        각막윤부 줄기세포의 줄기세포성 유지 및 증식에 대한 Wnt 단백질의 효과

        Purpose: To evaluate the effects of the Wnt protein on proliferation and stemness maintenance of cultured corneal limbal stem cells. Methods: We examined the expression of Wnt proteins by Western blot analysis. We then evaluated the effects of Wnt on cell proliferation by colony forming efficiency. β-catenin activation using Wnt proteins was examined by immunocytochemistry. We also examined the effects of Wnt on proliferation and stemness maintenance by reverse transcriptase polymerase chain reaction of p63 and connexin43. Results: Wnt has a different effect on corneal epithelial stem cells. Colony forming efficiency was also significantly higher in treated Wnt2 and Wnt4 cells compared with controls. The Wnt2 and Wnt4 treated cells showed nuclear accumulation of β -catenin. In addition, the limbal stem cell marker p63 was strongly expressed in Wnt2, Wnt4 Wnt5a, and Wnt5b. Connexin43 mRNA was also strongly expressed in Wnt5a, Wnt5b and Wnt7b cells. Conclusions: We suggest that Wnt2 and Wnt4 could lead to more effective proliferation and stemness maintenance for human corneal epithelial stem cells. J Korean Ophthalmol Soc 2009;50(4):588-593

      • 동작관찰 중 동작 수행 시 시점의 변화에 따른 뇌 활성의 변화

        Purpose: Treatments using a mirror neuron system, such as 3D virtual reality therapy, are used in stroke rehabilitation, but they need to be constructed according to a detailed procedure. The aims of this study were to analyze electroencephalograms (EEG) during relaxation and action while observing first person perspective (1AE) and third person perspective (3AE) videos of the right hand for 20's. Methods: Thirty participants (Male=4, Female=26) were recruited for this study. Participants were selected by a vividness of movement imagery questionnaire (VMIQ). EEG was measured during relaxxation and during action with 1AE and 3AE videos, focusing on the supination and pronation actions of participants' right hands. An absolute mu rhythm, a relatively high alpha power, and a relative beta power were identified. In each group, one-way repeated measures ANOVA was used for statistical analysis. Results: Measurement of absolute mu rhythms was significantly suppressed for both 1AE and 3AE compared with relaxation in C3 and C4 regions. High alpha wave measurements were significantly suppressed for both 1AE and 3AE in all regions, while beta wave measurements were significantly increased only for 3AE in F3 and F4 regions. Conclusion: Based on this study, we suggest that the mirror neuron system is activated during actions accompanied by action observation, especially actions with 3AE video observation, which can be a great therapeutic mathod in stroke rehabilitation.

        Streptozotocin-유발 당뇨쥐에 대한 클로렐라 열수 추출물의 혈당 강하 효과

        클로렐라의 생리활성작용 중 인슐린작용 활성 및 항당뇨 효능이 보고된 바 있으나 클로렐라 열수 추출물(Chlorella hot water extract, CE)에 의한 혈당치 조절에 관한 연구는 보고된 바 없다. 따라서 본 실험에서는 실험적인 당뇨유발물질 streptozotocin (STZ)을 성장기 SD계 수컷 흰쥐에 단회 복강 주사하여 당뇨를 유발시킨 CE 무첨가 STZ-Control군, CE 3% 첨가 STZ+CE3군 및 CE 6% 첨가 STZ+CE6군, 그리고 정상군(Normal군)으로 실험 설계하여 4주간 식이와 함께 물을 자유 급여하였다. 공복 및 비공복시 혈당치는 정상군보다 당뇨 대조군에서 현저히 증가하여 당뇨 유발이 확인되었고, 이러한 혈당치 증가는 클로렐라 열수 추출물 분말 3% 첨가 STZ+CE3군 및 6% 첨가 STZ+CE6군에서 각각 감소하였다. 혈중 인슐린 농도는 당뇨 대조군에서 낮게 나타났으나 클로렐라 열수 추출물 분말 첨가식이 투여군들에서 인슐린 농도 감소현상이 상쇄되었다. 또한 정상군보다 STZ-당뇨 대조군에서 Fructosamine 및 뇨당이 증가하였으나, 클로렐라 추출물 투여 STZ+CE3군 및 STZ+CE6군에서 감소하는 경향을 나타내었고, 특히 이러한 효과는 클로렐라 열수 추출물 첨가농도 의존적이었다. 클로렐라 추출물 투여군에서 혈당치 감소의 기작을 검토하기 위하여 인슐린 합성과 분비에 관여하는 췌장의 췌도 베타세포의 면역반응에 의한 조직검사에서 정상군의 정상적인 둥근 모양의 형태를 잘 유지하고 있었으나, STZ-당뇨 대조군에서는 베타세포가 심하게 파괴되어 극소수 존재함으로서 면역 반응성이 매우 미약하였다. 그러나 클로렐라 열수 추출물 투여군에서 농도 의존적으로 베타세포가 많아지면서 정상군과 비슷한 췌도 세포의 형태를 유지하고 있었다. 클로렐라 열수 추출물은 STZ에 의해 파괴된 베타세포를 회복시키는 효능이 강한 것으로 보여 진다. 이러한 결과는 클로렐라 열수 추출물에 의해 인슐린을 생산해 내는 췌도 베타세포가 회복됨에 따라 인슐린 증강 작용으로 혈당치 강하 효과가 실험 동물을 통하여 확인되었다. The effect of Chlorella hot water extract (CE) on hyperglycemia in streptozotocin-induced diabetic rats has not been studied. Therefore, hypoglycemic effect of CE in type I streptozotocin-induced diabetic rats was studied. Rats were fed a semisynthetic diet supplemented with either 3% (the STZ+CE3) and 6% (the STZ+CE6) CE or no supplement the Normal and the STZ-Control rats for 4 weeks. The concentrations of fasting and non-fasting blood glucose were higher in the STZ-Control rats than in the Normal rats, but this rise was lowered in the STZ+CE3 and the STZ+CE6 rats. Serum insulin concentrations were decreased with STZ injection, however, the decreased levels were almost restored to the Normal level with CE supplementation. The increased serum fructosamine levels associated with hyperglycemia were decreased with the CE treatment. The morphology of pancreatic islets in the Normal rat was round and maintained a typical arrangement. The STZ-Control pancreatic beta-cells were found to have significant swelling and severely morphological damaged, however, pancreatic tissue damage by STZ in the CE-supplemented diet group was ameliorated. This study shows that Chlorella hot water extract had a hypoglycemic effect on the STZ-diabetic rats via either increased insulin secretion during recovery or the prevention of STZ-induced pancreatic damage.

