<P>Administration of mountain ginseng (MG) extract can restore advanced cancer to a normal state. To elucidate the mechanism by which MG extract prevents the progression of lung cancer, the processes of proliferation and death of lung cancer cells (A549) were examined after treatment with MG extract. Butanol-extracted MG (BX-MG) showed a high inhibitory effect (IC(50) = 2 mg/ml) by attenuating proliferation and inducing apoptosis in lung cancer cells. By HPLC-UV analysis of BX-MG, ginsenosides, Rb1 was identified as the most abundant ginsenoside, followed by Rg1, Re, Rc and Rb2. BX-MG induced caspase-3 dependent apoptosis by inhibiting NF-관B. In addition, BX-MG activated p53 and p21, resulting in the attenuated proliferation of A549 cells. Reduced activity of the NF-관B promoter and increased activity of the p53 promoter indicate that BX-MG regulates apoptosis at the level of transcription in lung cancer cells. Furthermore, BX-MG blocked the nuclear translocation of RelA and the associated reduction in surviving. These results suggest that BX-MG inhibits lung cancer cell growth by activating tumor suppressors and inhibiting nuclear translocation of NF-관B.</P>

Citrus mosaic sadwavirus (CiMV) is a closely related virus with the Satsuma dwarf virus (SDV) along with Navel orange infectious mottling virus (NIMV), Natsudaidai dwarf virus (NDV), and Hyugagatsu virus (HV). The present study found that the typical symptoms of CiMV-infected citrus fruits include the appearance of dark blue speckles or ringspots on fruit rinds and the browning of oil glands in the spots as rind coloring began. As rind coloring progressed, the spots gradually faded, whereas the browning of the oil glands worsened to the point that the tissues surrounding the oil glands became necrotic. In very early satsuma mandarins (Citrus unshiu ‘Miyamoto Wase’) and ‘Setoka’ cultivar (C. hybrid ‘Setoka’) of late-maturity citrus, the symptomatic fruits were eventually dropped. And in early satsuma mandarin (C. unshiu ‘Miyakawa Wase’), the peel hardness of the virus-infected fruit (1,618.3 ± 305.5, g-force) was more than twice as hard as that of the healthy fruit (636.5 ± 39.1, g-force). The ratio of flesh weight to total fruit weight was higher for the healthy fruit (77.3 ± 1.7%) than for the infected fruit (70.7 ± 0.6) and peel puffing was more severe in the infected fruit (2.9 ± 0.4 mm) than in the healthy fruit (0.9 ± 0.2 mm). The soluble solids content in infected citrus fruits was less values than the healthy fruit by 0.5-1.5 °Brix. These findings reveal that CiMV infection on citrus trees reduces the fruit quality of citrus.
제주에서 적산온도를 이용한 볼록총채벌레 세대별 발생최성기 예측모형

볼록총채벌레는 제주도 감귤에서 경미한 피해만 보고되다가 2007년부터 다발생하여 그 피해가 갈수록 심각해지고 있다. 본 연구는 적산온도를 이용하여 볼록총채벌레 각 발생세대의 발생최성기를 예측할 수 있는 모형을 개발하고자 수행하였다. 볼록총채벌레 성충 발생세대수를 독립변수(x)로 취급하고 세대별 발생최성기의 적산온도를 종속변수(y)로 취급하여 직선 회귀식을 추정하였다. 감귤원에서 유살된 자료와 녹차 또는 키위과원에서 얻은 자료를 기반으로 각각 감귤기반모형(y = 310.9x + 69.0, $r^2$=0.99) 녹차기반모형(y = 285.7x + 84.1, $r^2$=0.99)을 개발하였다. 각 모형의 예측값과 독립된 포장 실측자료와의 잔차자승합을 토대로 모형의 적합성을 평가하였으며, 녹차기반모형의 적합력이 좋았다. 본 예측모형을 통한 계산값과 실측치의 불일치에 대한 원인과 모형의 포장 활용도에 대하여 고찰하였다. The yellow tea thrips, Scirtothrips dorsalis (Thysanoptera: Thripidae), has been regarded as a minor pest on citrus on Jeju Island. However, the damage of yellow tea thrips has gradually increased since 2007. This study was conducted to develop a forecasting model for generation peak time of S. dorsalis by using degree-days. Simple linear regression analysis was applied to determine the relationship between the generation number (x, dependent variable) and degree-days (y, independent variable). As a result, two regression models were established: citrus-based model (y = 310.9x + 69.0, $r^2$=0.99) and green tea-based model (y = 285.7x + 84.1, $r^2$=0.99). The models was fitted by independent data sets obtained from 2013 and evaluated using the technique of RSS (residual sum of square) and ${\chi}^2$-test. The green tea based-model showed a good fitting ability. The discrepancy between model outputs and actual data, and the practical application of models were discussed.

