      • Garcinol, an Acetyltransferase Inhibitor, Suppresses Proliferation of Breast Cancer Cell Line MCF-7 Promoted by 17β-Estradiol

        The acetyltransferase inhibitor garcinol, a polyisoprenylated benzophenone, is extracted from the rind of the fruit of Garcinia indica, a plant found extensively in tropical regions. Anti-cancer activity has been suggested but there is no report on its action via inhibiting acetylation against cell proliferation, cell cycle progression, and apoptosis-inhibtion induced by estradiol ($E_2$) in human breast cancer MCF-7 cells. The main purposes of this study were to investigate the effects of the acetyltransferase inhibitor garcinol on cell proliferation, cell cycle progression and apoptosis inhibition in human breast cancer MCF-7 cells treated with estrogen, and to explore the significance of changes in acetylation levels in this process. We used a variety of techniques such as CCK-8 analysis of cell proliferation, FCM analysis of cell cycling and apoptosis, immunofluorescence analysis of NF-${\kappa}B$/p65 localization, and RT-PCR and Western blotting analysis of ac-H3, ac-H4, ac-p65, cyclin D1, Bcl-2 and Bcl-xl. We found that on treatment with garcinol in MCF-7 cells, $E_2$-induced proliferation was inhibited, cell cycle progression was arrested at G0/G1 phase, and the cell apoptosis rate was increased. Expression of ac-H3, ac-H4 and NF-${\kappa}B$/ac-p65 proteins in $E_2$-treated MCF-7 cells was increased, this being inhibited by garcinol but not ac-H4.The nuclear translocation of NF-${\kappa}B$/p65 in $E_2$-treated MCF-7 cells was also inhibited, along with cyclin D1, Bcl-2 and Bcl-xl in mRNA and protein expression levels. These results suggest that the effect of $E_2$ on promoting proliferation and inhibiting apoptosis is linked to hyperacetylation levels of histones and nonhistone NF-${\kappa}B$/p65 in MCF-7 cells. The acetyltransferase inhibitor garcinol plays an inhibitive role in MCF-7 cell proliferation promoted by $E_2$. Mechanisms are probably associated with decreasing ac-p65 protein expression level in the NF-${\kappa}B$ pathway, thus down-regulating the expression of cyclin D1, Bcl-2 and Bcl-xl.

        Two new butyrolactones, asperphenol A (1) and B (2), together with four known butyrolactones (3-6) were isolated from the fermentation products of an endophytic fungus Aspergillus versicolor. Their structures were elucidated by spectroscopic methods including extensive 1D- and 2D-NMR techniques. Compounds 1-6 were also tested for their anti-tobacco mosaic virus (anti-TMV) activities. The results showed that compound 2 exhibited high anti-TMV activity with inhibition rate of 46.7%. The other compounds also exhibited potential anti-TMV activities with inhibition rates in the range of 21.8-28.4%.

      • The Interannual Variations of the Summer Monsoon Onset over the South China Sea

        Interannual variations of the summer monsoon onset over the South China Sea (SCS) have been studied using data from over seventeen years (1979-1995) of NMC global analysis and of Outgoing Longwave Radiation (OLR) observed with NOAA polar-orbitting satellites. It was found that the summer monsoon onset in the SCS occurs abruptly with a sudden change of zonal wind direction from easterly to westerly and an exploding development of deep convection in the whole SCS region in the middle of May. Based on the criteria defined in this paper for the SCS summer monsoon onset, the average onset date over the SCS from 1979 to 1995 is around the fourth pentad of May. The airflow and general circulation over the SCS changes dramatically after the onset. The ridge of the subtropical high in the western Pacific in the lower troposphere weakens and retreats eastward from the SCS region with an establishment of westerly winds over the whole region. During the SCS monsoon onset, the most direct impact in the vicinity of the SCS are the equatorial westerlies in the Bay of Bengai through their eastward extension and northward movement. An indirect influence on the SCS onset is also caused by the enhancement of the Somali cross-equatorial flow and the vanishing Arabian High over the sea; the latter may be a signal for the SCS onset. There are quite significant interannual variations in the SCS onset. In the years of a delayed onset, the most profound feature is that the easterly winds stay longer in the SCS than on average. Deep convection activities are suppressed. The direct cause is the abnormal existence of the western Pacific subtropical high over the SCS region. Moreover, compared to the average, the equatorial westerlies in the Bay of Bengal are also weaker in the years of a delayed onset. No significant changes for the crossequatorial flow at 105°E are observed for these years. It has also been found that the interannual variations of the SCS onset are closely related with the ENSO events. In the years of a delay, the Walker circulation is weaker, and the sea surface temperature (SST) anomalies in the western Pacific are negative.

        Effects of Copper-bearing Montmorillonite (Cu-MMT) on Escherichia coli and Diarrhea on Weanling Pigs

        Copper-bearing montmorillonite (Cu-MMT) was produced by $Cu^{2+}$ cation exchange reaction. X-ray diffraction analysis showed that that the (001) basal spacing of the MMT crystal lattice increased from 1.544 to 1.588 nm after $Cu^{2+}$ exchange. This indicated that $Cu^{2+}$ entered into interlayer position of MMT as a hydrated cation or composite cation. In vitro results indicated that Cu-MMT had antibacterial activity on Escherichia coli $K_{88}$. Cu-MMT had unbalanced positive charge after cation exchange. Its antibacterial activity resulted from two aspects, one was electrostatic attraction which made E. coli $K_{88}$ being adhered on the montmorillonite surface, the other was the $Cu^{2+}$ slowly released, which could kill bacteria. In an in vivo study, four replicates of eight weanling pigs were assigned to each of two dietary treatments to study the effects of Cu-MMT on diarrhea, E. coli in the lumen of the jejunum and morphology of jejunal mucosa. As compared to the control, supplementation of the diet with 0.2% Cu-MMT improved average daily gain by 12.50% (p<0.05) and decreased F/G by 9.42% (p<0.05). The mean diarrhrea incidence was decreased by 71.80% (p<0.05). The viable counts of Escherichia coli in jejunal contents were significantly reduced (p<0.05). Villus height and the villus height to crypt depth ratio at the jejunal mucosa were increased by 19.09% (p<0.05) and 37.10% (p<0.05), respectively.