        클로렐라 균주의 분리 동정 및 열수 추출물의 이화학적 조성

        클로렐라의 생리활성 물질을 활용하여 기능성 식품을 개발하기 위한 기초 자료를 얻기 위하여 클로렐라 배양장에서 생육이 빠른 클로렐라를 분리하여 동정하고 이화학적 특성을 조사하였다. 분리된 클로렐라는 전형적인 둥근 모양과 3㎛ 크기로 주름진 형태를 취하고 있었다. 분리된 클로렐라의 동정을 위하여 18S rRNA sequences를 실시한 결과 Chlorella sp. IFRPD 1018와 99%의 높은 상동성을 보여 Chlorella sp.와 거의 일치하여 Chlorella sp. CMS-1로 명명하였다. 클로렐라 열수 추출물에 조단백질 함량이 59.21%로 많이 함유되어 있었고, 조지방 함량은 0.01%로 미량 함유되어 있었다. 클로렐라 CMS-1의 무기질 농도는 K 2.52%, P 2.25%, Mg 0.63%, Ca 0.63% 순으로 함유되어 있었다. 클로렐라 CMS-1의 주요 구성 아미노산은 glutamic acid 6.21%, alanine 5.76%, aspartic acid 5.44% 순이였으며, 특히 유리아미노산 중 γ-aminobutyric acid (GABA)의 함량은 7.13%로 전체 유리아미노산의 63.8% 비율로 차지하였고 그 다음으로 L-alanine 1.44%, L-glutamic acid 0.90%, L-leucine 0.26%, L-glycine 0.20% 순으로 함유되어 있었다. CMS-1 strain was isolated from the outdoors cultivation pools in Culmansa Co., Ltd. This strain was found to be a rounded type of 3 ㎛. Phylogenetic analysis by the 18S rRNA sequencing using isolated strain is most similar to Chlorella sp. IFRPD 1018 gene at the level of nucleotide sequence identity at 99%. Accordingly, the isolated Chlorella strain was named as Chlorella sp. CMS-1 based on its morphological and phylogenetic properties. The concentrations of crude protein and fat were 59% and 0.01%, respectively. Major compositional amino acids (㎎%) were glutamic acid 6.21, alanine 5.76, aspartic acid 5.44%, glycine 4.29%, and threonine 3.09% and major free amino acids (㎎%) were γ-aminobutyric acid (GABA) 7.13%, L-alanine 1.44%, L-glutamic acid 0.90, L-leucine 0.26% and L-glycine 0.20%. The concentrations of major minerals were P 2.25%, K 2.25%, Na 1.09%, Mg 0.63%, and Ca 0.28%.

        상향식 연속주조법에 의한 Plate 및 Tube 형 Sn 결정의 성장 안정성

        The Upward Continuous Casting process which manufactures near-net shaped crystals directly from the melt has been improved in order to reduce the cost of products. Growth of plate and tube shaped crystals using the U.C.C. process was carried out with tin. Near-net shaped tin plates and tubes have been produced with the newly developed mold. The effects of process parameters on the size and the shape of the crystal were studied:these were the meniscus height(S), the length between the top surface of the mold and free surface(H), the mold temperature, and the pulling speed. Of these S and H were the most important parameters in determining the crystal shape stability. The U.C.C. process was compared with other shaped growth methods to determine its growth stability. It can be concluded that the U.C.C. process will find applications in the growth of various shaped crystals.
