch, Lateolabrax japonicus, by artificial digestion technique and fed to 2-week old chickens. Adult worms were recovered from small intestine of chickens 8 days after infection. For working solutions, praziquantel was diluted with TC199 medium at concentration of 0.01, 0.1. 1 and $10{\mu}g/ml$. To each petri dish containing 10ml of solution, 5~10 worms were introduced and incubated at $37^{\circ}C$. For the scanning electron microscopy(SEM), the worms were fixed in cold 2.5% glutaraldehyde, dehydrated in a series of graded ethanol and freeze-dried. Dried specimen was mounted on stub and coated with gold and observed in an SEM. The results were as follows: 1. Severe tegumental alterations were recognized by scanning electron microscope. Bleb formation of tegument was observed in 5 minute group and most pronounced on anterior tegument of worms. The number and size of blebs increased as incubation time prolonged. 2. The surface destruction was more pronounced at ventral margin between the oral and the ventral suckers. 3. The sensory papillae were slightly affected, but destruction of tegumental spine was not recognized. 4. The effect of praziquantel on the worm was found dependent on the concentration and incubation time, however, the effect was more dependent upon the incubation time.

This study was carried out to observe in vitro effect of praziquantel on the viability and internal organ changes of Heterophyopsis continua with light microscopy. Metacercariae were collected from the perch, Lateolabrax japonicus, by artificial digestion technique and fed to 2-week old chickens. Adult worms were recovered from the small intestines of chickens 8 days after infection. For working solutions, praziquantel was diluted with TC199 medium at the concentration of 0.01, 0.1, 1 and $10{\mu}g/ml$. To each petri dish containing 10ml of solution, 5~10 worms were introduced and incubated at $37^{\circ}C$. Motlity of worms was observed at 5, 15, 30, 60 minutes, 1, 2, 4 and 6 hours after incubation. For light microscopy, worms were fixed in 10% formalin under cover glass pressure and stained with Semichon's acetocarmine. The results were as follows: 1. In $0.01{\mu}g/ml$ praziquantel, the worms had their mobility until 6 hours post treatment. However, worms in over $0.1{\mu}g/ml$ of praziquantel contracted within 5 minutes and immobilized. 2. Intestine of the worm incubated in $0.001{\mu}g/ml$ praziquantel for 5 minutes was dilated and intestinal wall was thickened. 3. In incubated over $0.1{\mu}g/ml$ praziquantel, pharynx of the worm protruded out from oral sucker. 4. The lowest effective lethal concentration of praziquantel on H. continua was $0.1{\mu}g/ml$. The worms exposed to the drug were observed to be immobilized immediately after incubation in solutions of over $0.1{\mu}g/ml$ concentration. All of the worms in early period showed severe contraction and those in late period showed severe dilation.

The present investigation was focussed on histochemical and electron microscopical observations of utero-vaginal(U-V) glands in U-V junctions of domestic hens. In histochemical observations, fine granules by PAS technique for mucopolysaccharides and nile blue stain for acidic lipids were slightly stained on the cytoplasm of U-V glandular epithelium. Larger granules by Sudan black B stain for neutral fat and phospholipids and Sudan III stain for neutral fat were heavily stained on the perinuclear region of the U-V gland epithelial cells. These positive materials were heavily stained on the U-V glandular epithelium of lowfecundity hens and non-laying hens. In scanning electron microscopic findings of the U-V junction surface, the orifices of U-V glands are seen as the crater-like invagination. The neck of the U-V gland and the epithelium of U-V iunction were covered by ciliated epithelial cells. Aggregates of spermatozoa are observed often to be on the necks of the U-V gland. These spermatozoa heads are embedded in the glandular tubules and many spermatozoa tails are free on the epithelium of uterine surface. In transmission electron microscopic findings, the epithelial cells of the U-V glandular orifices were ciliated, columnar cells. The apical regions of these cells contained numerous electrondense, round secretory granules of uniformly size. The epithelial cells of the U-V glandular tubule were columnar or pyramid shape with round or oval nuclei. These epithelial cells have numerous microvilli and also contained electron-dense, round secretary granules of uniformly size and electron-lucent vesicles of various size. Spermatozoa are seen as the cross-sections of various regions of heads and tails in glandular tubules. Also spermatozoa arranged longitudinally parallel within the glandular tubules.
시판 감귤주스, 매실주스 및 키위 즙에서 Listeria monocytogenes와Salmonella Typhimurium의 생존성