Satsuma dwarf virus (SDV) or Citrus mosaic sadwavirus (CiMV) were not consistently detected in RTPCR assay with the primer sets based on gene of Japan isolates. SDV and CiMV isolates were distinctively divided into two groups based on phylogenetic analysis of PP2 gene cloned from 22 Korean isolates, and the Korean CiMV and SDV isolates shared 95.5-96.2% and 97.1-97.7% sequence identity with Japanese isolate, respectively. We developed PP2-1 primer set based on the PP2 gene sequence of Korean isolates to simultaneously and effectively detect SDV and CiMV. And CTLV-2013 and CTV-po primer sets were newly designed for detection of Citrus tatter leaf virus (CTLV) and Citrus tristeza virus (CTV), respectively. Using these primer sets, a new multiplex PCR assay was developed as a means to simultaneously detect 4 citrus viruses, CTV, CTLV, SDV, and CiMV. The degree of detection by the multiplex PCR were consistent with those of uniplex RT-PCR for detection of each of the viruses. Therefore, the new multiplex PCR provides an efficient method for detecting 4 citrus viruses, which will help diagnose many citrus plants at the same time. We verified that 35.2% and 72.1% of 775 trees in 155 orchards were infected with SDV or CiMV (SDV/CiMV) and CTV by the multiplex-PCR assay, respectively, and CTLV was not detected in any of the trees tested.

Elsinoe fawcettii and E. australis are two currently recognized scab pathogens of citrus. E. fawcettii has at least six pathotypes while E. australis has at least two pathotypes. Colonies of E. fawcettii and E. australis do not sporulate in artificial media including potato dextrose agar (PDA). Whiteside's method has been widely used for preparing conidial inoculum in vitro. This study was carried out to develop efficient method for conidia production from artificial media. We developed a shaking method which included the following steps: 1) Colony grown on PDA was mashed with a steel spatula; 2) Mycelia fragments were cultured in 50 ml sterilized rain water in a rotary shaker-incubator (180 rpm) at $25^{\circ}C$ for 24 h: 3) The conidia suspension was filtered through two layers of cheesecloth. Average conidia production of all isolates tested using this shaking method was approximately 13.1 times higher than that from Whiteside's method in this study.
First Report of Citrus Scab on Trifoliate Orange (Poncirus trifoliata)

Citrus scab symptoms were observed on leaves of trifoliate orange (Poncirus trifoliata) in open field of Citrus Research Institute, National Institute of Horticultural and Herbal Science. Typical scab pustules were formed and they were elevated protuberances form and light brown to grey. The pathogens were isolated from the symptomatic leaves and then were identified to Elsinoë fawcettii by morphological characteristics, pathoge- nicity and PCR assay. The morphological characteristics of colonies formed very slow-growing, pulvinate, or raised and deeply fissured, gummy to mucoid or tomentose colonies on potato dextrose agar medium. The pathogens were pathogenic to leaves of Satsuma mandarin, lemon, sour orange and grapefruit with typical scab symptoms. In PCR assay, specific amplified of products of 717 bp with Efaw-1 and 384 bp with Efaw-2 were observed from trifoliate isolates and E. fawcettii as reference but not from E. australis. This is first report of trifoliate orange being positive for citrus scab disease.