        A High-efficiency Trim Method for CFD Numerical Calculation of Helicopter Rotors

        In order to resolve the trimming difficulty in rotor CFD calculations, a high-efficiency and improved "delta trim method" is established to compute the blade control settings that are necessary to identify the blade motion. In this method, a simplified model which combines the blade element theory and different inflow models is employed to calculate the control settings according to the target aerodynamic forces, then it is coupled into a CFD solver with unsteady Navier-Stokes equations by the delta methodology, which makes the control settings and aerodynamics calculated and updated in the meantime at every trim cycle. Different from the previous work, the current research combines the inflow model based on prescribed wake theory. Using the method established, the control settings and aerodynamic characteristics of Helishape 7A, AH-1G and Caradonna-Tung rotors are calculated. The influence of different inflow models on trimming calculations is analyzed and the computational efficiency of the current "delta trim method" is compared with that of the "CFD-based trim method". Furthermore, for the sake of improving the calculation efficiency, a novel acceleration factor method is introduced to accelerate the trimming process. From the numerical cases, it is demonstrated that the current "delta trim method" has higher computational efficiency than "CFD-based trim method" in both hover and forward flight, and up to 70% of the amount of calculation can be saved by current "delta trim method" which turns out to be satisfactory for engineering applications. In addition, the proposed acceleration factor shows a good ability to accelerate the trim procedure, and the prescribed wake inflow model is always of better stability than other simple inflow models whether the acceleration factor is utilized in trimming calculations.

        Research on Grid Side Power Factor of Unity Compensation Method for Matrix Converters

        Input filters are very important to matrix converters (MCs). They are used to improve grid side current waveform quality and to reduce the input voltage distortion supplied to the grid side. Due to the effects of the input filter and the output power, the grid side power factor (PF) is not at unity when the input power factor angle is zero. In this paper, the displacement angle between the grid side phase current and the phase voltage affected by the input filter parameters and output power is analyzed. Based on this, a new grid side PF unity compensation method implemented in the indirect space vector pulse width modulation (ISVPWM) method is presented, which has a larger compensation angle than the traditional compensation method, showing a higher grid side PF at unity in a wide output power range. Simulation and experimental results verify that the analysis of the displacement angle between the grid side phase current and the phase voltage affected by the input filter and output power is right and that the proposed compensation method has a better grid side PF at unity.

      • Purification and Characterization of an Acid Deoxyribonuclease from the Cultured Mycelia of Cordyceps sinensis

        A new acid deoxyribonuclease (DNase) was purified from the cultured mycelia of Cordyceps sinensis, and designated CSDNase. CSDNase was purified by $(NH_4)_2SO_4$ precipitation, Sephacryl S-100 HR gel filtration, weak anion-exchange HPLC, and gel filtration HPLC. The protein was single-chained, with an apparent molecular mass of ca. 34 kDa, as revealed by SDS-PAGE, and an isoelectric point of 7.05, as estimated by isoelectric focusing. CSDNase acted on both double-stranded (ds) and single- stranded (ss) DNA, but preferentially on dsDNA. The optimum pH of CSDNase was pH 5.5 and its optimum temperature 55. The activity of CSDNase was not dependent on divalent cations, but its enzymic activity was inhibited by high concentration of the cation: $MgCl_2$ above 150 mM, $MnCl_2$ above 200 mM, $ZnCl_2$ above 150 mM, $CaCl_2$ above 200 mM, NaCl above 300 mM, and KCl above 300 mM. CSDNase was found to hydrolyze DNA, and to generate 3-phosphate and 5-OH termini. These results indicate that the nucleolytic properties of CSDNase are essentially the same as those of other well-characterized acid DNases, and that CSDNase is a member of the acid DNase family. To our knowledge, this is the first report of an acid DNase in a fungus.

        Colossal dielectric behavior in TbFeO3 ceramics

        TbFeO3(TFO) ceramic samples were prepared via the traditional solid-state reaction route. The lowtemperature (100-300 K) dielectric properties of TFO have been systematically investigated in the frequency range from 100 Hz to 5 MHz. Colossal dielectric behavior contributed from two thermally activated dielectric relaxations was found in the sample. The low-temperature relaxation with the activation energy of 0.22 eV was confirmed to be a polaron relaxation caused by electrons hopping between the mixed-valent states of Fe ions. The high-temperature relaxation shows the activation energy of 0.59 eV and can be enhanced by annealing treatment in oxygen and weakened in nitrogen. This relaxation was ascribed to be a MaxwelleWagner relaxation due to surface-layer effect.

        To improve students’ classroom engagement is not only the demand of curriculum revolution, but also the reflection of learning democracy. Students' responses and thinking are the main manifestations of students' participation in classroom learning. To reduce the amount of questions and increase the opportunities and time for students to think, this study, by employing SPSS, makes attempts to analyze the data by using multivariate GLM analysis to explore the effects of students' responses and thinking on learning results. The results indicated the students learning effect will be promoted through reducing the quantity and increasing the quality of question and adding the thinking opportunities.