Inactivation of Salmonella Typhimurium and Listeria monocytogenenes in mandarin orange, Pru-nus mume (maesil), and kiwi juices was evaluated. A three-strain mixture of S. Typhimurium or L. monocytogenes wasinoculated (7 log CFU/ml) into a commercial mandarin orange juice and maesil juice, and home-maid kiwi extract.The inactivation efect of Maesil juice was estimated by the addition into the other two fruit juices. All fruit juices hadacidic pH, ranging from 2.8 to 3.5 and it was not variable during all experimental period, being at 4oC for 14 days. TheL. monocytogenes within 7 days, while kiwi extractand mandarin orange juice archived 3.0-log inactivation and 1.0-log inactivation, respectively, until 14 days of stor-age. S. Typhimurium was completely reduced by Maesil juice and kiwi extract within 14 days, but mandarin orangejuice showed only 1.4-log inactivation. The inactivation of L. monocytogenes and S. Typhimurium was increased byadding 10% maesil juice to both mandarin orange juice and kiwi extract.
Prevalence of Babesia spp. in dogs of Seogwipo-si, Jeju-do, South Korea

Cases of canine babesiosis may present with a wide variation of severity of clinical signs, ranging from a hyper-acute, shock-associated, hemolytic crisis to an inapparent, subclinical infection. Dogs typically present with the acute form of babesiosis, which is characterized by general findings such as pyrexia, weakness, mucous membrane pallor, depression, hemorrhagic anemia. This study was conducted to investigate the prevalence of Babesia spp. infection in dogs of Seogwipo-si. A survey of canine Babesia spp. infections among 173 dogs in Seogwipo-si was performed from July 2008 to August 2008, taking a blood sample from dogs raised outdoors through cephalic vein and jugular vein. And Babesia spp. was diagnosed by examination of blood smear stained with Giemsa stain. Of 173 dogs, 9 dogs (5.2%) were infected with the Babesia spp. This result was a little lower than the prevalence of Babesia spp. in dogs of other areas.
제주도에서 사육되는 마의 질병과 기생충 감염에 관한 연구

The disease mobility and parasitic infection, and normal blood biochemistry values were investigated in Cheju horse. The ratio of the diseased horse was 23.3% in 1992 and 25.2% in 1993. The disease incidences of the digestive system, the respiratory system and the traumatic disease were 14.3%, 5.6% and 4.7%, respectively in the diseased horses. The rate of parasitic infection was 82.7% in Strongylus vulgaris, and 9.3% in Ascaris equiutn. The rate of bot's eggs on the equine hairs of Gastrophzlus spp. was 100% in the horses and the rate of infected larvae of Gastrophdus spp. was 37.5% in slaughtered horses. In the healthy horses, the values of RBC was 801.1 k79.1 l0^(4)/㎕, WBC was 9,090±1.533/㎕. PCV was 38.8±3.5%, hemoglobin was 13.0±1.5g/l00ml, total protein was 7.2±0.6g/l00ml, fibrinogen was 195.0±80.5mg/l00ml, sorbitol dehydrogenase was 0.62 ± 0.35IU/L, bilirubin was 2.5±0.5mg/100ml, blood urea nitrogen was 19.1±4.4mg/l00ml, creatinine was 1.1±0.2mg,/l00ml and glucose was 46.3±7.8mg/l00ml.
